Natural variation in brain gene expression profiles of aggressive and nonaggressive individual sticklebacks

ABSTRACT: We examine whether there are differences in gene expression between aggressive versus nonaggressive behavioral types of individuals within a natural population of male threespined sticklebacks (Gasterosteus aculeatus). We compared gene expression profiles of aggressive male sticklebacks to nonaggressive males in four regions of the brain (brainstem, cerebellum, diencephalon and telencephalon). We identified new candidate genes related to aggressiveness, including SLC38a, a solute carrier which has been linked to GABAergic neuron reuptake of glutamine.

Project description:We investigated the transcriptomic response to territorial intrusion in four regions of the brain (telencephalon, diencephalon, cerebellum and brain stem) of a teleost fish, the threespined stickleback (Gasterosteus aculeatus). We tested the hypothesis that gene regulatory networks modulate the transcriptional response to territorial intrusion in the different brain regions and thereby aggressive behavior. Indeed, transcription regulatory network analysis identified cis-regulatory transcription factor motifs enriched in upregulated genes in one brain region and downregulated in the others. We further validated our findings by showing that many of the differentially expressed genes in the diencephalon were positively correlated with levels of aggression in individual males. 2 conditions: control fish and fish that experienced intrusion into their territory, Biological replicates 5.Four brain regions. Sample 4BS was excluded from the analysis during data preprocessing because it was an outlier.

Personal and transgenerational cues are nonadditive at the phenotypic and molecular level

Project description:Organisms can gain information about their environment from their ancestors, their parents, or their own personal experience. “Cue integration” models often start with the simplifying assumption that information from different sources is additive. Here, we test key assumptions and predictions of cue integration theory at both the phenotypic and molecular level in threespined sticklebacks (Gasterosteus aculeatus). We show that regardless of whether cues about predation risk were provided by their father or acquired through personal experience, sticklebacks produced the same set of predator-adapted phenotypes. Moreover, there were nonadditive effects of personal and paternal experience: animals that received cues from both sources resembled animals that received cues from a single source. A similar pattern was detected at the molecular level: there was a core set of genes that were differentially expressed in the brains of offspring regardless of whether risk was experienced by their father, themselves or both. These results provide strong support for cue integration theory because they show that cues provided by parents and personal experience are comparable at both the phenotypic and molecular level, and draw attention to the importance of nonadditive responses to multiple cues.

Microarray analysis of di-n-butyl phthalate and 17α ethinyl-oestradiol responses in three-spined stickleback testes

Project description:The data comes from a study, where three-spined sticklebacks (Gasterosteus aculeatus) were exposed to di-n-butyl phthalate (DBP) and 17α ethinyl-oestradiol (EE2) at nominal concentrations 35 μg/L and 40 ng/L, respectively, for four days. The aim of the study was to obtain insight into the acute transcriptional responses putatively associated with endocrine disruption. RNA samples from the testes of eight individuals fish per treatment (including solvent controls, exposed only to DMSO) were used in the microarray analysis, covering the expression of approximately 21000 genes.

Project description:The aim of this study was to describe the excretory–secretory proteins from the helminth Schistocephalus solidus and its intermediate host, the three-spined stickleback Gasterosteus aculeatus L., which are likely to be involved in interactions between them. Combined samples of washes from the G. aculeatus sticklebacks cavity infected with the S. solidus, and washes from the parasite surface were used as experimental samples (samples 5 to 8), while washes from the uninfected fish body cavity were used as control (samples 1 to 4). The obtained samples were analyzed using mass-spectrometry nLC–MS/MS.

Proteome of Schistocephalus solidus (Cestoda) after short- and long-term heating

Project description:The protein composition of the cestode Schistocephalus solidus was measured in an experiment simulating the transition of the parasite from a cold-blooded to a warm-blooded host. Infective S. solidus plerocercoids obtained from the three-spined stickleback Gasterosteus aculeatus were heated at 40 °C for 1 hour or cultured in vitro at 40 °C and 22 °C for 48 hours. Plerocercoids taken out from the body cavity of sticklebacks immediately after their sacrifice were used as a reference group.
