Project description:Total RNA samples were isolated from M. catarrhalis grown in BHI (iron-replete condition, designated DF0) and in BHI containing 30 mM Desferal (iron-limiting condition, designated DF30). Total RNA and M. catarrhalis genome-directed primers (GDPs) were used for synthesis of Cy3- or Cy5-labeled cDNA samples. Three independent M. catarrhalis growth experiments and total RNA isolations were used for synthesis of cDNA samples, and a total of six DNA microarray hybridizations were performed in this study. Transcriptional Response to Iron Limitation in Pelagibacter ubique

Project description:We investigated the gene expression responses of Candidatus Pelagibacter ubique cultures to iron limitation. Differential expression was observed for genes in iron acquisition and incorporation operons. SfuC in particular was 16 times higher in iron-limited cultures and encodes a periplasmic iron-binding protein. Six natural seawater cultures were amended with minimal nutrients and inoculated with P. ubique. Close to maximum cell density, all carboys were supplemented with 100 nM ferrichrome (an iron-chelating siderophore) and three carboys were additionally supplemented with 1 µM FeCl3. Each of the six carboys was sampled for microarray analyses one, two, and eleven days after the ferrichrome addition.

Project description:The iron-binding glycoprotein lactoferrin exhibits anti-bacterial activity. To investigate the overall response of Bacillus cereus to lactoferrin, total transcriptome analyses were performed. Growth conditions were mMOD minimal medium, minimal medium plus 10 mg/ml lactoferrin, as well as minimal medium without iron source for comparison. Differential regulation of genes involved in iron acquisition as well as genes encoding enterotoxins and further virulence factors was of special interest.
