MiRNA expression profiles in decidualized and non-decidualized endometriotic cyst stromal cells (ECSCs).

ABSTRACT: To identify the miRNAs which regulate the decidualization of ECSCs, we explored differentially expressed miRNAs between untreated ECSCs (n=4) and decidualized ECSCs (n=4) using miRNA microarray. The miRNA expression profiles in untreated ECSCs and decidualized ECSCs were obtained, then the miRNAs were filtered by signal intensity (50.0<, at least one out of 8 samples have values within range) and flags (at least 17% of cases have good flags). We established the criteria for regulated genes: ratio ≥2.0-fold for up-regulated genes and ratio ≤0.5 for down-regulated genes.

Project description:To identify the miRNAs which play a role in the development of endometriosis, we explored differentially expressed miRNAs between ECSCs (N=8) and normal endometrial stromal cells (NESCs, n=8) using miRNA microarray. The expression profiles in ECSCs and NESCs were obtained, then the miRNAs were filtered by signal intensity (20.0<, at least one out of 16 samples have values within range) and flags (at least 70% of cases have good flags). The differentially expressed miRNAs in ECSCs compared with NESCs were identified using volcano plot (Fold change >= 2.0, Mann-Whiteny U test p<0.05). The primary culture of ECSCs and NESCs (controls) were utilized. Total RNA was extracted from them, and miRNA microarray analysis was performed. The miRNAs differentially expressed between ECSCs and NESCs were identified, and aberrantly expressed miRNAs in ECSCs were defined as the endometriosis related miRNAs.

Project description:Genome-wide gene expression analysis of new cardiomyocytes (CMs) derived from resident c-kitpos endogenous cardiac stem cells (eCSCs) after myocardial injury by Isoproterenol (ISO) in mice. These data show that of new CMs derived from resident c-kitpos eCSCs have a gene expression profile that closely resembles the specific gene expression of adult cardiomyocytes. This result supports a role of c-kitpos eCSCs in the regeneration and repair of myocardial damage. To test the identity and the degree of differentiation of new cardiomyocytes (CMs) derived from resident c-kitpos endogenous cardiac stem cells (eCSCs) after myocardial injury by Isoproterenol (ISO) in mice. Global gene expression profiles by microarray was obtained in c-kitpos eCSCs, c-kitpos eCSC-derived YFPpos CMs and normal adult CMs. c-kitposCD45neg eCSCs were isolated from B6.129X1-Gt(ROSA)26Sortm1(EYFP)Cos/J mice and harvested at 4th passage (n=3). YFPpos cardiomyocytes were FACS sorted from left ventricle apex of Lentirius-c-kit/cre recombined B6.129X1-Gt(ROSA)26Sortm1(EYFP)Cos/J mice 28 days after ISO (N=3).
