Analysis of gene expression in grape berries due to diurnal effects (DIU_06).

ABSTRACT: This is a 24 hour time course experiment to investigate changes in gene expression in deseeded berries sampled every three hours within a 24 hour period at 2 time points during development i.e. T4 (preveraison) & T15 (postveraison). Changes in gene expression within the 24 hour period were analyzed to identify expression profiles that coincided with diurnal changes. The gene expression profiles were then to be compared between the two developmental time points T4 v T15). Keywords: Diurnal time course

Diurnal oscillations in murine brown adipose tissue analyzed by ChIP-seq

Project description:Brown adipose tissue (BAT) burns fatty acids to produce heat, and shows diurnal oscillation with peak activity in glucose and triglyceride-derived fatty acid uptake at wakening. To gain further insight in the diurnal regulation of metabolic BAT activity, we performed chromatin immunoprecipitation sequencing (ChIP-seq) on pooled interscapular (i)BAT samples collected from chow-fed male C57BL/6J mice (10 weeks old) exposed to mild cold (22°C) at 3-hour intervals throughout a 24-hour period (n=8 mice per time point). All mice were entrained to a 12h:12h light:dark cycle, and therefore time is denoted as Zeitgeber Time (ZT) in which ZT0 indicates the onset of the light (active) phase.

Tissues from wild type mice were harvested every 3 hrs over a 36 hour time period, for a total of 13 time points. Striatal, cortical, hypothalamic and liver samples were harvested, flash frozen, and transferred to Expression Analysis for RNA extraction and miRNA-seq sequencing. Samples passing QC were analyzed to produce high quality, 50-base paired-end RNA-seq results.
