Project description:Maternal nutrition during different stages of pregnancy can induce significant changes in the structure, physiology, and metabolism of the offspring. These changes could have important implications on food animal production especially if these perturbations impact muscle and adipose tissue development. The objective of this study was to evaluate the effect of different maternal diets on the transcriptome of fetal tissues in sheep. Ewes were bred to a single sire and from days 67 ± 3 of gestation until necropsy (days 130 ± 1), they were fed one of three isoenergetic diets: alfalfa haylage (H; fiber), corn (C; starch), or dried corn distillers grains (D; fiber plus protein plus fat). Longissimus dorsi (M), subcutaneous adipose depot (S), and perirenal adipose depot (R) tissues from individual fetuses were pooled and then analyzed by RNA sequencing. A total of 26 fetuses were removed from 15 dams. From the fetuses three different tissues were collected including one muscle, longissimus dorsi muscle (M), and two adipose tissues, perirenal adipose depot (R) and subcutaneous adipose depot (S). The RNA samples from the 26 fetuses were pooled to generate four biological replicates per maternal diet and tissue. In particular, per each diet and tissue, two RNA pools were created from male fetuses and two RNA pools were created from female fetuses. Overall, a total of 36 pools (i.e., 12 pools per tissue, 3 tissues in total) underwent RNA extraction, library generation, and subsequent sequencing.

Project description:Toll-like receptor 4 (TLR-4) is a transmembrane protein. Its activation leads to NF-kB signaling pathway and proinflammatory cytokines’ production that are responsible for activating innate immunity system. Here, we design a co-immunoprecipitation based cross-linking proteomics approach to reveal the TLR-4 interactome upon treatment of lipopolysaccharides and statin drug in HA-TLR-4 transfected HEK293 cells. A total of 712 differentially expressed proteins were identified and quantified in this study. Selected candidate proteins, macrophage myristoylated alanine-rich C kinase substrate, and creatine kinase, exclusively identified in the treatment of statin along with cross-linkers, were further validated by immunoblotting.

Project description:When macrophages are activated by sensing bacterial lipopolysaccharides (LPS), they greatly increase their motility, mRNA synthesis and protein production. Most of the ATP needed for these responses is derived from the uptake and catabolism of glucose, a relatively inefficient ATP source. Although the stimulated cells also increase their uptake of free fatty acids, they store a large fraction as triglycerides (TAG). We report here that both Toll-like receptor 4 (TLR4) and TLR2 agonists stimulate prolonged TAG retention by primary murine and human macrophages. Agonist-induced TAG storage lasted at least 72-96 hrs in vitro and was associated with increases in fatty acid (FA) uptake, FA esterification, and FA incorporation into TAG; FA oxidation decreased. The results of expression and inhibitor studies support a prominent role for increases in long chain acyl CoA synthase 1 (ACSL1) and diacylglycerol acyltransferase-2 (DGAT2) during the sustained response to TLR2/4 activation; decreases in adipose triglyceride lipase (ATGL, Pnpla2) and monoacylglycerol lipase (MgII) may also contribute. Stimulated murine macrophages that retained TAG carried out phagocytosis more effectively and were protected from saturated fatty acid-induced cell death (lipotoxicity). TLR agonist-induced TAG retention in macrophages is thus an active, sustained process that may have important adaptive functions. It may also contribute to the persistence of lipid-laden macrophages in infected tissues, host susceptibility to some microbial pathogens, and the pathogenesis of atherosclerosis. RNA from macrophage loaded with Fatty Acids, stimulated with bacterial lipopolysaccharides (LPS), or both compared to untreated controls (FA, LPS, FA+LPS, untreated). Project description:We conducted proteome analysis of basilar (cerebral) arteries from three control baboon fetuses and four fetuses that were exposed to alcohol in utero. Three alcohol-exposure episodes took place during second trimester-equivalent of human pregnancy, while fetal arteries were harvested during cesarean sections performed near-term. Supernatants from whole artery lysates were processed for TMT-labeling, fractionated, and subjected to LC/MS analysis.

Project description:To investigate fetal gene expression in obese compared to lean women in the second trimester, by performing global gene expression analysis of amniotic fluid (AF) cell-free RNA Analysis of paired data from obese cases and lean controls revealed differential expression of 205 genes. Functional analysis of differentially expressed genes suggested down-regulation of apoptosis in fetuses of obese women, particularly within nervous system pathways involving the cerebral cortex, and activation of pro-estrogenic, pro-inflammatory transcriptional regulators. Prospective whole transcriptome microarray study analyzing cell-free RNA in AF from obese cases and lean controls in the second trimester. Significantly differentially-regulated genes in fetuses of obese cases (N= 8) vs. matched lean controls (N = 8) were identified and functional analyses were performed. Tissue-specific differential gene expression in fetuses of obese women was also examined.
