Affymetrix SNP array data for long time/short time survivors

ABSTRACT: Ovarian cancer is the seventh-most common cancer and the eight-most common cause of death from cancer in woman. Some women with ovarian cancer survive a long time (> 10yrs). We want to know if there’s a genetic difference between these long survivors and the broader population of interest. Affymetrix SNP arrays were performed according to the manufacturer's directions on sample DNA.

Gene expression profile in human ovarian granulosa cells during long-term in vitro culture

Project description:Granulosa cells (GCs) have many endocrine functions. However, in long-term in vitro culture GCs can change their properties. GCs were collected from hyper-stimulated ovarian follicles from woman during IVF procedures. They were cultured in in vitro long-term culture. RNA was collected after 1, 7, 15 and 30 days of culture. Expression microarrays were used for analysis, which allowed to identify groups of genes characteristic for particular cellular processes. In this study, we demonstrated the gene expression profile of long time primary cultured human ovarian granulosa cells.

Expression data from mouse ovarian surface epithelium cells at different stages of malignancy

Project description:Ovarian cancer is one of the most deadly cancers accounting for only 3% of diagnosed cancers, but is the fifth leading cause of cancer deaths among woman; however, the progression of ovarian cancer is poorly understood. To study and further understand the early events that lead to epithelial derived ovarian cancer, we previously developed a cell model of progressive ovarian cancer. Mouse ovarian surface epithelial (MOSE) cells have undergone spontaneous transformation in cell culture and represent pre-neoplastic, non-tumorigenic to an aggressive malignant phenotype. Microarray analysis was performed with RNA isolated from different stages of MOSE cells to examine changes in gene expression as MOSE cells transition from a pre-neoplastic to a malignant state.

Expression data from the CTCR-OV01 study

Project description:All patients with suspected ovarian cancer (Raised CA 125 and a complex pelvic mass in a perimenopausal woman) were radiologically staged using CT scan and a chest x-ray. Patients with evidence of intra-abdominal metastasis and/or malignant pleural effusion were approached for entry to the study. Tissue biopsy was obtained either under radiological control (core needle biopsy) or via laparoscopic surgery (punch biopsy). Patients with histologicaly confirmed epithelial ovarian cancer were randomized to receive either three cycles of carboplatin (AUC 7) or paclitaxel (175 mg/m2). Patients with suspected ovarian cancer had a biopsy obtained before and after 3 cycles of either Carboplatin or Paclitaxel. Samples were immediately frozen for RNA extraction.

Cancer-associated fibroblasts-secreted CXCL14-regulated LINC00092 mediates ovarian cancer progression by altering glycolysis

Project description:Epithelial ovarian cancer (EOC) constitutes a major gynecological malignancy, with a reported incidence rate of 3-12/100 000 woman annually. As early symptoms of ovarian cancer are often clinically atypical or absent, the majority of ovarian cancer patients are diagnosed at a late stage, when the five-year survival rate is extremely low. This condition underscores the urgency of early detection of these patients and establishment of new therapeutic targets for successful intervention. Considering that the predominant biological characteristic that differentiates malignant from benign tumors is the ability to metastasize, it is necessary to identify novel metastasis-related molecules for ovarian cancer. In this study, we found that CAFs could significantly increase the metastatic potential of ovarian cancer cells compared with non-cancer associated fibroblasts(NAFs), which is associated with over-expression of CXCL14 in CAFs. We examined the impact of CAF-secreted CXCL14 on the lncRNA expression profiles in ovarian cancer during metastasis. We treated A2780s ovarian cancer cell line with recombinant CXCL14 protein and control respectively and subjected them to Arraystar Human LncRNA microarray v3.0 to profile differential lncRNAs in ovarian cancer upon treatment of CXCL14

Investigation of the potential mechanisms underlying nuclear F-actin organization in ovarian cancer cells by high-throughput screening in combination with deep learning

Project description:We sequenced a set of ovarian cancer cell lines with or without overexpression of a fluorescent nuclear F-actin probe. It is to compare different ovarian cancer cell types which have distinct nuclear F-actin patterns, and try to seek the difference behind that phenotype. Meanwhile, we also want to check whether the nuclear F-actin probe would affect the transcriptional profile of the cell line.
