CnAb1 knock down in R1 mESCs differentiated to mesoderm in Ebs

ABSTRACT: R1 mESCs were trasnfected with siRNAs against CnAb1 or Control (Luciferase) using Lipofectamine. mRNA was extracted 48 hours using Qiagen RNeasy Mini kit using DNAse treatment Microarray was performed after QPCR validation of the downregulation of CnAb1.

Project description:To confirm that the SMAD1/5- and SMAD4-associated genes are direct transcriptional regulators in mESCs in response to BMP, we treated undifferentiated R1 ES cells with BMP4 or with the BMP4 antagonist noggin, which can inhibit BMP signaling effectively for 4 h. Undifferentiated R1 ES cells were treated for 4 h with BMP4 or with the BMP4 antagonist noggin, which can inhibit BMP signaling effectively. Untreated R1 ES cells served as the control.

Project description:Stem cell-derived tissues have wide potential for modelling developmental and pathological processes as well as cell-based therapy. However, it has proven difficult to generate several key cell types in vitro, including skeletal muscle. In vertebrates, skeletal muscles derive during embryogenesis from the presomitic mesoderm (PSM). Using PSM development as a guide, we establish conditions for the differentiation of monolayer cultures of mouse embryonic stem (ES) cells into PSM-like cells without the introduction of transgenes or cell sorting. We differentiated mouse ESCs in serum-free medium supplemented with Rspo3 ( or as an alternative with Chir 9902) and the Bmp inhibitor LDN193189. In vivo, the PSM cells are first expressing Msgn1 (posterior PSM marker) and then mature to express Pax3 (anterior PSM marker). After 4 days of differentiation of mESCs, Msgn1-positive cells were FACS-sorted and their transcriptome analyzed. After 6 days of differentiation, Pax3-positive cells were sorted and their transcriptome analyzed. Laser-capture microdissection and RNA sequencing of aleurone and starchy endosperm of maize R1 genotypes

Project description:Three homozygous R1 genotypes, R1-g (WT), R1-sc and r1-m3 were laser microdissected and sequenced. The data showed that in aleurone (AL), expression of R1 in R1-sc and r1-m3 is higher than R1-g, and R1 is ectopically expressed in starchy endosperm (SE) in R1-sc and r1-m3. Interestingly, the ectopic expression of R1 in SE may be associated with RNA splicing process.
