Project description:Identification of early transcriptional responses to CPR5 function loss in Arabidopsis Expression of CPR5 C terminal half efficiently interferes with native CPR5 protein function and causes a dominant negative effect. Four-week-old leaves of Col-0 WT and transgenic Dex:YFP-CPR5-C plant were sprayed with water or 50 uM dexamethasone to induce expression of CPR5-C. Samples were collected at 24 hours after treatment. Three biological replicates were performed per genotype per treatment.

The dependency of cpr5-altered genes on SIM and SMR1 in Arabidopsis

Project description:In plants, effector-triggered immunity (ETI) is often associated with programmed cell death (PCD). Although the intracellular immune receptors involved in ETI have been studied extensively, how their activation leads to PCD and disease resistance is poorly understood. We found that the Arabidopsis nuclear envelope protein, CPR5 (constitutive expresser of PR genes 5), plays a crucial role in controlling cell fate in response to stress, as the cpr5 mutant exhibits spontaneous cell death and heightened immunity. A genetic screen revealed that the Cip/Kip CKIs (cyclin-dependent kinase inhibitors), SIM (siamese) and SMR1 (siamese-related 1), are essential for CPR5 signaling, as the sim smr1 double mutant fully suppressed the cpr5 phenotype. More significantly, PCD and ETI are compromised in sim smr1 even with the wild-type CPR5.

Project description:Innate immune responses of plant cells confer the first line of defence against pathogens. Signals generated by activated receptors are integrated inside the cell and converge on transcriptional programmes in the nucleus. The barley NLR intracellular receptor MLA is fully functionnal in Arabidopsis and the MLA N-terminal coiled-coil domain (MLACC) is sufficient for initiating the MLA-dependent immune signalling. Using RNA-seq, we analyzed the early MLACC-mediated transcriptional changes over time in a dexamethasone-inducible and pathogen-independent system.

Transcription profiling by array of Arabidopsis transgenic plants overexpressing YFP and VND7-YFP

Project description:The Arabidopsis thaliana NAC domain transcription factor, VASCULAR-RELATED NAC-DOMAIN7 (VND7), acts as a master regulator for differentiation of xylem vessels. In order to identify a set of genes regulated by VND7, we carried out a microarray experiment with the Arabidopsis full-genome GeneChip array ATH1 (Affymetrix) for transgenic Arabidopsis roots overexpressing VND7. Experiment Overall Design: Total RNAs were isolated from roots of 5-day-old seedling of transgenic plants overexpressing YFP and VND7-YFP under control of Cauliflower mosaic virus 35S promoter. Three independent biological replicates were performed for each sample.

XPO4 negatively regulates immune activation in the cpr5 mutant

Project description:Purpose: To understand the severe phenotype caused by the genetic interaction between CPR5 and XPO4, we performed the whole genome transcriptome analysis on 10-day-old seedlings of WT, xpo4, cpr5, and xpo4cpr5. Conclusions: XPO4 is a critical negative regulator of plant immunity, which is required to prevent an otherwise catastrophic induction of immune and cell death responses in the cpr5 mutant.

Gene expression profiling in senescence leaves from senescence-inducible LEC2 in Arabidopsis

Project description:The profiling was conducted with the 385K Microarray manufactured at NimbleGen, Inc (http://www.nimblegen.com/) designed from 30,386 genes of Arabidopsis (TAIR v.6). RNA samples of leaves of S1 (100% green leaf from 40 days plant) and S3 (more than 50% yellow leaf) senescence stages from senescence-inducible LEC2 transgenic plant, OIL21 were subjected for analysis of Arabidopsis 385K transcriptome. Expression of 3,025 genes were significantly changed with log2 ratios greater than 1.0 or less than -1.0 in S1 or S3 stage (p value <0.05), among which 1491 were up-regulated and 1534 were down regulated. Gene annotation revealed that 7.4% genes, 223 out of 3,025 changed by LEC2 were encoded transcription factors (TF). 106 TFs were up-regulated and 115 TFs were down-regulated.
