Nonkeratinizing Carcinoma Tissues: Tumor vs. Adjacent non-tumor tissues

ABSTRACT: Nasopharyngeal carcinoma (NPC) is one of the common malignant tumors, accounting for first place in head and neck malignant tumors. Nonkeratinizing carcinoma (NKC) is the major histologic type of NPC. We performed Transcriptional profiling of NKC comparing tumor with adjacent non-tumor tissues using. Goal was to identify and investigate dryregulated lncRNAs and mRNA in NKC at genome-wide scale.

Project description:MicroRNAs are biomarkers of prognosis and survival for many types of cancer. We evaluated whether microRNAs can predict the survival and efficacy of concurrent chemotherapy in nasopharyngeal carcinoma (NPC) patients. We retrospectively analyzed microRNA expression in 312 paraffin-embedded NPC specimens and 18 normal nasopharyngeal tissues using microarray. We found Forty-one microRNAs are differentially expressed between NPC and normal tissues, and a five-microRNA signature can predict survival independent of stage. Genome-wide expressing profiling of nasopharyngeal carcinoma primary tumors versus non-cancerous nasopharyngeal tissues.

Project description:To explore differentially expressed genes between nasopharyngeal carcinoma (NPC) primary tumors and non-cancerous nasopharyngeal tissues, 18 NPC tissue samples versus 18 control samples were utilized to perform genome-wide expressing profiling. Consequently, 2992 genes were found to be differentially expressed in NPC tissues relative to the control (FC>2, P<0.05). Among these 2992 genes, uPA was ranked as the top one in all upregulated genes sorted by ascending order of P-value. Moreover, expression of uPAR was also upregulated in NPC tissues with FC=3.34 and P=7.52M-CM-^W105. 18 nasopharyngeal carcinoma primary tumors and 18 non-cancerous nasopharyngeal tissues were used to perform genome-wide expressing profiling. The median ages of patients were 46 (range, 19-77) for NPC patients and 45 (range, 18-78) for the non-cancerous cohort. Almost one third of patients were female. All samples were collected before any anti-cancer treatment.

Genome-wide analysis of DNA methylation between nasopharyngeal carcinoma tissues and normal nasopharyngeal epithelial tissues

Project description:Nasopharyngeal carcinoma (NPC) is rare malignancy in most parts around the world but common in Southern China and Southeast Asia, Annually, approximately 80,000 new NPC cases and 50,000 deaths are reported worldwide. In NPC, recent advances have shown that many genes that are predominantly or even exclusively silenced by DNA methylation in epithelial cells during pathogenesis of NPC. However, the causal relationship between DNA methylation status and outcome in NPC remains not well understood. To investigate this problem, we used Illumina 450K BeadChips to examined methylation patterns and survival in NPC. and ultimately provided insight into prognostic value of DNA methylation in NPC clinical management. We employed Illumina Human Methylation450K Beadchip to analyze a genome-wide of DNA methylation in a cohort of 48 samples (between 24 nasopharyngeal carcinoma tissues and 24 normal nasopharyngeal epithelial tissues) to identify aberrant methylation genes.

Project description:Nasopharyngeal carcinoma (NPC) is a common cancer in southern China and South East Asia where more than 50,000 new cases are diagnosed each year. We used microarrays to identify down or upregulated genes in NPC compared with non-malignant controls. Experiment Overall Design: Snap frozen nasopharyngeal biopsies from 25 patients with histologically confirmed undifferentiated NPC were included in the microarray analysis. Controls were obtained from 3 patients with no evidence of malignancy.

Transcription profiling of human xenografted nasopharyngeal carcinoma tumor cell lines and an untransformed diploid fibroblast cell line

Project description:The aim of this experiment was to investigate the gene expression profile of two xenografted nasopharyngeal carcinoma tumors - C15 and C17 which are international reference tools for biological investigations of this type of disease. C15 and C17 transcriptome was profiled by comparison with human untransformed diploid fibroblasts, MRC5. These fibroblasts are very sensitive to contact inhibition and were collected after 3 days of confluence, therefore in a quiescent state. We acknowledge that malignant nasopharyngeal carcinoma cells and MRC5 cells do not belong to the same lineage (epithelial cells versus fibroblasts). However this combination was expected to highlight genes involved in proliferation of NPC cells. Competitive hybridisation of cDNA for each pair C15 and MRC5, C17 and MRC5 was performed on Agilent microarrays containing 22 000 60mer oligonucleotides related to 16 000 human transcripts. The analysis was focused mainly on genes overexpressed in both C15 and C17 by comparison with MRC5

Project description:Epstein-Barr virus (EBV) has been etiologically linked to human malignancies including Nasopharyngeal Carcinoma (NPC). Although EBV-encoded miRNAs have been shown to contribute to viral latency, host cell survival and immune evasion, their direct impact on cancer progression in their human host remains unclear. In the current study, based on a miRNA expression profiling analysis of a larger number of clinical specimens using a miRNA array platform containing human, EBV and other species miRNA probes., we found that some EBV-miR-BARTs were highly expressed in NPC. Accordingly, further exploration of the potential roles and regulatory mechanisms of some important EBV-miR-BARTs in NPC progression was carried out. We applied a miRNA expression profiling analysis in 20 poor or undifferentiated NPC (Nasopharyngeal Carcinoma) matched with 20 benign chronic nasopharyngitis (CNP) specimens using a miRNA array platform containing human, EBV and other species miRNA probes. Two-group experiment, NPC vs CNP. Biological repelicates: 20 NPCs, 20 CNPs. One replicate per array.
