Project description:Mesenchymal stem cells (MSCs) are a population of multipotent cells with an attractive ability to promote tissue repair by regulating regeneration and inflammation. Ironically, the immune regulatory capacity of MSCs, regardless of their tissue origin, is endowed by inflammatory cytokines (e.g., IFNg plus TNFa). Effective application of MSCs relies on the production of relatively homogeneous MSCs. However, the cellular heterogeneity, and the differentiation trajectories of in vitro expanded MSCs remains unclear. We profiled the transcriptomes of 361 single MSCs derived from two umbilical cords (UC-MSCs). The MSCs are from different passages and culture conditions with or without inflammatory cytokine stimulation. Weighted gene correlation network analysis for the expression of their highly variable genes revealed that UC-MSCs surprisingly possess only limited heterogeneity. Cell cycle based principal component analysis showed that the limited heterogeneity identified in these UC-MSCs was strongly associated with their entrance into the G2/M phase. This was further proven by the observation that one featured gene CD168 was expressed in a cell-cycle dependent manner. When CD168high UC-MSCs were sorted and cultured in vitro, they repopulated into a new community that is similar to the original status. Our results demonstrated that in vitro expanded UC-MSCs is a well-organized population with limited heterogeneity dominated by cell cycle status. Thus, our studies argues that standardization of MSCs for disease treatment is possible.

Project description:Human mesenchymal stem cells (MSCs) have demonstrated promise when delivered to damaged tissue or tissue defects for their cytokine secretion and inflammation modulation behaviors that can promote repair. Insulin-like growth factor 1 (IGF-1) has been shown to augment MSCs’ viability and survival and promote their secretion of cytokines that signal to endogenous cells, in the treatment of myocardial infarction, wound healing, and age-related diseases. Biomaterial cell carriers can be functionalized with growth factor-mimetic peptides to enhance MSC function while promoting cell retention and minimizing off-target effects seen with direct administration of soluble growth factors. Here, we functionalized alginate hydrogels with three distinct IGF-1 peptide mimetics and the integrin-binding peptide, cyclic RGD. One IGF-1 peptide mimetic (IGM-3) was found to activate Akt signaling and support survival of serum-deprived MSCs. MSCs encapsulated in alginate hydrogels that presented both IGM-3 and cRGD showed a significant reduction in pro-inflammatory cytokine secretion when challenged with interleukin-1β. Finally, MSCs cultured within the cRGD/IGM-3 hydrogels were able to blunt pro-inflammatory gene expression of human primary cells from degenerated intervertebral discs. These studies indicate the potential to leverage cell adhesive and IGF-1 growth factor peptide mimetics together to control therapeutic secretory behavior of MSCs.

Project description:UC-MSCs have been classified as an alternative niche for expanding HSPCs. However, inconsistent outcomes in hematopoietic recovery and the shift toward specific lineage differentiation were observed following co-cultures. Moreover, the underlying molecular mechanisms and regulatory roles of UC-MSCs during hematopoiesis have not been fully understood. The co-cultures of UC-MSCs UCB-CD34+ cells were established. Integrative transcriptomic analysis of UC-MSCs collected on day 3 of co-cultures has highlighted the molecular signature and comprehensive regulatory networks. The lncRNA-RNA binding protein interaction network and lncRNA cis- and trans-regulation networks were evident. The 3 gene modules and 10 hub genes were identified in the protein-protein interaction (PPI) network, including RPS16, CD74, RPL35, COX7C, RPL38, RPS28, RPS27, RPS10, TARDBP, and TOMM7. These have shed light on niche activity of UC-MSCs in regulation of cell differentiation, genomic stability, and hematopoietic supportive during co-culture with HSPCs.

Project description:Discography is an imaging procedure typically performed on patients who report extended periods of pain.Usually radiocontrast agents are used to diagnose the discogenic low back pain. This study was focused on investigating the effects of two clinically used, non-ionic radiocontrast dyes, isovue-370 and omnipaque-240, on the human umbilical cord mesenchymal stem cells (UC-MSCs). Human UC-MSCs were cultured and treated with various concentrations of Omnipaque and Isovue for 24 hr. The cell growth was inhibited by both the dyes. The results showed that both the dyes significantly affected the cell morphology and viability as well as caused apoptosis in UC-MSCs. The global gene expression analysis showed that the most effected genes were those involved in the apoptosis, DNA damage and toxicity.

We evaluated the effects of human adipose tissue-derived MSCs (AD-MSCs) and umbilical cord tissue-derived MSCs (UC-MSC) in a dextran sulfate sodium (DSS)-induced mouse model. Methods: Human AD-MSCs and UC-MSCs (1 × 106 cells) were injected intravenously into a 7-day DSS-induced colitis model. The therapeutic effects of cell origin, injection timing, and supernatants obtained from MSC cultures were evaluated. We also analyzed mRNA expression in MSCs, tissues, and intestinal flora. Results: AD-MSCs and UC-MSCs had strong anti-inflammatory effects when injected on day 3 in a mouse model. On day 11, mRNA levels of inflammatory factors in colon tissues were significantly decreased after injection of MSCs on day 3. Supernatants from MSCs culture decreased mRNA levels of tumor necrosis factor (Tnf)-α, but had reduced therapeutic effects compared with MSC cell injection. RNA sequencing using colon tissues obtained the day after cell injection revealed changes in the TNF-α/nuclear factor-κB and T-cell receptor signaling pathways. Additional analyses showed that several factors, including chromosome 10 open reading frame 54, stanniocalcin-1, and TNF receptor superfamily member 11b were increased in MSCs after adding serum from DSS colitis mice. Furthermore, both AD-MSCs and UC-MSCs maintained the balance of intestinal flora. Conclusion: AD-MSCs and UC-MSCs showed therapeutic effects against inflammation after early cell injection while maintaining the intestinal flora. Although supernatants showed therapeutic effects, cell injection was more effective against inflammation.

Project description:Umbilical cord blood-derived mesenchymal stem cells (UCB-MSCs) have attracted attention as a potential cell therapy due to their immunomodulatory properties, paracrine trophic effects, and differentiation capacity that promote tissue repair and functional recovery. However, exposure to diabetic conditions impairs mitochondrial function in UCB-MSCs, leading to reduced stem cell activity and increased cell death, ultimately compromising their therapeutic efficacy. Hyperglycemic stress abnormally activates mTOR signaling, driving dysregulated glucose metabolism and mitochondrial dysfunction. To assess whether everolimus, an mTOR inhibitor, can mitigate these effects, we examined glucose metabolism-related enzyme expression in UCB-MSCs exposed to high glucose with or without everolimus treatment. Microarray profiling targeted 84 glucose metabolism-related genes, including eight housekeeping genes, of which 64 genes were reliably detected. Comparison of high glucose (H) versus high glucose plus everolimus (HE) conditions (two replicates each) revealed limited transcriptional changes, with only a subset of genes showing differential expression between groups. Overall, these results indicate that everolimus does not significantly alter glucose metabolism gene expression in UCB-MSCs.
