Investigating gene expression changes upon ageing in chm mutants

ABSTRACT: We report here mRNA-seq data of adult male Drosophila head tissues. We compare two different ages: young and midlife as well as chm/chameau (CG5229) heterozygous mutants.

Chameau (HBO1) regulates starvation in a temperature-dependent manner in Drosophila melanogaster

Project description:The body temperature of Drosophila melanogaster depends on the extrinsic temperature. Numerous studies in fruit flies have shown that environmental temperature has an effect on metabolism, lifespan and starvation resilience. We have previously shown that Chameau (Chm), a MYST-domain acetyltransferase, promotes aging but also increases starvation resilience. As starvation resilience is highly temperature dependent, we explored the effect of temperature on starvation resilience in fruit flies with reduced chm expression. Strikingly, we found that an increase of 2°C was sufficient to restore starvation resilience in chm mutants. The increase in temperature rescued the dampened expression of genes involved in insulin, hormone and starvation response as well as a reduced rate of weight loss and misregulation of trehalose, we observed in chm mutants at 23°C. Our data show that whereas Chm has an important role in regulating starvation at 23°C and below, it becomes obsolete at higher temperatures. Our finding that a gene plays an important role only under specific environmental conditions has important implications in light of the recent global change of climate conditions.

The fruit fly acetyltransferase chameau promotes starvation resilience at the expense of longevity

Project description:Proteins involved in cellular metabolism and molecular regulation can extend lifespan in various organisms in the laboratory. However, any improvement in aging would only provide an evolutionary benefit if the organisms were able to survive under non-ideal conditions. We have previously shown that Drosophila melanogaster carrying a loss of function allele of the acetyltransferase chameau (chm) has an increased healthy life span when fed ad libitum. Here, we show that loss of chm and a reduction of its activity result in a substantial reduction of weight and a decrease in starvation resistance. This phenotype is caused by a failure to properly regulate the genes and proteins required for energy storage and expenditure. The previously observed increase in survival time thus comes with the inability to prepare for and cope with nutrient stress. As the ability survive in environments with restricted food availability is likely a stronger evolutionary driver than the ability to live a long life, chm is still present in the organism’s genome despite its apparent negative effect on lifespan.

The acetyltransferase chameau (chm) functions as metabolic sensor in regulating energy homeostasis and starvation response in Drosophila melanogaster

Project description:The coupling of metabolism to the posttranslational modification of proteins has been suggested to play an important role in the modulation of protein function in response to physiological variation. This interplay could potentially establish feedback loops that buffer extreme environmental challenges. Protein acetylation is one such posttranslational protein modification that is tightly coupled to metabolic variation. Here, we show that the acetyltransferase chameau (chm), which affects life span in Drosophila, is also involved in the regulation of metabolism upon environmental cues. While chm haploinsufficiency increases life span in flies that have sufficient access to nutrients, it reduces their ability to cope with starvation. We hypothesize the starvation susceptibility in mutant flies is caused by their reduced ability to modulate energy homeostasis. Interestingly, this effect is highly dependent on the ambient temperature, suggesting that it evolved to allow organisms to adapt to a wider range of environmental conditions by modulating their energy metabolism.

Genome-wide single nucleotide polymorphism array analysis unveils the origin of heterozygous androgenetic complete moles

Project description:Heterozygous CHMs have been believed as synonymous with dispermic moles (two sperm origin). The possibility of diploid sperm origin has not been considered. We assessed whether heterozygous CHMs would be of dispermic or diploid sperm origin. In all the cases, centromeric zygosity was random, i.e., mixed status. Theoretically, if the heterozygous CHMs were of diploid sperm origin, the centromeric status will be all homozygous or all heterozygous. Thus, all the analyzed heterozygous CHMs were considered to be of dispermic origin. Three cases showed the trisomy while we did not notice them with STR analysis. Diploid sperm fertilization can be rare during the development of heterozygous androgenetic CHM.

Project description:Proteins involved in cellular metabolism and molecular regulation have been shown to extend lifespan in various laboratory organisms. However, for any improvement in aging to confer an evolutionary benefit, organisms must also be able to survive under non-ideal conditions. Earlier, we discovered that the loss of chm function and reduced activity resulted in several notable effects. Firstly, flies with a loss-of-function allele of chm exhibited an extended healthy lifespan when provided with an unrestricted food supply, indicating that chm plays a role in lifespan regulation. In this study, we demonstrate that these flies also experience a significant reduction in weight and decreased starvation resistance compared to flies with normal chm function. Furthermore, based on observations from transcriptomic, proteomic, and acetylome data, we hypothesize that chm is important for regulating metabolism in Drosophila, particularly in energy storage and utilization. Moreover, we demonstrate that reduced chm activity affects carbohydrate metabolism in flies at the basal level and during changes in nutrient status. In summary, this study suggests that chm is also required for starvation resilience by regulating carbohydrate metabolism, and the previously observed increase in survival time likely accompanies the inability to prepare for and cope with nutrient stress. Finally, as the ability to adapt and survive in a food-restricted environment was likely a stronger evolutionary driver than the ability to live a long life, chm is still present in the organism's genome despite its apparent deleterious effect on lifespan.
