Project description:Biotic and abiotic stresses limit agricultural yields, and plants are often simultaneously exposed to multiple stresses. Combinations of stresses such as heat and drought or cold and high light intensity, have profound effects on crop performance and yeilds To analyze such responses, we initially compared transcriptome changes in ten Arabidopsis thaliana ecotypes using cold, heat, high light, salt and flagellin treatments as single stress factors or their double combinations. Arabidopsis thaliana plants of ecotypes (Col, Ler, C24, Cvi, Kas1, An1, Sha, Kyo2, Eri and Kond) were subjected to the following stress treatments: Salt, Cold, Heat, High Light (HL), Salt+Heat, Salt+HL, Cold+HL, Heat+HL, as well as FLG (Flagellin, flg22 peptide), Cold+FLG, Heat+FLG

Analysis of reversibility of transcriptome responses to abiotic stress conditions in Arabidopsis thaliana

Project description:Plants acclimate to environmental fluctuations by transitory reconfigurations the homeostatic network. Primary studies suggested that transcriptome responses to deal with fluctuations in light intensity and temperature tend to reversibility after stress removal in the model plant Arabidopsis thaliana. To gain more insight into this pattern in the context of acclimation, RNA-Seq analysis were conducted in Arabidopsis thaliana after different abiotic stress treatments consisting in high light (HL), high humidity, drought, heat, cold and combinations among factors or after recovery periods. Our transcriptome study is in line of a general pattern wherby transcriptome changes in response to adverse environments are prone to return to the basal state during the de-acclimation phase.

Seedling transcriptomes of Arabidopsis thaliana HULK mutants

Project description:We produced RNA-seq reads from messenger RNA isolated from aerial seedling tissue for Arabidopsis thaliana mutants in the HULK gene family. The read data were generated with biological replication (two replicates). The resulting RNA-seq data provide a resource to assess the function of HULK genes in the control of downstream gene expression in A. thaliana.

Nontargeted and targeted metabolomics measurements of abiotic stress responses in three-week-old Arabidopsis thaliana plants' rosette leaf tissue

Project description:Nontargeted and targeted metabolomics measurements of abiotic stress responses in three-week-old Arabidopsis thaliana plants' rosette leaf tissue for Col-0 wild type plants and double/triple knockout mutants of aquaporins (pip2;1 pip2;2 and pip2;1 pip2;2 pip2;4) treated with drought, heat at different air humidities, or combined drought-heat stress at different air humidities. This experiment contains FT-ICR-MS measurements for 103 Arabidopsis thaliana rosette leaf samples covering three genotypes under six different environmental conditions. The three genotypes comprise the Col-0 wildtype and two loss-of-function mutants of aquaporins, a pip2;1 pip2;2 double mutant and a pip2;1 pip2;2 pip2;4 triple mutant (respective AGI locus identifiers: AT3G53420, AT2G37170, AT5G60660). The six conditions include control condition (well-watered, 22 °C, 70% relative air humidity), drought stress (one week without watering), heat stress without changing the absolute humidity of the ambient air (6 hours at 33 °C, 37% relative air humidity), heat stress with supplemented air humidity to maintain a constant vapor pressure deficit before and during the heat episode (6 hours at 33 °C, 84% relative air humidity), and the combinations of drought pretreatment with each of the two heat stress variants (one week of drought followed by 6 hours of heat stress). Samples from all conditions were harvested at the same time (within 15 min starting at 5 pm). For validation, GC-TOF-MS measurements were done for two genotypes (wildtype, double mutant) and two conditions (drought, control) on partially overlapping samples.

Transcriptomic profiling of Arabidopsis thaliana in response to combined stress treatment under altered GSH conditions.

Project description:The Arabidopsis seeds of Col-0 and pad2.1 were grown for three weeks and were further placed in filter paper for 5 min at 4°C and then transferred to 1/2MS media with 30% PEG at 4°C for an additional 6 hours; the leaves were collected for RNA isolation. Trancriptomic profiling of the combined stress treated Col-0 and pad2.1 indicated downregulation of various abiotic and oxidative stress responsive genes in pad2.1 in response to combined stress treatment.
