Arabidopsis mTERF9 protein promotes chloroplast ribosomal assembly and translation by establishing ribonucleoprotein interactions in vivo

Project description:The mitochondrial transcription termination factor proteins are nuclear-encoded nucleic acid binders defined by degenerate tandem helical-repeats of ~30 amino acids. They are found in metazoans and plants where they localize in organelles. In higher plants, the mTERF family comprises ~30 members and several of these have been linked to plant development and response to abiotic stress. However, knowledge of the molecular basis underlying these physiological effects is scare. We show that the Arabidopsis mTERF9 protein, promotes the accumulation of the 16S rRNA and the formation of the 23S rRNA first hidden break in chloroplasts, and co-immunoprecipitates with the 16S rRNA. Furthermore, mTERF9 is found in large complexes containing ribosomes and polysomes in chloroplasts. The analysis of the in vivo mTERF9 protein interactome identified many ribosomal proteins whose assembly into 70S ribosomes is compromised in the null mterf9 mutant. mTERF9 additionally interacts with putative ribosome biogenesis factors and CPN60 chaperonins. Yeast protein interaction assays revealed that mTERF9 has the capacity to directly interact with these proteins. Our data demonstrate that mTERF9 integrates protein-protein and protein-RNA interactions to promote chloroplast ribosomal assembly and translation. Besides extending our knowledge of mTERF molecular functions in plants, these findings provide important insight into the chloroplast ribosome biogenesis.

The plants were watered with nitrogen-sufficient or nitrogen-limiting solutions. 10 biological replicates were produced. For each sample, 3 rosettes were pooled to extract RNA. The libraries were generated with the Illumina Truseq stranded with ribozero plant leaf and sequenced on a NextSeq500 in single-end 75 bases.

Ribosome footprinting of wild-type Col-0 and rlmnl3 mutant

Project description:In the present study, we discover the presence of m2A in chloroplast rRNA and tRNA, as well as cytosolic tRNA, in multiple plant species. We identify six m2A-modified chloroplast tRNAs and two m2A-modified cytosolic tRNAs across different plants. Furthermore, we characterize three Arabidopsis m2A methyltransferases—RLMNL1, RLMNL2, and RLMNL3—which methylate chloroplast rRNA, chloroplast tRNA, and cytosolic tRNA, respectively. Our findings demonstrate that m2A37 promotes a relaxed conformation of tRNA, enhancing translation efficiency in chloroplast and cytosol by facilitating decoding of tandem m2A-tRNA-dependent codons. This study provides insights into the molecular function and biological significance of m2A, uncovering a layer of translation regulation in plants.

Arabidopsis TRM5 encodes a nuclear-localised tRNA guanine-N1-methyltransferase and is important for plant growth

Project description:We show that trm5 plants have less m1G in tRNAs, a slower leaf initiation rate, delayed flowering, and reduced primary root length. Late flowering correlates with reduced transcription of floral activators CO, FT, SOC1 and AP1. We also show that TRM5 methylates tRNA position 37 of several nucleus-encoded tRNA species, trm5 plants have reduced m1G and TRM5 is localized to the nucleus. Our findings highlight the importance of the post-transcriptional tRNA modification m1G37 in plant growth and development.

The thiolation modification of tRNA is essential for plant immunity

Project description:Plants have evolved sophisticated mechanisms to regulate gene expression to activate immune responses against pathogen infections. However, how the translation system contributes to plant immunity is largely unknown. The evolutionarily conserved thiolation modification of tRNA ensures efficient decoding during translation. Here we show that tRNA thiolation is required for plant immunity in Arabidopsis. The Arabidopsis cgb mutant is hyper-susceptible to the pathogen Pseudomonas syringae. CGB encodes ROL5, a homolog of yeast NCS6 required for tRNA thiolation. ROL5 physically interacts with CTU2, a homolog of yeast NCS2. Mutations in either ROL5 or CTU2 result in loss of tRNA thiolation. Further analyses reveal that tRNA thiolation is required for both transcriptional reprogramming and translational reprogramming during immune responses. The translation efficiency of immune-related proteins reduces when tRNA thiolation is absent. Our study not only uncovers a new biological function of tRNA thiolation but also reveals a new mechanism for plant immunity.
