HAL2 overexpression induces iron acquisition and enhanced salt resistance in bdf1Δ

Project description:The strain bdf1Δ+HAL2 improved salt resistance of bdf1∆. To gain further insight into the mechanism of bdf1∆ salt sensitivity, DNA microarray analysis was performed to determine the reason for the salt sensitivity of bdf1∆ cells and the process of how HAL2 overexpression and HDA1 deletion improves salt resistance. Transcriptomic analysis under salt treatment (0.5 mol.L-1 NaCl for 45 min) was performed using four different strains: bdf1∆, W303, bdf1Δ+HAL2 and bdf1∆hda1∆. The transcription of 1,393 genes were significantly changed( > 2-fold) in bdf1∆ compared with W303 upon NaCl stress (0.5 mol.L-1 NaCl for 45 min). Only 108 genes were significantly changed ( > 2-fold) in bdf1Δ+HAL2 compared with bdf1∆ upon NaCl stress (0.5 mol.L-1 NaCl for 45 min). Only 88 genes were significantly changed ( > 2-fold) in bdf1Δ+HAL2 compared with bdf1∆ upon NaCl stress (0.5 mol.L-1 NaCl for 45 min).

Project description:The strain bdf1∆bdf2∆[BDF2 L][SIR2 H] improved salt resistance of bdf1∆. To gain further insight into the mechanism of BDF1 in suppressing bdf1∆ salt sensitivity, DNA microarray analysis was performed to determine the reason for the salt sensitivity of bdf1∆ cells and the process of how coexpression of SIR2 and BDF2 improves salt resistance. Transcriptomic analysis under salt treatment (0.6 mol.L-1 NaCl for 45 min) was performed using three different strains: bdf1∆bdf2∆[BDF2 L][SIR2 H], bdf1∆bdf2∆[BDF2 L][pYX242] and bdf1∆[pRS316][pYX242]. The transcription of 3244 genes were significantly changed( > 2-fold) in bdf1∆bdf2∆[BDF2 L][SIR2 H] compared with bdf1∆[pRS316][pYX242] upon NaCl stress (0.6 mol.L-1 NaCl for 45 min). Rhodobacter sphaeroides 2.4.1 pPos19 versus pBBR under singlet oxygen stress

Project description:To learn more about the function of Pos19, we constructed an over-expression strain. For this purpose, the Pos19 gene together with its native RpoE promoter was cloned into the middle-copy plasmid pBBR1 and expressed in the R. sphaeroides wild-type 2.4.1 background. The corresponding strain consequently exhibits a singlet oxygen-induced Pos19 over-expression compared to an empty vector control. Total RNA from the over-expression (pPos19) and from the empty vector control (pBBR) strain was extracted after 7 min of singlet oxygen stress and used for microarray analysis to investigate Pos19-induced changes on transcriptome level.

Rhodobacter sphaeroides 2.4.1 pRKSorX144 versus pRK415 under singlet oxygen stress.

Project description:To learn more about the function of SorX, we constructed an over-expression strain. For this purpose, the sorX gene together with its 28-bp upstream region was cloned into the over-expression vector pRK4352 and expressed in the R. sphaeroides wild-type 2.4.1 background. The corresponding strain consequently exhibits a SorX over-expression that is driven by the constitutive 16S rRNA (RSP_4352) promoter. The over-expression strain was compared to an empty vector control (pRK415). Total RNA from the over-expression (pRKSorX144) and from the empty vector control (pRK415) strain was extracted after 7 min of singlet oxygen stress and used for microarray analysis to investigate SorX-induced changes on transcriptome level.
