SENP2 suppresses browning of white adipose tissues by de-conjugating SUMO from C/EBPβ

Project description:The adipose tissue is a key site regulating energy metabolism. One of the contributing factors behind this is browning of white adipose tissue (WAT), however, knowledge of the intracellular determinants of browning process remains incomplete. By generating adipocyte-specific Senp2 knockout (Senp2-aKO) mice, here we showed that SENP2 negatively regulates browning by de-conjugating SUMO from C/EBPβ. Senp2-aKO mice were resistant to diet-induced obesity and insulin resistance due to increased energy expenditure and heat production. Senp2 knockout promoted beige adipocyte accumulation in inguinal WAT by upregulation of thermogenic gene expression. In addition, SENP2 knockdown promoted thermogenic adipocyte differentiation of precursor cells isolated from inguinal and epididymal WATs. Mechanistically, sumoylated C/EBPβ, a target of SENP2, suppressed expression of HOXC10, a browning inhibitor, by recruiting a transcriptional repressor DAXX. These findings indicate that a SENP2-C/EBPβ-HOXC10 axis operates for the control of beige adipogenesis in inguinal WAT.

Comparison of white adipose tissue and bone marrow adipose tissue from New Zealand White rabbits

Project description:Microarray analysis of white adipose tissue (WAT) and bone marrow adipose tissue (BMAT) from 22-week-old or 13-week-old male New Zealand White rabbits. From both cohorts, BMAT was sampled from the distal tibia (dBMAT) and the radius and ulna (ruBMAT), while WAT was sampled from the inguinal (iWAT) and gonadal (gWAT) depots. From the 13-week-old cohort, BMAT was also sampled from the proximal tibia (pBMAT). Sufficient RNA could not be isolated from all tissues for all rabbits, so for some rabbits only a subset of tissues is included.

Post-natal development of brown adipocytes in white fat: Phases independent and dependent of the ambient temperature

Project description:Decreased ambient temperature, from birth to weaning, significantly reduced fat mass (FM) and adiposity in developing mice. Changes in FM content in 10 day-old mice raised at 17°C were not accompanied by precocious induction of brown adipocytes in inguinal white adipose tissue (WAT). Brown phenotype was induced in 21 day-old mice independently of ambient temperature (17°C or 29°C) however the expression of Ucp1 and other brown fat biomarker genes was greater in animals raised at 17°C suggesting increased adaptive thermogenesis. Accordingly, we predict that the induction of brown adipocytes in WAT follows a strict developmental program and the number of potential brown adipocytes in WAT is determined genetically. Microarray analysis of gene expression was performed on inguinal white adipose tissue dissected from 10 and 21 day-old mice kept at different temperature conditions, that is, in mice maintained at 17°C or 29oC during early post-natal development.
