Project description:Cardiotoxin was injected into left soleus muscle of adult rats. Right muscle was kept intact. Ambulation recovery was allowed for 8 weeks. After 8 weeks of regeneration period, both left and right soleus muscles were overloaded by the transection of synergists' tendons for 2 weeks. We tested 6 muscles for each group. RNA samples were mixed with same amount from each muscle toward analysis by microarray.

Project description:Skeletal muscle possesses the ability to adapt its size in response to milieus, which is called plasticity. Resistance training induces the increment of muscle mass called muscle hypertrophy. Muscle stem cells (MuSC; also known as muscle stem cells) function to supply new nuclei for myofiber during the overload in muscle. We found that Yap1 and Taz in mesenchymal progenitors (also called FAPs) are critical for MuSC proliferation in overloaded muscles. We hypothsize that Yap1/Taz-dependent mesenchymal progenitors derived factor induces MuSC proliferation. In order to identify such factors, RNA-seq of overloaded FAPs were performed.

Project description:We report the mRNA sequencing of right and left atria from an adult inducible, cardiomyocyte-specific Notch gain-of-function mouse model (iNICD). Using the tetracycline-on system, we activated Notch by feeding doxycycline chow for 3 weeks to mice that were at least 8 weeks old. We asked what transcriptional changes occur in right versus left atrial cardiomyocytes in response to the same stimulus (Notch signaling). mRNA sequencing on separated right and left atria revealed that there are more differentially dysregulated transcripts (1,011) than similarly regulated transcripts (447) in the right and left atria, which is a simiar paradigm as what occurs in human atrial cardiomyocytes of patients with atrial fibrillation.

Project description:One hundred ninety wildtype male C57BL/6J mice age 7-10 weeks were purchased from Jackson Laboratory and entrained to a 12:12 light:dark cycle for 2 weeks. Mice were placed in light-tight boxes on a 12:12 LD cycle for 4 weeks, then released into constant darkness. Starting 30 hours after entry into DD (CT18), the left leg muscle from 5 wildtype mice, and the right leg muscle from the same wildtype mice were collected every 4 hours for 48 hours, for a total of 12 timepoints. Total RNA from each side (left or right) were separately pooled to make a single sample for amplification and hybridization. Therefore, duplicate arrays (from left and right side) were performed at each time point. At timepoints 34 through 58 hours in DD, tissues from age-matched male C57BL/6J Clock/Clock homozygous mutant mice that had been treated with the same light entrainment protocol as the wildtype were collected. Tissues were collected from 5 Clock/Clock mutants at each timepoint except for 34 and 46 hours after the onset of DD, when tissues from 10 Clock/Clock mice were collected. As for the wildtype mice, right and left sides were collected, prepared, amplified, and hybridized in separate pools. Mice were sacrificed by cervical dislocation, and the optic nerves were severed in complete darkness; brain dissection was performed using illumination from an infrared viewer (FJW Industries, Palatine, IL). Project description:Left-right asymmetry is a fundamental organizing feature of the human brain, and neuro-psychiatric disorders such as schizophrenia sometimes involve alterations of brain asymmetry. As early as 8 weeks post conception, the majority of human foetuses move their right arms more than their left arms, but because nerve fibre tracts are still descending from the forebrain at this stage, spinal-muscular asymmetries are likely to play an important developmental role. Here we have used gene expression profiling in 18 human embryos to show that the left side of the human spinal cord, between four and eight weeks post conception, matures slightly faster than the right side, even though both sides transition from transcriptional profiles associated with cell division and proliferation at earlier stages, to later neuronal differentiation and function. The hindbrain showed a left-right mirrored pattern compared to the spinal cord.

Summary: Brain asymmetry in humans, both structurally and functionally, is observed very early during development, and is important for healthy development. Using RNA sequencing, we investigated gene expression profiles of left and right sides of various brain structures in human foetuses aged 5 to 13 weeks post conception. All brain structures showed significant differences between the expression profiles of left and right sides. The differences partly correlated with gene expression changes with age, suggesting left/right differences in maturation rate. In particular on the faster side (right for cerebral cortex, left for all other investigated tissues), different tissues were lateralised for tissue-specific processes. In the youngest forebrain (5.5 weeks), KCTD12, SNAI1 and GATA1 were significantly right-lateralised. SNAI1 is involved in visceral asymmetry in vertebrates, and KCTD12 is important in lateralisation of fish brains. In the youngest midbrain, SOX1 was significantly left-lateralised.
