IGF2BP3 promotes tumour cells survival in organoid pancreatic adenocarcinoma model

Project description:Transcriptional profiles of two PDAC cell lines (CF_PAC1 and PANC1) are highly divergent. Knocking down IGF2BP3 resulted in significant transcriptional divergence at 24H, 48H and 72H compared to control, most strikingly in the PANC1 cell line. The dominant affect of knockdown was widespread reduction in expression levels of genes functionally enriched for Proliferation, TNFA signalling and Apoptosis. This highlights IGF2BP3 in these cell lines to be acting as a positve regulator of gene programs broadly associated with progression through the cell cycle and identifies many candidate IGF2BP3-linked genes. A high confidence, consistent motif of genes identified as IGF2BP3-regulated in both cell lines included the genes SLC22A18, GET3 and CCNG1.

IGF2BP3 Promotes Stemness and Leukemogenesis in Acute Myeloid Leukemia

Project description:N6-methyladenosine (m6A) is the most abundant internal modification in mRNA, and its modulators have been implicated in various biological processes. Here, we demonstrate that the mRNA m6A reader IGF2BP3 is highly expressed in human acute myeloid leukemia (AML), particularly in AML leukemia stem/initiating cells (LSCs/LICs). IGF2BP3 is required for the development and maintenance of AML and the self-renewal of LSCs/LICs but dispensable for normal hematopoiesis. IGF2BP3 exhibits promising anti-AML efficacy both in vitro and in vivo, providing new therapeutic strategies for targeting AML.

The RNA binding protein IGF2BP3 is required MLL-AF4 mediated leukemogenesis

Project description:In our study, we found that MLL-AF4 transcriptionally induces IGF2BP3 and is required for MLL-Af4 mediated leukemogenesis. Deletion of murine Igf2bp3 significantly increased the survival of mice with MLL-Af4 driven leukemia and greatly attenuated disease. Furthermore, Igf2bp3 was necessary for the development of and the self-renewal capacity of MLL-Af4 leukemic initiating cells. eCLIP and transcriptome analysis of MLL-Af4 transformed stem and progenitor cells and primary cells from MLL-Af4 leukemic mice revealed an IGF2BP3-regulated post-transcriptional operon governing tumor cell survival and proliferation. Critical mRNA targets include the Hoxa locus and numerous genes within the Ras signaling pathway. Together, our findings show that IGF2BP3 is an essential positive regulator of MLL-AF4 mediated leukemogenesis and is a potential therapeutic target in this disease.

IGF2BP3 promotes mRNA degradation through internal m7G modification [PARCLIP]

Project description:Recent studies have suggested that mRNA internal m7G and its writer protein METTL1 are closely related to cell metabolism and cancer regulation. Here, we identify that IGF2BP family proteins IGF2BP1-3 can preferentially bind internal mRNA m7G. Such interactions, especially IGF2BP3 with m7G, could promote the degradation of m7G target transcripts in cancer cells. IGF2BP3 is more responsive to changes of the m7G modification, while IGF2BP1 prefers m6A to stabilize the bound transcripts. We also demonstrate that p53 transcript, TP53, is m7G-modified at its 3’UTR in cancer cells. In glioblastoma, the methylation level and the half lifetime of the modified transcript could be modulated by tuning IGF2BP3, or by site-specific targeting of m7G through a dCas13b-guided system, resulting in modulation of cancer progression and chemosensitivity.
