Connexin 43-dependent mesenchymal stem cell mitochondrial transfer activates PGC1a disrupting oxidative inflammation axis in ischemia-reperfusion injury

Project description:Ischemia Reperfusion Injury-related Acute Kidney Injury (IRI-AKI) is a significant clinical challenge characterized by interrupted blood flow to the kidneys, leading to substantial complications such as prolonged hospital stays, chronic kidney disease, and increased mortality rates. Current management strategies are limited to supportive care, underscoring the need for targeted therapies. This study investigates the potential of Mesenchymal Stromal Cells (MSCs) to transfer functional mitochondria to damaged renal cells via gap junctions, specifically connexin 43 (Cx43), in a clinically relevant surgical model of IRI-AKI. We conducted a time-course analysis to identify critical time points for mitochondrial dysfunction and the optimal therapeutic window for MSC administration, comparing the reno-protective effects of bone marrow-derived MSCs (BM-MSCs) and umbilical cord-derived MSCs (UC-MSCs) in both in vivo and in vitro models. Additionally, we explored the activation of mitochondrial biogenesis and dynamics signaling pathways, particularly PGC1α, to assess the restoration of mitochondrial homeostasis in IRI-AKI, and its potential to disrupt the oxidative stress-inflammation axis, paving the way for precision therapies in treating IRI-AKI.

Induction of circular RNA in fetal heart development recapitulated in in vitro differentiation

Project description:We discovered induction of circular RNA in human fetal tissues, including the heart. In this study, we were able to recapitulate this induction by in vitro directed differentiation of hESCs to cardiomyocytes, paving the way for future studies into circular RNA regulation. We harvested hESCs at sequential stages of differentiation: undifferentiated (day 0), mesoderm (day 2), cardiac progenitor (day 5) and definitive cardiomyocyte (day 14). We performed RNA sequencing in biological triplicate, with 3-8 technical replicates each.

Project description:Sarcomatoid renal cell carcinoma (sRCC) is an aggressive trans-differentiation of epithelioid clear cell RCC (ccRCC) tumors that shows heightened response to immunotherapy. The underlying biology leading to sarcomatoid transformation and mechanisms contributing to immunotherapy response are not well understood. Novel single cell spatial techniques were used in ccRCC and sRCC tumors from 40 patients to understand the spatial sRCC transformation and corresponding immune changes. A transcriptional transition state in epithelioid ccRCC cells along a continuum to mesenchymal sRCC was identified which expresses high levels of pro-inflammatory cytokines and an immune infiltrate. In vitro studies demonstrated that M2-like macrophages, recruited to the tumor by the transition state, induce full transition to the sarcomatoid state. A combination of increased PD-L1 expression and T cells recruited by the transition state was observed consistent with the increased immunotherapy response. This study enriches our understanding of the mechanisms leading to development and immune responsiveness of sRCC paving the way for novel approaches to diminish RCC progression.
