Sex and Insulin Resistance Define a Cell Autonomous Supernetwork of Protein Phosphorylation

Project description:Insulin resistance is an important factor in the pathophysiology of many metabolic disorders. The aim of this study was to uncover the cell autonomous determinants of insulin resistance using induced pluripotent stem cell (iPSC)-derived myoblasts (iMyos). Here, we found that iMyos from insulin resistant non-diabetic individuals show impaired insulin signaling, defective insulin-stimulated glucose uptake and glycogen synthase activity compared to insulin sensitive individuals. Global phosphoproteomic analysis uncovered a large network of altered protein phosphorylations in insulin resistance, mostly outside the canonical insulin-signaling cascade. More surprisingly, we observed striking differences in the phosphoproteomic signature from male versus female subjects, including changes in DNA and RNA processing, GTPase signaling, and SUMOylation/ubiquitination. These findings unravel cell autonomous mechanisms underlying insulin resistance and demonstrate a previously unrecognized supernetwork of cell signaling differences in males and females that must be considered in understanding the molecular basis of sex-based differences in normal physiology and disease.

Impact of Preoperative Oral Branched-chain Amino Acids on Reducing Postoperative Insulin Resistance.

Project description:Postoperative insulin resistance refers to the phenomenon that the body’s glucose uptake stimulated by insulin is reduced due to stress effects such as trauma or the inhibitory effect of insulin on liver glucose output is weakened after surgery. There is a clear link between postoperative insulin resistance and poor perioperative prognosis. Therefore, exploring interventions to reduce postoperative stress insulin resistance, stabilize postoperative blood glucose, and reduce postoperative complications are clinical problems that need to be solved urgently. In recent years, research on branched-chain amino acids and metabolic diseases has become a hot spot. Studies have found that in the rat model, preoperatively given a high branched-chain amino acid diet can inhibit postoperative insulin resistance and stabilize blood glucose levels. This research plan is to try to add branched-chain amino acids before surgery to observe the occurrence of postoperative insulin resistance in patients.

Human Insulin Resistance and Thiazolidinedione-Mediated Insulin Sensitization

Project description:Cellular and tissue defects associated with insulin resistance are coincident with transcriptional abnormalities and are improved after insulin sensitization with thiazolidinedione (TZD) PPAR? ligands. We transcriptionally profiled 364 biopsies gathered from 72 human subjects harvested before and after hyperinsulinemic-euglycemic clamp, at baseline and after three-month TZD treatment. Subjects range from insulin-sensitive to insulin-resistant. Insulin resistant subjects responded to TZD treatment with varied improvements in insulin sensitivity, thus they were ranked by their degree of TZD response to define responder and non-responder subgroups. Human adipose insulin resistance

Project description:Adipocytes play a major role in whole body fuel homeostasis. Integrated proteomic analysis of adipose from insulin resistant versus healthy humans or mice or in 3T3-L1 adipocytes revealed defects in the mevalonate/coenzyme Q (CoQ) biosynthesis pathway as a unifying feature of adipocyte insulin resistance. CoQ was decreased selectively in mitochondria in all insulin resistant conditions and this was associated with a selective increase in mitochondrial oxidative stress. CoQ supplementation lowered mitochondrial oxidative stress and reversed insulin resistance in vitro and in vivo. Decreasing mitochondrial CoQ by genetic or pharmacological means caused insulin resistance by increasing mitochondrial oxidants from complex II. Our data suggest that loss of mitochondrial CoQ is a proximal driver of mitochondrial oxidative stress and insulin resistance. These findings may explain why statin use is associated with insulin resistance, and suggest that mitochondrial CoQ may be an effective therapeutic target for treating insulin resistance in humans.

Effect of exercise on insulin resistance and adiponectin in colorectal cancer survivors

Project description:Background and study aims Cancer has become a major public health problem in South Korea. The lifestyle factors are important determinants of colorectal cancer risk. Previous studies have shown a strong association between the level of physical activity and the outcomes of colorectal cancer. However, the effects of exercise on insulin resistance and adipocytokines in colorectal cancer patients still need to be investigated. The aim of this study are to assess the effects of a 12-week home-based exercise intervention on insulin resistance and adiponectin (the hormone produced by fat cells which influences the bodys response to insulin) in colorectal cancer survivors. Who can participate? Patients with stage II-III colorectal cancer, who have completed all standard cancer treatment at least 4 weeks earlier but no more than 2 years. What does the study involve? Patients are randomly allocated to either the exercise group or the control group. Exercise group patients will be encouraged to walk more than 10,000 steps per day. They will get exercise DVDs, will record their daily activities and will visit the clinic five times during the 12 weeks. Control group patients will be instructed to continue with their usual daily activities.
