Effects of low-doses of antiandrogenic substances on Wistar Rat testes [Transcriptome analysis]

ABSTRACT: Three antiandrogenic substances (Flutamide, Prochloraz, Vinclozolin) were administered in aqueous preparations at 3 doses: i) Acceptable Daily Intake (ADI) ; ii) No Observed Adverse Effect Level (NOAEL) ; iii) Low Observed Adverse Effect Level (LOAEL) . Pregnant females (20/group) were treated by gavage from gestation day (GD) 6 until postnatal (PND) day 21. After weaning, the offspring received the same test-substance as their mothers by oral gavage. Twenty males were selected to be raised until the weaning (Subset 1, PND 21), the day of sexual maturation (Subset 2, puberty) or early adulthood (Subset 3, PND 83±2). At each subset, testes were used for transcriptome and miRNome analyses.

Mammary Gland Morphology and Gene Expression Signature of Prepubertal Male and Female Rats Following Exposure to Exogenous Estradiol

Project description:In order to properly understand whether xenoestrogens act as estrogens, it is essential to possess a solid portrait of the physiological effects of exogenous estradiol. Because the estrogen-dependent gene expression is one of the primary biomarkers of estrogenic action, we have assessed effects of three doses of exogenous estradiol (0.1, 1.0 and 10 ug/kg of body weight/day) on the mammary gland morphology and gene expression profiles by microarray analysis of prepubertal male and female rats of both sexes compared to untreated controls. Estradiol was administered subcutaneously with minipumps from weaning at PND21 to the end of the experiment at PND33. The data suggest that the male mammary is a sensitive tissue for estrogenicity assessment. Groups of male and female rats were infused subcutaneously with mini-osmotic pumps with 17-beta-estradiol from weaning on post-natal day (PND) 21 until PND 33. Pumps delivered doses of 0, 0.1, 1.0 and 10 ug/kg/d. At sacrifice (PND 33) abdominal mammary gland pairs were collected and fresh frozen in liquid nitrogen.

Expression data from PBDE treated rats at PND22 and Week 13

Project description:Liver gene transcripts patterns were used to characterize toxicity from exposure to polybrominated diphenyl ethers (PBDEs), flame retardant components. In this study, Wistar Han dams were exposed by gavage to the PBDE mixture (DE71) starting at gestation day 6 (GD 6) and continuing to weaning on postnatal day 21 (PND 21). Offspring from the dams began PBDE direct dosing on PND 12 and were dosed daily through PND 21. After weaning, they were dosed 5 days per week for another 13 weeks. Liver samples were collected at PND 22 and week 13 for liver gene expression analysis and interrogated with the Affymetrix Rat Genome 230 2.0 Array. PBDE treatment induced 1,066 liver gene transcript changes in females and 1,200 transcriptional changes in males at PND 22 (false discovery rate (FDR) < 0.01), but only 263 liver transcriptional changes at 13 weeks in male rats (FDR <0.05). No significant differences in dose response were found between male and female pups. There were a total of 6 groups and 5x replication for each group, for 30 total samples that were analyzed. The groups were (1) pup-male-CTL, (2) pup-female-CTL, (3) pup-male-PBDE, (4) pup-female-PBDE, (5) rat-male-CTL, (6) rat-male-PBDE. We generated the following pairwise comparisons using R/maanova: malePups(PBDE vs CTL), femalePups(PBDE vs CTL), maleRats(PBDE vs CTL), CTLpups(male vs female), PBDEpups(male vs female). We also performed ANOVA test for SEX-by-DOSE (pups) and AGE-by-DOSE (males). For pups, genes with an FDR≤1% were selected; for rats, genes with FDR < 5% were selected.

Dietary zinc deficiency and supplementation in early life and mammary tumors development in adulthood female rats

Project description:Zinc deficiency during pregnancy and postnatal life can adversely affect the health and predispose to an increased risk of developing human diseases at adulthood. The present study was designed to evaluate whether dietary zinc deficiency or supplementation during the pregnancy, lactation and juvenile stages interferes with the development of mammary tumors induced by 7,12-dimethylbenzanthracene (DMBA) in female Sprague-Dawley (SD) rats. Pregnant females SD were allocated into three groups: dams received diets containing adequate (35 mg/Kg chow), deficient (3 mg/Kg chow) or supplemented (180 mg/Kg chow) zinc levels during gestational day 10 (GD 10) until the litters weaning. Female offspring were allocated into three groups and received the same diets as their dams until postnatal day (PND) 51. At PND 51, females SD (n=16 each group) received a single dose of DMBA (50 mg/kg, ig) for initiation of mammary carcinogenesis, adequate zinc diets and were euthanized at PND 180. Tumors samples were collected and processed for histological evaluation and microarray analysis. Dietary zinc deficiency induced a significant reduction in female offspring body weight evolution at parturition, weaning and pubertal phases. At late in life, the early in life dietary zinc deficiency or supplementation did not alter the latency, incidence, multiplicity and volume or histological types of mammary tumors in relation to the adequate zinc group, and changed the expression of a small amount of genes. The present findings indicate that early-in life dietary zinc deficiency or supplementation did not significantly modify the susceptibility to development of mammary tumors induced by DMBA.

Microarray analysis of rat testis following combined fetal and postnatal DBP exposure

Project description:Provided later Pregnant Fisher 344 rats will be purchased from Charles River Laboratories, Inc. and delivered to CIIT on gestational day (GD) 7 (GD0 = day first vaginal plug positive). At gestational day 12 (GD12), the dams will be exposed once/day until GD20 to 50 mg/kg dibutyl phthalate (DBP) in corn oil vehicle via oral gavage. Each dose group will contain 4-6 vehicle control or phthalate treated dams. Groups of animals will be sacrificed at GD20, postnatal day (PND) 35, and PND90 for endpoint analysis. At GD20, treated and control animals will be examined for various endpoints including body weight, testicular histopathology, gene expression profile via microarray analysis, and anogenital distance (AGD). AGD (at parturition; PND1) and nipple number/location (at PND14 and day of sacrifice) will be determined on animals in the postnatal groups. At PND35 or 90, one male from each in utero corn oil vehicle or DBP exposed group will receive a second gavage of either corn oil or 500 mg/kg DBP. 6 hours after the second gavage, the following endpoints will be examined: 1) testis histopathology; 2) spermatid head quantification (PND90 only); 3) testis and body weights; 5) genome-wide gene expression (via microarray); and 6) germ cell apoptosis (TUNEL assay).

Project description:This study investigates the effects of non-thermal levels of 900 MHz radiofrequency electromagnetic field (RF-EMF) which is used in mobile phone technology on neurodevelopment and neural stem cell differentiation. In vivo in the rat, the effects of pre- and post-natal 0.08 and 0.4 W/kg exposure were assessed on the proteomic profile at PND postnatal day 0 (PND) and on proliferation, synaptogenesis, and oxidative stress in the hippocampus and primary motor cortex of rats at PND 8 and PND 17. Complementary Mechanistic endpoints on cell differentiation were studied in vitro using stem cells exposed to 900 MHz RF-EMF at a non-thermal level for 3 or 7 days in vitro at the lowest SAR (0.08 W/kg). Our in vivo results showed a decrease in BDNF level and cells proliferation with a decrease in synapses balance. Increased proliferation, apoptosis, and double-strand DNA breaks in neural stem cells (NSCs) were also observed. Smaller ratio of B1 cells and a higher ratio of oligodendrocyte progenitor cells and astrocytes were observed in the exposed NSPCs. These findings suggest that developmental RF-EMF exposure induce changes in neurodevelopment in vivo from PND 0 and until PND 17 specifically on cellular proliferation supported by alterations in NSPCs in vitro, leading to the hypothesis of a vulnerability of developing central nervous system towards RF-EMF exposures at regulatory thresholds.
