Project description:Human mesenchymal stem cells (hMSCs), which are multipotent cells to differentiate into several cell types, are expected to be a useful tool for cellular therapy. In some clinical settings, hMSCs have immuno-suppressive effects for GVHD (Graft-versus-host disease) and are expanded in vitro before application. To find biomarkers that indicate the culture stage of hMSCs, we performed microarray analysis for hMSCs derived from bone marrow, using Affymetrix GeneChip Human Genome U133 Plus 2.0 (54,613 probe sets). Experiment Overall Design: We profiled the gene expression in 6 different batches of hMSCs. The cells were cultured and subjected to total RNA extraction. We performed microarray analysis using 6 batches provided by 6 different individuals in early stage (passage #4 or 5), middle stage (#7 or 9) or senescing stage (#22, 24, or 28).

Project description:In this study, we have addressed how cellular senescence influences the immunomodulatory potential of human mesenchymal stem cells (hMSCs). We induced cell senescence in a panel of bone marrow-derived hMSC samples by means of gamma-irradiation, and performed both gene expression and miRNA microarray analyses on the untreated and senescent samples. We also compared the gene expression profile of untreated and senescent hMSCs with those obtained from several hMSCs samples used in an ongoing allogeneic clinical study of Graft Versus Host Disease (GVHD), of which their therapeutic efficacy is known. We have identified several genes (PLEC, C8orf48, TRPC4, and ZNF14) differentially expressed in senescent hMSCs that are similarly regulated in hMSC samples that did not show a therapeutic effect in the GVHD study. These genes might be useful as markers to evaluate the therapeutic potential of hMSCs used in future clinical studies.

The samples used in the clinical study were classified in two groups, depending on whether they elicited a therapeutic response (G1) or not (G2). A total of four independent samples (biological replicates) were used for WT and SEN conditions. For the samples used in the clinical study, a total of five samples were used for the G1 group, and three samples for the G2 group.

However, detailed pathophysiology of acute GVHD developed after UCBT has not yet been clarified. Here, we examined the roles of inflammatory cells in the pathogenesis of acute GVHD following UCBT, by expression profiling of a total of 615 genes in each of 4 subsets (CD4+, CD8+, CD14+, and CD56+) of peripheral blood mononuclear cells (PBMCs), which were taken from 5 patients with hematologic malignancy, in whom acute GVHD had developed after UCBT. By comparing expression profiles measured during acute GVHD with those measured upon discharge (recovery phase), for each subset of PBMCs, we identified immuno-regulatory genes which were assumed to be linked to the pathogenesis of acute GVHD. Project description:Allogeneic hematopoietic stem cell transplantation (HSCT) is the treatment of choice for high-risk hematological malignancies, yet a major complication associated with this therapy is acute graft-versus-host disease (GVHD). Despite a well-defined pathophysiological mechanism, there are no definitive markers for predicting acute GVHD development or progression to advanced stages. In the current study, we enrolled four acute GVHD and four acute GVHD-free recipients of allogeneic HSCT and collected peripheral blood just prior to onset of clinical acute GVHD for analysis on Affymetrix GeneChip Human Genome U133 Plus 2.0 microarrays. We noted significant differences in expression of 1,658 genes between control and acute GVHD patients, based on an analysis of covariance (ANCOVA) by type of transplant, a pooled error estimate, and a false discovery rate (FDR) of 10%. In conclusion, we offer the first report of a preliminary molecular signature of acute GVHD in allogeneic HSCT patients.

Project description:Human mesenchymal stem cells (hMSCs) promote endogenous tissue regeneration and have become a promising candidate for cell therapy. However, in vitro culture expansion of hMSCs induces a rapid decline of stem cell properties through replicative senescence. Here we characterize metabolic profiles of hMSCs during expansion. We show that alterations of cellular nicotinamide adenine dinucleotide (NAD+ /NADH) redox balance and activity of the Sirtuin (Sirt) family enzymes regulate cellular senescence of hMSCs. Treatment with NAD+ precursor nicotinamide increases the intracellular NAD+ level and re-balances the NAD+ /NADH ratio, with enhanced Sirt-1 activity in hMSCs at high passage, partially restores mitochondrial fitness and rejuvenates senescent hMSCs. By contrast, human fibroblasts exhibit limited senescence as their cellular NAD+ /NADH balance is comparatively stable during expansion. These results indicate a potential metabolic and redox connection to replicative senescence in adult stem cells and identify NAD+ as a metabolic regulator that distinguishes stem cells from mature cells. This study also suggests potential strategies to maintain cellular homeostasis of hMSCs in clinical applications.
