TLR signaling in C57BL/6J TLR7-tolerant CD3+ T-cells over the unmanipulated state.

ABSTRACT: Murine CD3+ T-cells were immunomagnetically purified from the spleens of C57BL/6J mice and were pretreated in vitro for three days in the presence of R848 (5 μg/ml). Unmanipulated T-cells served as negative control. We used Qiagen Toll-like Receptor RT2 Profiler PCR Array kit to quantitate gene expression profiling of the TLR signaling pathway.

Project description:The spleenocytes were isolated from 3 male C57Bl/6 mice respectively (wt- dob: 7/14/09; rsq- dob: 6/27/09). The spleens were homogenized in a gentleMACSM-^Y Dissociator (Miltenyi Biotech, Germany). For red blood cell lysis the cells were resuspended in 3ml ACK lysis buffer (Sigma) and incubated for 3 min at RT. 7ml Medium (RPMI/2.5% FCS/1%PSG/1xbeta-mercaptoethanol were added. The cells were washed 1x in medium and resuspended in 5ml. 6x10^5 cells were treated with the following compounds for 4h. (final volume of 500 ul): Treatments: R0006 (5 ul/10E+06 cells + 5 ul DOTAP), DOTAP (5 ul + 5ul water x2 replicates), 10 uM R848 and DMSO. RNA extracted using QUIAShredder/RNeasy columns and stored at -80C until labeling was performed. Labeling was caried out following Agilent QUICKAMP 2-color labeling protocol. Briefly, 400 ng RNA was retrotranscribed and labeled using cyanine3 (reference) or Cyanine5 (treatment) and purified with RNeasy columns. Yield and efficiency of dye incorporation were measured at NanoDrop. Only reactions with yields >0.825 ug cRNA and yields >8 pmol dye/ug cRNA were used for hybridization. TLR agonists on human dendritic cells

Project description:Toll like receptors (TLRs) sense microbial products and initiate adaptive immune responses by activating dendritic cells (DCs). Since pathogens may contain several agonists we asked whether different TLRs may synergize in DC activation. We report that in human and mouse DC TLR3 or TLR4 potently synergize with TLR7, TLR8 or TLR9 in the induction of selected cytokine genes. Upon synergistic stimulation, IL-12, IL-23 and Delta-4 are induced at levels 50-100 fold higher than those induced by optimal concentrations of single agonists, leading to enhanced and sustained TH1 polarizing capacity. Using microarray analysis we show that only 1.5% of the transcripts induced by single TLR agonists are synergistically regulated by combinations of TLR4 and TLR8 agonists.. These results identify a combinatorial code by which DCs discriminate pathogens and provide (suggest) a rationale to design adjuvants for TH1 responses.
