ABSTRACT: We analyzed the transcriptome of dormant and after-ripened imbibed seeds of the Arabidopsis accession Cape verde Islands. We analyzed the transcriptional changes in temporal and spatial detail by sampling at four imbibition time-points (3, 7, 12 and 24 hours after sowing) and two seed compartments (RAD and MCE).

Gene expression in dormant, germinating and thermoinhibited Arabidopsis seeds

Project description:To establish the basis for understanding molecular mechanism of seed germination response to temperature, we analyzed transcriptomes in freshly harvested dormant and dry stored after-ripened seeds. The after-ripened seeds started to show visible germination from 36h after the start of imbibition, and almost all the seeds germinated after 3 days. The freshly harvested seeds stayed dormant by imbibition at 26°C, and germination of the after-ripened seeds was almost completely inhibited at 34°C. Total RNA was prepared from 0 (dry), 6 and 24h imbibed seeds to find regulatory genes of seed dormancy and germination.

Project description:In the dry state, orthodox seeds have little antioxidant capacity, relying on reduced glutathione and tocochromanols for antioxidant defences rather than ascorbate or ascorbate peroxidase (APX). Ascorbate is synthesised de novo within a few hours of imbibition resulting in rapid increases in the ascorbate pool in the developing embryo. The knock-on effects of enhanced oxidation on embryo development before and after imbibition remain poorly characterised. A T-DNA insertional mutant line (SAIL_769_H05) line was used in these studies together with vtc2-1 mutants that were originally identified in an ethyl-methanesulfonate (EMS) mutagenesis screen. Gene expression profiling predicts processing properties in short-term imbibition of wheat

Project description:Objective: Germination of wheat maximizes phytochemical content and antioxidant activity while altering chemical composition, gluten content, and pasting properties. We previously reported marked changes in gene expression in soybean prior to germination before the radicle had grown from a seed. The present study investigated whether imibibition induces similar changes in wheat. Methods: Changes in gene expression profiles of wheat during short-term imbibition (0, 16, and 24 h) were evaluated by DNA microarray analysis. Gene Ontology (GO) analysis was carried out to categorize the function of genes with altered expression. The expression of genes encoding enzymes associated with starch breakdown was evaluated by quantitative real-time PCR, and changes in enzymatic activity were assessed with functional assays. Pasting properties of flour made from wheat seeds imbibed for different times were examined with a Rapid Visco Analyzer. The protein profile was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis, and gluten content was quantified. Results: The GO analysis revealed that genes related to cellulose and cell wall synthesis were upregulated by imbibition for 16 h whereas those associated with polysaccharide catabolism and nucleosome assembly were upregulated in the subsequent 8 h. α-Amylase expression was highest after 24-h imbibition, with a corresponding increase in enzymatic activity. The pasting properties of wheat flour decreased when seeds were imbibed for over 16 h. Gluten was not degraded until 48 h imbibition. Conclusion: Short-term imbibition of wheat can produce a new type of starch with improved physical and functional properties that may be more appealing to consumers.
