Circadian cycle-The impact of some genes mutations implicated in the amidon metabolism

ABSTRACT: af30_starch_circadian_rythm - circadian cycle - The scientific aim is to study the impact of some genes mutations implicated in the amidon metabolism on the expression profils of other genes and in particular those of the carbonated metabolism. Those analyses will be done from RNAs (total or messenger) extracted from leaves at various time of the day and the night - The first experiments will deal with the wild reference plant (WS) . The RNAm of leaves would be harvested according to day and night hours. Some harvesting will be done every 3 hours during a 24H period (so 10 different points ). Large scale analysis of leaf gene expression upon treatment of apple grafted scions with acibenzolar-S-methyl or water at 20 or 35°C

Project description:Investigation of whole genome gene expression level changes in the youngest expanded leaves of GDDH13 grafted scions either sprayed with reverse osmosis water or acibenzolar-S-methyl (ASM) either exposed to control temperature (21°C/day ; 18°C/night) or three successive heat days (35°C for 4 h during the day ; 18°C/night). The morning of the third heat day, treatment was performed by spraying to runoff (with a pressurized hand sprayer) with the commercial product Bion 50 WG (Syngenta, Basel, Switzerland; 50% of ASM) prepared in reverse osmosis water at a final concentration of 0.4 g/L. All plants were then returned to control thermal conditions (21°C/day ; 18°C/night) for 2 days. The youngest developped leaf of each plant was sampled 72 hours after the treatment.

Photooxidative stress and light acclimation in the PSII antenna-deficient ch1 mutant

Project description:cea10-02_light - photooxidative stress - To characterize the metabolic pathways implicated in oxidative stress responses and in acclimation mechanisms in the ch1 mutant - The extracts are carried out starting from sheets of Arabidopis thaliana having pushed on compost in controlled conditions (light: 250 µmol.m-2.s-1, temperature: 22°C day 18°C night, moisture: 55%, photoperiod: 8:00 jour/16h night) during 4 and 8 weeks for respectively the Col0 genotype and the mutant ch1. The acclimatization of the plants is done during 48 H under an average luminous intensity (450 µmol.m-2.s-1, photoperiod 8:00), whereas the stress requires a strong luminous intensity (900 µmol.m-2.s-1, photoperiod 8:00) and low temperature (10°C day, 14°C night) also during 48 hours.

Agron-omics Long Day Optimal Watering (LOW) experiment

Project description:Plants adapt to the prevailing photoperiod by optimally adjusting growth and flowering to the availability of energy. When Arabidopsis thaliana plants are grown in long days individual leaf growth is favoured, whereas whole plant leaf area is decreased because of the rapid shift to floral stages and, consequently, the low number of total leaves. To understand the molecular profiles of adaptation to long-day conditions we profiled Arabidopsis leaf number six of plants grown in 16 hours of light at four developmental stages both at the end of the day and the end of the night and compared the profiles to those acquired in short day conditions.

After sterilization, the seeds were placed on Murashige and Skoog medium supplemented with 2% (w/v) sucrose for 10 days and then the seedling were transferred to soil under 16 hours light (22°C) / 8 hours dark (18°C) period in growth chamber at a light intensity of 120?150 µmol m-2 s-1. 20-day-old Arabidopsis leaves without bolting were immediately frozen in liquid nitrogen for RNA and protein and metabolites extraction. Leaves were harvested at three different time points: t = 0 hr (end of night), t = 1 hr (one hour after light turn on) and t = 8 hr (eight hours after light turn on), respectively.

FPB - 2021 - Azri et al - Physiological and proteomic analyses of Tunisian local grapevine cultivar Razegui in response to drought stress.

Project description:Cuttings from Razegui cultivar were cultivated in sand for 2 months according to the method we have described previously (Azri et al. 2020). Then grapevine plantlets were further grown in pots containing sand/peat mixture (2/1, v/v), in a controlled greenhouse (16 h light/8 h dark at PAR 300 μmol m-2 s-1 photoperiod, 24 °C day/20 °C night temperature and 65-75% relative humidity). They were watered daily with tap water and supplemented once a week with 100 mL of Gibeaut’s nutrient solution (Gibeaut et al. 1997). Drought stress was applied according to our previously described procedure (Azri et al. 2020): plants with 10-15 leaves were either irrigated daily with water (control) or non-irrigated at all (water deficit). After 4 time points (day 0, 4, 8 and 16) of water deficit, the 4th fully-developed leaf was harvested and used for physiological analyses. Three independent biological experiments were performed consisting each in 2 treatments (control and water deficit) and 3 plants per time point and treatment. Proteomic analysis was performed on the 5th fully developed leaves at the time point 16 days of drought treatment. Irrigated and non-irrigated leaves were harvested immediately in liquid nitrogen and stored at - 80 °C until analysis. Three biological replicates were carried out for each treatment.

The effect of simulated night shift work on the circadian regulation of the human transcriptome

Project description:Eight healthy human subjects were enrolled in a 6-day simulated shift work protocol. Blood samples were collected during the two 24-hour measurement periods. Blood samples were collected every 4 hours during both measurement periods. Subjects entered the lab on Day 1. At the start of Day 2, the first 24-hour measurement period was started. Subjects slept according to their habitual sleep/wake schedule, followed by a 16-hour constant posture procedure. On days 3-6, the sleep period was delayed by 10 hours. Following the third night on this schedule, subjects underwent another 24-hour measurement period. During both measurement periods, 7 blood samples were collected and PBMCs were isolated. mRNA was extracted, labelled, and hybridized to microarrays.
