Activated Notch-induced transcriptional profile modulation in human primary dermal lymphatic endothelial cells

Project description:Human Notch1 intracellular domain (NICD) was overexpressed in human primary lymphatic endothelial cells (LECs) for 10 and 24 hours by adenovirus. A GFP-control adenovirus-infected cells (24hours) and uninfected cells were also analysed as controls. Total RNAs were harvested and subjected to Affymetrix U133A microarray. Human primary lymphatic endothelial cells (LECs) were isolated from human foreskin and cultured and expanded to population passages 5~6. Healthy subconfluent primary LECs were infected with adenovirus expressing human Notch1 intracellular domain (NICD) for 10 or 24 hours. In parallel, LECs were also infected with a GFP-expressing control adenovirus for 24 hours. Uninfected LECs were also used as a negative control in the same experiments

The NICD is released from the cytoplasmic tail of the Notch receptor by gamma-secretase treatment and translocated as a transcription factor to the nucleus. Here, virally transduced recombinant NICD constructs (wild-type and deltaEP hyperactive mutant) were employed for AP-MS with the aim of identifying novel NICD interactors in a murine T cell leukemia cell line.

Expression profiling of Notch constitutive activation induced HCC in mice

Project description:Notch intracellular domain (NICD) is the active form of the Notch receptor. In this mouse model, NICD is inserted in the Rosa26 locus downstream of a loxP-STOP-LoxP (lsl) sequence and therefore NICD expression is dependant on Cre recombinase expression. These mice are crossed with the AFP-Cre strain that expresses Cre in hepatoblasts due to its regulation by the AFP promoter and albumin enhancer. Mice from 6 to 12 months are sacrificed and liver RNA samples from control monotransgenic Rosa26-lsl-NICD and confirmed HCC lesions from bitransgenic AFP-Cre/Rosa26-lsl-NICD (AFP-NICD) are obtained. Exon expression profiling of these samples are submitted.

Gene expression analysis of SJG6 CONTROL and NICD

Project description:Mutational landscape of Oral Squamous Cell Carcinoma (OSCC) is predominated by frequent inactivating mutations in EGF- like ligand binding domain of NOTCH1. However role of NOTCH1 signalling in tumorigenesis is highly context and cell type dependent. In this study, we investigated phenotypic effect of NOTCH1 mutations in primary line derived from OSCC biopsy. Based on gain-of-function assays in NOTCH1 mutant line we demonstrated regulation of cell behaviour and morphology, by NOTCH1 intracellular domain (NICD) leading to changes in cell proliferation, migration, clonal growth and differentiation. Importantly, overexpression of NICD results in upregulation of ETV7/TEL2 which negatively regulates SERPINE1 expression and confers malignant cells with less aggressive phenotype. Further knockdown of SERPINE1 expression simulated the phenotypes observed upon rescue of NICD. In accord with our cell-based model we observed strong correlation between NOTCH1, ETV7 and SERPINE1 expression in OSCC primary tumours indicating similar deregulated mechanisms during oral carcinogenesis. Overall this study suggests a tumour suppressive role of NOTCH1 in OSCC and highlights possible mechanisms leading to changes in cell behaviour and morphology in absence of NICD potentially by regulating TEL2 and SERPINE1 expression.

Gene profiles induced by overexpression of wild-type and mutant Notch1 variants in MCF10A mammary epithelial cell line

Project description:Expression of a constitutively active Notch-1 intracellular domain (NICD) in MCF-10A cells was found to induce two distinct types of 3D structures: large, hyperproliferative structures and small, growth-arrested structures with reduced cell-to-matrix adhesion. These heterogeneous phenotypes reflect differences in Notch pathway activation levels. High Notch activity caused loss of cell adhesion and inhibition of proliferation, whereas low Notch activity maintained matrix adhesion and provoked a strong hyperproliferative response. In order to gain insight into the dosage-dependent transcriptional events triggered by Notch1 activation, gene expression profiles induced 48 hours after infection of MCF-10A cells with retroviral vectors expressing full-length Notch-1, L1601P+âP, or NICD were compared. Full-length Notch-1 induced the weakest effect, L1601P+âP induced an intermediate effect and NICD induced the strongest effect. Results provide insight into the dichotomous activites of Notch during development and tumorigenesis. Twelve samples were harvested 48 h after retroviral infection with either vector control, full-length Notch1, Notch1 L1601P+âP, or NICD. Each condition was performed in triplicate.
