Differential gene expression of Andrographis paniculata and andrographolide enriched andrographis paniculata extract in liver cells

ABSTRACT: Andrographis paniculata Nees and its major compound andrographolide is known to exhibit a pleothera of activities. Active component enriched extracts are known to provide various beneficial effects, and therefore it was interesting to investigate whether increased amount of major compound alters gene expression. To acheive this, HePG2 cells treated with Andrographis paniculata extract (AP) and andrographolide enriched –extract (AP20) were subjected to microarray analysis to check their effect on global gene expression.

Oct3/4 expression and notch signaling in bone marrow stromal cells reprogrammed by Xenopus egg extract

Project description:Bone marrow stromal cells (BMSCs) are multipotent stem cells that preferentially differentiate into mesenchymal cells. If they can be dedifferentiated into embryonic stem cell-like cells, they will be a highly attractive source for cell therapy. Cell and egg extracts have been used in a few studies to evaluate nuclear reprogramming, but these have not examined cell pluripotency in any detail. In this study, we used a cell reversible permeabilization method to treat BMSC with Xenopus laevis mitotic egg extract. We observed an upregulation of the pluripotent protein Oct3/4 in BMSCs treated by this extract. We also further evaluated transcriptional changes with a focused stem cell oligonucleotide array. A number of genes involved in the Notch or Wnt signaling pathways were upregulated in BMSC exposed to Xenopus egg extract. In conclusion, our microarray data from BMSCs exposure to egg extracts may provide interesting clues regarding factors involved in nuclear reprogramming. Our approach is an alternative method towards dedifferentiation of cells without genetic modification, which is preferable in the clinical situation. Keywords: Cell type comparison

Analysis of global transcriptomic response of lettuce seedlings (Lactuca sativa L.) to microalgae extract used as biostimulant agents

Project description:The use of new natural eco-sustainable products is becoming an interesting option in order to reduce the use of chemical fertilizers and increase crop yields. Seaweed extracts are gained major attention as plant biostimulants due to their positive effect on plant-growth as well as on improving plants’ tolerance against abiotic stresses. Among the horticulture crops, lettuce (Lactuca sativa L.) is a major fresh vegetable crop in the Mediterranean area, which often requires the use of natural biostimulants to improve both the quantity and quality of production. The aim of this work was to assess the effect of either Chlorella vulgaris or Scenedesmus quadricauda extracts on lettuce seedlings (Lactuca sativa L.) by motoring the induced transcriptomic modifications using a RNASeq approach. The results showed that both C. vulgaris and S. quadricauda extracts positively influence the growth of lettuce seedlings. However, a higher reprogramming of the gene expression occurred in the case of C. vulgaris treatment than in S. quadricauda extract. Considering the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway functional enrichments, the main KEGG terms are in the “Biosynthesis of secondary metabolites”, “Metabolic pathways”, “Carbon metabolism” and “Biosynthesis of amino acids” categories. This study lays the basis for understanding the mechanisms and processes triggered by the use of microalgal extracts, which can represent an easier-to-handle and cheaper method for an eco-sustainable cultivation of lettuce plants than the application of chemicals.

Comparative transcriptomic profiling of peripheral blood mononuclear cells (PBMCs) isolated from patients in secondary prevention of CVD taking placebo, grape extract or resveratrol-enriched grape extract

Project description:We used Affymetrix microarrays to investigate gene expression changes in PBMCs isolated from male patients ongoing secondary prevention of CVD to determine significant modulatory effects that may have been induced by the intake of an initial dose of 8 mg of resveratrol-enriched grape extract for 6 months and then, 16 mg for a further 6 months. The aim of this work was to determine whether the daily intake of dietary levels of resveratrol (RES) for a total of 12 months exerted any modulatory effects, at the level of gene expression, in PBMCs isolated from patients in secondary prevention of CVD. Male patients were divided in 3 groups: placebo (A), grape extract (B) and resveratrol-enriched grape extract (C). Total RNA was extracted from isolated PBMCs belonging to a total of 18 diabetic male patients (6 from each group) in 3 time points (at day 0, after 6 months and after 12 months) to compare differential gene expression between the groups. Project description:We used Affymetrix microarrays to investigate gene expression changes in the liver of lean female Zucker rats exposed to a normal diet supplemented with a rosemary extract rich in the diterpenic compound, carnosic acid (CA). The aim of this work was to determine whether the daily intake (with the standard diet) of the rosemary extract (RE) enriched in CA (40%) for 64 days exerted any modulatory effects, at the level of gene expression, in the liver of lean Zucker female rats. Female lean Zucker rats were randomly assigned to two experimental groups (n= 7 animals per group) as follows: (1) control group, fed a standard diet (CT); (2) treated group, fed a standard diet supplemented (0.05% w/w) with the RE rich in CA (RE). At the end of the experimental procedure (64 days), total RNA was extracted from the liver to compare differential gene expression between the two groups. Liver differential gene expression after 64 days of supplementation between the CT group and the RE group (effects of the extract supplementation).

RNA-seq analysis reveals modulation of inflammatory pathways by an enriched-triterpene natural extract in mouse and human macrophage cell lines

Project description:Chronic Inflammation has a key role in the development of insulin resistance and type 2 diabetes. Previously, we demonstrated that OBE100, a natural extract from the leaves of Eucalyptus tereticornis, has anti-inflammatory properties. Three pentacyclic triterpenoids, ursolic acid, oleanolic acid, and ursolic acid lactone are the major compounds present in OBE100. These molecules have shown multiple biological activities. In this study we analyzed how the compounds of OBE100 modify macrophage gene expression using RNA sequencing. Triterpenoids regulate the inflammatory program in activated macrophages inhibiting the expression of many cytokines, chemokines, and inflammatory mediators. However, the OBE100 extract has a more powerful immunomodulatory effect than the triterpene mixture increasing the number of genes regulated, both in mouse and human models. Our study shows that OBE100 is a promising extract for the treatment of diabetes that can break the link between inflammation and insulin resistance.
