Gene expression signatures for murine alveolar type II lung epithelial cells after tumor bearing

ABSTRACT: To determine the influence of primary tumors on pre-metastatic lungs, we have employed whole genome microarray expression profiling as a discovery platform to identify gene signatures of alveolar type II epithelial cells (AT-II) in TLR3 deficient mice (Tlr3-/-) and wide-type (WT) littermates with tumor bearing. We subcutaneously inoculated Tlr3-/- and WT mice with Lewis lung carcinoma (LLC). Two weeks later, lung tissues from Tlr3-/- and WT mice were dissociated and AT-II cells were sorted. AT-II cells from mice without tumor bearing were set as controls.

Whole transcriptome of Lewis lung carcinoma cells of Mus musculus

Project description:Purpose: To study the alteration of whole transcriptome of Lewis lung carcinoma (LLC) cells after the decreasing of malignant properties of tumor by treatment of tumor-bearing mice with RNase A. Methods: Whole transcriptome profile of Lewis lung carcinoma before and after RNase A treatment were generated by deep sequencing using SOLiD 5.5. The sequence reads were mapped by Bioscope 1.3 software, differential expression was evaluated by Cufflinks v.2.0.1 package. Results: Difference in expression was found for 966 genes. Conclusions: Our study represents the first detailed analysis of alteration of transcriptome of Lewis lung carcinoma after the decrease of malignant prtoperties of the tumor (proliferation and invasion) by RNase A. Whole transcriptome profile of Lewis lung carcinoma before and after RNase A treatment were generated by deep sequencing using SOLiD 5.5.

Comparison of mRNA expression in white adipose tissue from LLC tumor-bearing mice and controls

Project description:Cancer-associated cachexia (CAC) is a wasting syndrome caused by malignant tumors. Fat loss plays a significant role in the pathophysiology of cancer-associated cachexia. The mechanism of fat loss in CAC includes enhancement of lipolysis, inhibition of lipogenesis and browning of white adipose tissue. In order to discover potentially functional genes in white adipose tissue of CAC, we used transcriptome sequencing technology to find research-valuable genes in white adipose tissue (WAT) of CAC rodent model. C57/bl6 mice with LLC tumors were used as the in vivo model for CAC, while normal mice subcutaneously injected with phosphate buffer solution were used as the control group. Overall, our data revealed genes that were differentially expressed in the white adipose tissue of LLC tumor-bearing mice, providing a molecular framework for the investigation into CAC fat loss.

Project description:To determine the different gene signatures between primary tumor and tumor-derived exosomes, we have employed RNA-sequencing as a discovery platform to identify gene signatures of tumor-derived exosomes, taking the original tumors as a control. We subcutaneously inoculated C57BL/6 mice with Lewis lung carcinoma (LLC). Three weeks later, tumor tissues were cut and tumor-derived exosomes were isolated as described in the "treatment protocol". Then, both exosomal RNA and tumor RNA were extracted and sequenced. From sequencing, we found that exosomal RNAs showed quite different transcript profiles from tumor RNAs.
