ABSTRACT: The functionality of dendritic cells might be influenced by alterations of their biophysic microenvironment, e.g. changes in salt concentration. Microarray analysis aims to evaluate whether dendritic cells cultured in medium containing different salt concentrations modulate their gene expression profile.

Project description:Mouse bone marrow derived dendritic cells were generated by culturing bone marrow cells at a density of 0.5x10E6 cells/ml in RPMI-1640 supplemented with 5% FCS, 1% Pen/Strep, 5microM 2-mercaptoethanol, 20ng/ml GM-CSF. At day 7 dendritic cells were stimulated or not with 500 ng/ml LPS, and collected at day 10. 2x10E8 cells were used to prepare whole cell extracts and to perform PU.1 immunoprecipitaion with PU.1 antibody (T-21 Santa Cruz). IgG was used as control.

Proteomic analysis identifies the potential effects of trichostatin A and bone marrow-derived dendritic cells on NIH3T3 cells under oxygen–glucose deprivation conditions

Project description:Fibroblasts usually mediate acute wound healing and long-term tissue remodeling with scarring in tissue injury. In myocardial infarction (MI), following a prolonged lack of oxygen supply, necrotized cardiomyocytes become replaced by secreted extracellular matrix proteins produced by fibroblasts. Dendritic cells (DCs) act as inflammatory cells and can migrate from the bone marrow to the infarct areas and infarct border areas to mediate collagen accumulation after MI, whereas trichostatin A (TSA) can regulate the apoptosis and proliferation of the fibroblasts and affect DCs functions under oxygen–glucose deprivation (OGD) conditions. In this study, we used proteomics to investigate the effects of TSA and bone marrow-derived dendritic cells (BMDCs) on NIH3T3 fibroblasts under OGD conditions. Results showed that the fatty acid degradation pathway was significantly upregulated in NIH3T3 cells under OGD conditions, and the fatty acid synthesis pathway was significantly downregulated in NIH3T3 cells treated with BMDCs conditioned media with TSA (BMDCs-CM[TSA]) under OGD conditions. Meanwhile, the BMDCs-CM(TSA) significantly decreased the levels of triglycerides and free fatty acids and mediated ten fatty acid metabolism-related proteins in the NIH3T3 cells under OGD conditions. Summarily, the proteomic analysis showed that TSA and BMDCs affect fatty acid metabolism in NIH3T3 cells under OGD conditions.

RNAseq of conventional dendritic cells generated from mouse bone marrow cultures under different conditions

Project description:Type 1 conventional dendritic cells (cDC1s) are infrequent immune cells with an essential role in orchestrating immune responses to malignancies or infections. Despite their significance in regulating adaptive immunity, the absence of efficient manufacturing techniques to produce adequate yields of cDC1s has hindered their application in therapeutic settings. Here, we find that Interleukin-4 (IL-4) enhances the yield of cDC1 cells produced from Flt3 ligand (Flt3L) cultures of hematopoietic progenitors from either mice or humans, while inhibiting the generation of plasmacytoid DCs. IL-4 acted predominantly on DC progenitors and its activity relied on the cell intrinsic expression of IL4-RA or STAT6. Both in vitro and in vivo, cDC1s that had been stimulated by IL-4 were effective in activating and promoting the expansion of cytotoxic CD8 T cells. Gene expression analysis pointed to a critical role for IL-4 in promoting cDC1 proliferation in vitro. Overall, our research reveals an unanticipated role for IL-4 in fostering the development and manufacture of bona-fide cDC1s, making it easier to employ them for further mechanistic studies, and opening the door to leveraging their potential use in human diseases.

Sumoylation-deficient bone marrow derived dendritic cells transcriptomic analysis after LPS stimulation

Project description:Bone marrow derived dendritic cells were generated from Ubc9[fl;-] and Ubc9[+/+] mice. After in vitro derivation in the presence of GM-CSF, dendritic cells were treated with tamoxifen for four days to cause CreERT2 activation, and induce Ubc9 floxed allele deletion. This allowed comparative transcriptomic analysis of Ubc9[+/+] and Ubc9[-/-] dendritic cells unstimulated or stimulated with 10ng/ml LPS for one hour and six hours. Three biological replicates of each conditions were used. Analyzed bone marrow derived dendritic cells were Ubc9 WT or Ubc9 KO. Cells were left unstimulated or stimulated with LPS 10 ng/ml for one hour and six hours.
