Project description:The effects of 7.5 micromolar parthenolide (PTL) were assessed on primary CD34+ acute myelogenous leukemia specimens obtained from 12 patients. Experiment Overall Design: Acute myelogenous leukemia (AML) specimens were obtained from 12 patients and CD34+ cells were isolated. For each patient, cells were cultured in vitro and exposed to either 7.5 micromolar parthenolide (PTL) or left untreated (UT) for 6 h. Total RNA was then harvested for global gene expression analysis.

Parthenolide disrupts mitosis by inhibiting ZNF207/BUGZ-promoted kinetochore microtubule attachment

Project description:Parthenolide is a natural compound that has shown highly promising anticancer activity. Even though its mode of action has been studied for decades, its antimitotic activity has been largely overlooked, limiting the understanding of its full anticancer potential. In this study, we combined click chemistry with quantitative mass spectrometry and cell biology to elucidate the mechanism of action of parthenolide in mitosis. We show that parthenolide does not act as a microtubule-targeting agent in cells. Instead, it binds to the kinetochore protein ZNF207/BUGZ, preventing the establishment of proper kinetochore-microtubule attachment. Our results show that parthenolide covalently binds to Cys54 of BUGZ via Michael addition to its α-methylene-γ-lactone moiety. Since Cys54 is located within the second zinc finger domain of BUGZ microtubule-targeting region, we propose that parthenolide interferes with the microtubule-binding ability of BUGZ, consequently preventing kinetochore-microtubule attachments required for accurate chromosome congression to the spindle equator.

Project description:Desmocollin-1 (DSC1) is a desmosomal transmembrane glycoprotein that maintains cell-to-cell adhesion. DSC1 was previously associated with lymph node metastasis of luminal A breast tumors and was found to increase metastatic potential of MCF7 cells in vitro. Therefore, we focused on DSC1 role in cellular and molecular mechanisms in luminal A breast cancer and its possible therapeutic modulation using parthenolide. We performed comparative gene expression analysis using data obtained from RNA-seq of DSC1-overexpressing and control cells treated and untreated with parthenolide in biological duplicates.

Effects of parthenolide on amino acid metabolism and oxidative stress in lung adenocarcinoma based on quantitative proteomic analysis, targeted amino acid metabolomics, network pharmacology, and experimental validation

Project description:Background: Lung adenocarcinoma (LUAD) causes millions of deaths annually worldwide. Parthenolide (PTL), extracted from traditional Chinese herbal medicines, has various biological activities. In this study, we investigated the effects of PTL on amino acid metabolism and oxidative stress in LUAD cells. Methods: This study identified differential proteins and potential mechanisms of action of PTL in LUAD cells through label-free quantitative proteomics and protein-protein interaction networks. Combined with targeted amino acid metabolomics, we confirmed the results of GO and KEGG analyses. On this basis, the potential targets of PTL in LUAD were identified through network pharmacology, molecular simulation docking, and molecular dynamics simulations. Finally, the effects of PTL on amino acid metabolism and oxidative stress in LUAD were verified using in vivo and in vitro experiments. Results: PTL treatment of LUAD cells resulted in significant changes in expression of 157 proteins. GO and KEGG enrichment analyses showed that these proteins were involved in amino acid metabolism and oxidative stress response. Targeted amino acid metabolomics further confirmed that PTL affected amino acid metabolism in LUAD. Network pharmacology, molecular docking, and molecular dynamics simulations identified GCTG as a potential target of PTL in LUAD. Meanwhile, in vitro and in vivo experimental results indicated that PTL targeting GCTG affected the proliferation, amino acid metabolism, and oxidative stress levels of LUAD cells. Conclusion: PTL affects proliferation, amino acid metabolism, and oxidative stress levels of LUAD cells via targeting GCTG. Therefore, our study provides new insights into the prevention and treatment of LUAD with PTL, which may lay the foundation for future research directions
