Ribosome quality control is a central protection mechanism for yeast exposed to Deoxynivalenol and Trichothecin

ABSTRACT: Trichothecenes, like deoxynivalenol (DON) are secondary metabolites of Fusarium species, and are major pollutants in cereal food and feed products. They inhibit eukaryotic protein synthesis, however with only partly resolved consequences for the cellular homeostasis. Here we report a systematic functional investigation of the effects of DON on exponential phase growing yeast cells using either short doses of DON or continuos treatment with DON.

Project description:Deoxynivalenol (DON) is an important Fusarium toxin of concern for food safety. The inhalation of powder contaminated with deoxynivalenol is possible and may cause lung toxicity. In this study, we investigated the gene expression profile of A549 cells treated with 0.2 microg/mL deoxynivalenol by microarray analysis. In total, 16 genes and 5 noncoding RNAs were significantly affected by deoxynivalenol treatment. 4 samples; 2 samples from nontreated cells and other 2 samples from 0.2 microg/mL DON-treated cells for 24 hours

Transcriptome analysis of Escherichia coli under deoxynivalenol and nivalenol conditions

Project description:We constructed E. coli transcriptome under deoxynivalenol (DON) and nivalenol (NIV) condition from Fusarium spp. To identify differentially expressed genes in mycotoxin condition, we compared mycotoxin (DON and NIV) transcriptome to acetonitrile (ACN) transcriptome as the solvent of myxotoxin. As a result, 435-929 transcripts were identified from all conditions, respectively.

Project description:Fusarium head blight (FHB) is a major disease of cereal crops caused by the fungus Fusarium graminearum (Fg). FHB affects the flowering heads (or spikes). This study compare the gene expression profile in wheat spikelets from the very susceptible spring wheat cultivar Roblin inoculated with either water (H2O), a Fg strain (GZ3639) producing the mycotoxin deoxynivalenol (+DON), or a GZ3639-derived Fg strain which has been inactivated at the Tri5 locus (-DON).

Transcriptome analysis of Caco-2 Cells upon deoxynivalenol and its acetylated exposure

Project description:Deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON) and 15-acetyldeoxynivalenol (15-ADON) are type B trichothecenes and are major pollutants in food and feed products. Although the toxicity of DON has been well documented, the information of toxicity of its acetylated derivative remains incomplete. To acquire more detailed insight in 3-ADON and 15-ADON, Caco-2 cells under 0.5 µM DON, 3-ADON and 15-ADON treatment for 24 h were subjected to RNA-seq analysis. In the present study, 2656, 3132 and 2425 differentially expressed genes (DEGs) were selected respectively and were enriched utilizing KEGG and GO database. The upregulation of ataxia-telangiectasia mutated kinase (ATM), WEE1 homolog 2 (WEE2) and downregulation of proliferating cell nuclear antigen (PCNA), minichromosome maintenance (MCMs), cyclin dependent kinase (CDKs), E2Fs indicate that the three toxins induced DNA damage, inhibition of DNA replication and cell cycle arrest in Caco-2 cells. Apart from these, the upregulation of sestrin (SENEs) and NEIL1 implied that the reason of DNA damage may attribute to oxidative stress. Our study provides an insight in the toxic mechanism of 3-ADON and 15-ADON.

RNA-Seq Revealed Differences in Transcriptomes between 3ADON and 15ADON Populations of Fusarium graminearum

Project description:Fusarium graminearum is the major causal agent of Fusarium head blight (FHB) in North America and other regions of the world. The pathogen causes direct yield losses and produces various types of trichothecenes mycotoxins [Deoxynivalenol (DON) and its acetylated forms (3-acetyl-4-deoxynivalenol=3ADON and 15-acetyl-4-deoxvevalenol=15ADON), nivalenol (NIV). Recent studies indicated that 3ADON-type isolates were significantly increased in North America and China in recent years and appears to be more aggressive based on growth rate, disease severity on different cultivars, and DON production in vitro. Thus the overall objective of this this study was to understand the molecular mechanisms that make 3ADON- and 15ADON-type populations different during infection using a susceptible cultivar, and to compare the transcriptomes of the 3ADON- and 15ADON-type populations in vitro and in planta using the RNA-seq approach as well as to identify the expression differences of candidate genes related to aggressiveness and DON production.

Influence of Deoxynivalenol-contaminated diet on the liver gene expression of chicken.

Project description:The mycotoxin deoxynivalenol (DON) is a secondary metabolite from Fusarium species and is frequently present on wheat and other cereals. The main effects of DON are a reduction of the feed intake and reduced weight gain of broilers. At the molecular level DON binds to the 60S ribosomal subunit and inhibits subsequently protein synthesis at the translational level. It has been suggested that cells and tissues with high protein turnover rate, like the liver and small intestine, are most affected by DON. However, little is known about other effects of DON e.g. at the transcriptional level. Therefore we decided to perform a microarray analysis, which allows us the investigation of thousands of transcripts in one experiment. The one-day old broiler chicks were separated into four groups. The diets consisted of a control diet and of three diets with moderate concentrations of 1.0, 2.5 and 5.0 mg DON/kg feed, which was attained by exchanging uncontaminated wheat with naturally DON-contaminated wheat up to the intended concentration. The chicken were held at standard conditions for 23 days and received their diet ad libitum. After slaughter the gene expression was determined in the liver of three samples per group.

Proteomic analysis using a two-dimensional differential gel electrophoresis (2D-DIGE) technique coupled with LC-MS/MS was used to investigate the quantitative or qualitative differences between the experimental variants.
