ABSTRACT: The purpose of this study was to determine what are the effects of Src deficiency on innate antiviral response upon virus infection in RAW264.7 cells. Wild type and Src-/- RAW264.7 cells were infected with vesicular stomatitis virus (VSV) or herpes simplex virus 1 (HSV-1) for 6h. Then the differentially regulated genes were analyzed.

Then the differentially regulated genes were analyzed, focusing on F-box proteins and E3 ubiquitin ligases. RAW264.7 cells were infected with Vesicular Stomatitis Virus (VSV, MOI=1) or herpes simplex virus 1 (HSV-1, MOI=5) for 6h. Project description:We demonstrate that phosphoenolpyruvate (PEP) generally inhibits viral infection in vitro and in vivo. In order to further explore whether PEP controls viral replication through a general mechanism such as enhancing innate response, and if not, what's the critical downstream target(s) of PEP, we performed RNA sequencing (RNA-seq) analysis on the RAW264.7 cells that were pretreated with 250 μM PEP or PBS for 2 hours, and then infected with or without vesicular stomatitis virus (VSV) for 8 hours.

Project description:iBMDM cells were infected with VSV (Vesicular Stomatitis Virus) or HSV-1 (Herpes Simplex Virus 1) for 8 hours, followed by CHIP-seq analysis with H3K27AC antibody (Abcam, ab4729). We find that chromatin state changed with virus infection, some enhacer and superenhancer were induced to promote anti-viral gene expression. Conclusions: virus infection induce enhancer and superenhancer activation.

Effect of depletion of Igf2bp3 on gene expression during VSV infection in RAW 264.7 cells and mice PMs

Project description:N6-methyladenosine (m6A) modification is the most abundant RNA modification in both host mRNA and viral RNA transcripts. Dynamic regulation and recognition of m6A modification widely participate in post-transcriptional regulation of many biological processes. This project aims to define the regulation role of IGF2BP3, a m6A recognition protein, in viral infection, especially in RNA viral infection. In this study, the transcriptomes of Vesicular stomatitis virus (VSV)-infected WT/Igf2bp3-KO RAW264.7 cells and mouse peritoneal macrophages (PMs) were analyzed, suggesting the role of IGF2BP3 in the host's innate immune responses during viral infection and enhancement.
