Modeling human embryo adhesion using a microfluidic platform

Project description:Single-cell RNA-seq dataset of models of human embryo adhesion in a microfluidic platform (ADOC). Embryo adhesion represents the first step of implantation, yet understanding this process has been hindered by the lack of human in vitro platforms that replicate endometrial physiology. Here, we present a dual-channel microfluidic platform containing organoid-derived endometrial epithelium and primary stromal cells. Our model recapitulates important endometrial hallmarks including epithelial polarization, stromal decidualization, extracellular vesicle release, and hormone-induced receptivity. We tested its function by introducing mouse and human blastocysts and showed that embryos displayed features of initial adhesion. These included establishment of embryo-epithelial contacts initiated via the polar trophectoderm, inner cell mass repositioning, and lineage segregation. Moreover, human embryos secreted βhCG indicating a functional trophoblast. Thus, this work provides a platform to study key features of embryo adhesion and endometrial receptivity, and disorders affecting embryo-endometrium interactions.

A microfluidic co-culture platform to generate synaptically separated networks that uncover the role of astrocytes in the initiation and progression of neuron pathology

Project description:Communication between astrocytes and neurons plays a pivotal role in the development, maintenance and dysfunction of cellular networks within the central nervous system. Further understanding the contribution of astrocytes to the initiation and progression of neuropathology has, however, been hindered by the complexity of cell networks in traditional in vivo and ex vivo systems. We have designed and constructed a novel three-compartment microfluidic cell culture device that enables us to overcome these limitations and uncouple the roles of astrocytes and neurons in the transfer of pathology through complex cell networks. Our microfluidic device integrates a novel maze-like structure that prevents synaptic connectivity between two fluidically isolated neuron populations, while allowing astrocyte infiltration and growth throughout. We use primary neuron/astrocyte co-cultures, proteomic analysis and immunocytochemistry to validate the application of this device. Using this device, we identify and describe a novel calcium-dependent role for astrocytes in the transfer of excitotoxic pathology between the segregated neuron populations.

A plug and play microfluidic platform for standardized sensitive low-input Chromatin Immunoprecipitation

Project description:Epigenetic profiling by chromatin immunoprecipitation followed by sequencing (ChIP-seq) has become a powerful tool for genome-wide identification of regulatory elements, for defining transcriptional regulatory networks and for screening for biomarkers. However, the ChIP-seq protocol for low-input samples is laborious, time-consuming and suffers from experimental variation, resulting in poor reproducibility and low throughput. Although prototypic microfluidic ChIP-seq platforms have been developed, these are poorly transferable as they require sophisticated custom-made equipment and in-depth microfluidic and ChIP expertise, while lacking parallelisation. To enable standardized, automated ChIP-seq profiling of low-input samples, we constructed microfluidic PDMS-based plates capable of performing 24 sensitive ChIP reactions within 30 minutes hands-on time and 4.5 hours machine-running time. These disposable plates can be conveniently loaded into a widely available controller for pneumatics and thermocycling. In light of the Plug and Play (PnP) ChIP plates and workflow, we named our procedure PnP-ChIP-seq. We demonstrate high-quality ChIP-seq on hundreds to few thousands of cells for all six post-translational histone modifications that are included in the International Human Epigenome Consortium set of reference epigenomes. PnP-ChIP-seq robustly detects epigenetic differences on promoters and enhancers between naïve and more primed mouse embryonic stem cells (mESCs). Furthermore, we used our platform to generate epigenetic profiles of rare subpopulations of mESCs that resemble the 2-cell stage of embryonic development. PnP-ChIP-seq allows non-expert labs worldwide to conveniently run robust, standardized ChIP-seq, while its high-throughput, consistency and sensitivity paves the way towards large-scale profiling of precious sample types such as rare subpopulations of cells or biopsies.

High throughput microfluidic 3D biomimetic model enabling quantitative description of human breast tumor microenvironment

Project description:Cancer is driven by genetic aberrations in the tumor cells and fundamental changes in the tumor microenvironment (TME). These changes offer potential targets for novel therapeutics, yet lack of in vitro 3D models recapitulating this complex microenvironment impedes such progress. Here, we generated tumor-stroma spheroids reflecting the in vivo TME of early and advanced breast tumor, using a high throughput microfluidic system. The spheroids, composed of tumor cells, fibroblasts and activated immune cells, were generated and cultivated on-chip in alginate (Alg) or alginate-alginate sulfate (Alg/Alg-S) hydrogels.
