Analysis of the expression profile of skin macrophages FACS-sorted from mice overexpressing activin and/or oncogenes of human papilloma virus 8 in keratinocytes.

ABSTRACT: We have shown that activin promoted skin tumorigenesis in mice induced by the human papilloma virus 8 oncogenes. Activin attracted blood monocytes to the skin as revealed by depletion of CCR2-positive monocytes. To determine if activin also altered the gene expression profile of these cells, we performed RNA-Sequencing of macrophages FACS-sorted from the pre-cancerous ear skin. We have found that activin induces a pro-migratiory, pro-angiogenic and pro-tumorigenic genes in skin macrophages in vivo. This largely contributes to the pro-tumorigenic function of activin, since macrophage depletion delayed spontaneous tumorigenesis in HPV8-transgenic mice by reducing keratinocyte proliferation and angiogenesis.

Project description:Overexpression of activin by keratinocytes promotes HPV8-induced skin tumorigenesis in mice. Activin-promoted tumorigenesis is mediated via the stroma, specifically by cancer-associated fibroblasts (CAFs). To determine if activin alters the gene expression profile of skin fibroblasts, we performed RNA-sequencing of fibroblasts FACS-sorted from pre-cancerous ear skin of activin overexpressing mice. We found that activin induces a CAF-like gene expression profile of fibroblasts, leading to the upregulation of genes involved in proliferation, migration and cytoskeletal remodeling.

Re-education of tumor-associated macrophages by CXCR2 blockade drives senescence and tumor inhibition in advanced prostate cancer

Project description:Tumor-associated macrophages (TAMs) represent a major component of the tumor microenvironment that supports tumorigenesis. Here we found that prostate tumors are strongly infiltrated by TAMs expressing the C-X-C chemokine receptor type 2 (CXCR2). Pharmacological blockade of the CXCR2 receptor by a selective antagonist promoted the re-education of TAMs towards a pro-inflammatory phenotype. Strikingly, CXCR2 knockout monocytes infused in Ptenpc-/-; Trp53pc-/- mice preferentially differentiatied in TNFα-releasing pro-inflammatory macrophages leading to senescence induction and tumor inhibition. Moreover, tumor cells harboring PTEN deletion were more sensitive to TNFα-induced senescence, when compared to PTEN WT tumors, due to increased levels of TNFR1. Our results identify TAMs as a target in prostate cancer therapy and describe a novel therapeutic strategy based on CXCR2 blockade to harness the anti-tumorigenic potential of macrophages against this disease.

Epidermal growth factor shedding directs macrophage education in cancer

Project description:Macrophages in the tumor microenvironment have a significant impact on tumor progression. Depending on the signaling environment in the tumor, macrophages can either support or constrain tumor growth and metastasis. It is therefore of therapeutic interest to identify the tumor-derived factors that control macrophage education. With this aim, we correlated the expression of ADAM proteases, which are key mediators of cell-cell signaling, to the expression of pro-tumorigenic macrophage markers in human cancer cohorts. We identified ADAM17, a sheddase upregulated in many cancer types, as a protein of interest. Depletion of ADAM17 in cancer cell lines reduced the expression of several pro-tumorigenic markers in neighboring macrophages in vitro as well as in mouse tumor models. Accordingly, ADAM17-/- educated macrophages demonstrated a reduced ability to induce cancer cell invasion. Using quantitative mass spectrometry-based proteomics, we identified HB-EGF, shed by ADAM17 in the cancer cells, as the implicated molecular mediator of macrophage education. Additionally, RNA-seq and ELISA experiments revealed that ADAM17-dependent HB-EGF release induces the expression and secretion of CXCL chemokines in macrophages, which in turn stimulates cancer cell invasion. In conclusion, we provide evidence that ADAM17 mediates a paracrine HB-EGF-chemokine feedback loop, whereby cancer cells hijack macrophages to promote tumor progression.

The impact of HPV8 E6 on Lrig1+ hair follicle stem cell populations

Project description:Despite epidermal turnover, the skin is host to a complex array of microbes including viruses, such as the human papillomavirus (HPV), which must infect and manipulate skin keratinocyte stem cells (KSC) to survive. This crosstalk between the virome and KSC populations remains largely unknown. Here, we investigated the effect of HPV8 on KSCs using various mouse models. We observed that the HPV8 early region gene E6 specifically caused Lrig1+hair follicle junctional zone KSC proliferation and expansion, which would facilitate viral transmission. Within Lrig1+KSCs specifically, HPV8 E6 bound intracellular p300 to phosphorylate the STAT3 transcriptional regulatory node. This induces a transcriptional switch from TAp63 to ΔNp63 expression, resulting in expansion of KSC into the overlying epidermis. HPV8 was associated with 70% of human actinic keratoses (AK). Together these results redefine human AK as an expansion of KSC, which lack protecting melanosomes and are thus susceptible to sun-light-induced malignant transformation.

Project description:Overexpression of activin A by keratinocytes accelerates excisional wound healing in mice. Activin-promoted wound healing is mediated via the stroma, specifically by the wound immune cells and fibroblasts. To determine if activin A alters the gene expression profile of wound fibroblasts, we performed RNA-sequencing of fibroblasts FACS-sorted from excisional skin wounds of activin overexpressing mice. We found that activin induces a pro-fibrotic gene expression profile of these fibroblasts, leading to the upregulation of genes involved in collagen biosynthesis and remodeling.

In vitro differentiation of M1, M2, and TAM-like macrophages from healthy donor PBMC

Project description:To understand the immunological role of different macrophage populations in the tumor, we generated transcriptional profiles of macrophages differentiated from PBMC-derived monocytes in vitro under three conditions to compare with: M1-like macrophages were developed in vitro through polarization of human monocytes by IFNg and TLR4 stimulation. This population is considered to have anti-tumor activities; releasing proinflammatory cytokines, performing phagocytosis, and possessing antigen cross presentation abilities. Second, M2-like macrophages were developed in vitro through intensive IL4 stimulation of monocytes. M2 macrophages are known to promote many pro-tumorigenic processes such as angiogenesis, immune suppression, hypoxia induction, tumor cell proliferation, metastasis. Lastly, we deployed a third population that represents a better in vitro model for tumor associated macrophages (TAM), developed under conditions better resembling the TME.Monoctyes derived from two individual patient donors were cultured separately under four (4) different conditions in vitro, for a total of eight (8) samples to be processed for bulk RNA sequencing.
