No template was used as a negative control Triplicates for each line were analyses for epigenome-wide methylation (>480,000 CpG sites) using Infinium Human Methylation 450 BeadChip Kit (Illumina).

Differential blood biomarkers and PBMC transcriptome profiles in normal-weight and obese individuals

Project description:The aim of study was to investigate correlation bewteen peripheral blood mononuclear cell (PBMC) transcriptome profiles and plasma lipid profiles of Korean adult with varying BMI. In Korean, BMI cut-off points are 18.5~22.9 kg/m2 (normal range), ≥ 23 kg/m2 (overweight), ≥ 25 kg/m2 (obese), ≥ 30 kg/m2 (severely obese). Thus, the obese group was subdivided into mildly obese (BMI 25~27 kg/m2, OA) and highly obese (BMI 27~30 kg/m2, OB). This study indicates that lipid, glucose and inflammation metabolism-related gene expressions were altered according to cahnge of varing phenotype biomarkers such as BMI, plasma total-C, TG, FFA and HDL-C. Thus, blood biomarkers and PBMC gene expression profiles identified in this study may be useful as indicative biomarkers for obese susceptibility in Korean adult as well as response to various intervention for treating obesity.

Whole tissue mRNA expression from severely obese individuals: subcutaneous and visceral adipose tissues

Project description:Subcutaneous adipose tissue and visceral adipose tissue samples were obtained from severely obese individuals that underwent bariatric surgery. The goal of this study was to compare genome-wide gene expression levels in the two tissue types from healthy and unhealthy severely obese individuals. Whole-transcriptome subcutaneous adipose tissue gene expression levels were determined in 73 individuals with a BMI >35 kg/m2. Whole-transcriptome visceral adipose tissue gene expression levels were determined in 69 individuals with a BMI >35 kg/m2. Modules of co-expressed genes likely to be functionally related were identfied and correlated with BMI, plasma levels of glucose, insulin, HbA1c, triglycerides, non-esterified fatty acids, ALAT, ASAT, C-reactive protein, and LDL- and HDL cholesterol.

Microarray of isolated adipocytes from subcutaneous adipose tissue of severely obese patients compared to lean controls

Project description:The aim of the project was to compare global gene expression in adipocytes from obese patients and lean controls. Subcutaneous adipose tissue was collected from severely obese patients undergoing bariatric surgery (average body-mass index (BMI) of 45.5 kg/m2 (n = 12, thereof 4 men) and healthy lean patients undergoing hernia repairs (average BMI of 24.2 kg/m2 (n = 12, thereof 7 men), between 27 and 56 years of age. Adipocytes were isolated by collagenase treatment of adipose tissue, followed by filtering and centrifugation. Floating adipocytes were lysed in Qiazol before RNA purification and microarray analysis.

Expression data from Adipose Stem Cells (ASC) from morbidly obese and non-obese individuals

Project description:The adipose tissue is an endocrine regulator and a risk factor for atherosclerosis and cardiovascular disease when by excessive accumulation induces obesity. Although the adipose tissue is also a reservoir for stem cells (ASC) their function and “stemcellness” has been questioned. Our aim was to investigate the mechanisms by which obesity affects subcutaneous white adipose tissue (WAT) stem cells. We used microarrays to analyze differences in transcriptomic profiles between the adipose stem cells from morbidly obese and non-obese individuals. Subcutaneous white adipose tissues that were obtained during bariatric surgery (Obese) or liposuction (Lean) were donated by patients after obtaining informed consent. Three cases with BMI>40 kg/m2 (ASCmo) and three controls with BMI<25 kg/m2 (ASCn) were selected and processed extracting total RNA, processing and hybridizating on microarrays.

Project description:Mesenchymal stem cells (MSCs) are generally considered the main tool box for cell-based therapies and compared to embryonic stem cells (ESCs), MSCs exhibit many advantages. It has been shown that UC matrix (the Wharton’s Jelly surrounding umbilical vessels) contains a great number of mesenchymal cells. The abundant amount of Wharton’s Jelly (WJ) makes it an attractive source of MSCs for cell-based therapies. recently have been highlighted the changes in protein expression profiling during in vitro expansion of WJ-MSC, probably related to the gradual reduction of their staminal plasticity and to the biological cellular mechanism occurring in the cellular aging. The aim of the present work was to analyze the transcriptomic profile useful to the definition of WJ-MSC characteristics and therefore identify possible different new marker candidates. In these contest, changes in gene expression profiles occurring during WJ-MSC in vitro growth wile by analyzed in order to find possible modulation related to their long expansion and fast growth abilities. We analyzed the changes in total RNA expression at the 12th in vitro expansion cycle compared to the 4th in vitro expansion cycle.

Gene expression patterns of mesenchymal stem cells from bone marrow and Wharton's jelly of umbilical cord

Project description:Mesenchymal stem cells (MSCs) are promising tools for the treatment of diseases such as infarcted myocardia and strokes because of their ability to promote endogenous angiogenesis and neurogenesis via a variety of secreted factors. MSCs found in the Wharton's jelly of the human umbilical cord are easily obtained and are capable of transplantation without rejection. We isolated MSCs from WhartonM-bM-^@M-^Ys jelly and bone marrow (WJ-MSCs and BM-MSCs, respectively) and compared their transcriptional profiles. In this study, BM-MSCs and WJ-MSCs were isolated from 4 and 3 independent donors, respectively.

The study group consisted of 17 obese women with normal glucose tolerance and 15 obese women with T2DM classified according to WHO standards. The groups were matched for age, BMI and waist circumference. All the women had been morbidly obese (BMI>40 kg/m2) for at least five years.
