Parvovirus B19 NS1 protein induces cell cycle arrest at G2 phase

ABSTRACT: We construct two stable UT7/Epo-S1 cell lines which could be inducible expressing B19 NS1 and NS1 TAD2 domain mutation proteins. After treated with or without doxycycline induction, we extract the total RNA for RNA-seq analysis.

The NS1 Protein of the Parvovirus MVM Aids in the Localization of the Viral Genome to Cellular Sites of DNA Damage

Project description:The autonomous parvovirus Minute Virus of Mice (MVM) localizes to cellular sites of DNA damage to establish viral replication centers, visualized by focal depositions of the essential MVM phosphoprotein NS1. However, how such foci are established remains unknown. Here, we show that MVM NS1 localized to cellular sites of DNA damage independently of its known ability to covalently bind the viral genome at its known consensus DNA binding site. NS1 that retained its capacity to bind its cognate DNA binding sequence could relocate either the viral genome, or heterologous DNA, containing those sites, to cellular sites of DNA damage. These findings suggest that another way that NS1 can facilitate virus infection is by localizing the genome to cellular sites which contain replication and expression proteins.

qRT-PCR analysis of primary CD36+ cells infected with parvovirus B19

Project description:The goal of our present work was to understand the influence parvovirus B19 infection may have on the thyroid hormone signaling pathway, as well as the nuclear receptors (NR) pathway overall. We demonstrated that B19 infection of CD36+ erythroid progenitor cells leads to downregulation of the thyroid hormone receptor α isoform. In addition to that we have shown that B19 infection modulates the expression of other members of the NR superfamily such as estrogen and retinoid receptors. CD36+ cells (StemCell Technologies) were mock-infected or infected with B19, 48 hours post infection cells were collected, total RNA was isolated, and cDNA was obtained as described above. TaqMan® array human nuclear receptors fast 96-well plates obtained from Applied Biosystems (Carlsbad, CA) were utilized in order to assess the differences of 92 nuclear receptors’ expression in mock- and B19-infected CD36+ cells. Relative quantity (RQ) values were calculated using the 2-ΔΔCt method.

TP53 sequence of human glioblastoma stem cells sorted for p53-Ser15 induction and NS1 virus protein expression upon parvovirus MVMp infection

Project description:We have analyzed whether mutations in the TP53 gene and post-translational phosphorylation in the Ser-15 residue of the p53 protein (Pp53) may confer permissiveness to the prototype strain of the parvovirus Minute Virus of MIce ( MVMp) infection in human glioblastoma stem cells (GSCs) isolated from brain tumours. Infected (MVMp samples) and control uninfected (mock samples) GSCs from four patients (GS5, GS7, GS8, GS9) were sorted for the Pp53 modification (which is required for virus genome replication) and the NS1 protein (a major marker of virus cytotoxic gene expression). Cells sorted through restrictive gates for the NS1+/Pp53+ and NS1+/Pp53- phenotypes were proceeded for mRNA purification, the TP53 gene was amplified with specific primers and subjected to NGS analysis.

Design and characterization of mutated variants of the oncotoxic parvoviral protein NS1

Project description:Oncotoxic proteins such as the non-structural protein 1 (NS1), a constituent of the rodent parvovirus H1 (H1-PV), offer a novel approach for treatment of tumors that are refractory to other treatments. In the present study, mutated NS1 variants were designed and tested with respect to their oncotoxic potential in human hepatocellular carcinoma cell lines. We introduced single point mutations of previously described important residues of the wild-type NS1 protein and a deletion of 114 base pairs localized within the N-terminal domain of NS1. Cell-viability screening with HepG2 and Hep3B hepatocarcinoma cells transfected with the constructed NS1-mutants led to identification of, the single-amino acid NS1-mutant NS1-T585E, which lead to a 30% decreased in cell viability as compared to NS1 wildtype. Using proteomics analysis, we could identify new interaction partners and signaling pathways of NS1. We could thus identify new oncotoxic NS1 variants and gain insight into the modes of action of NS1, which is exclusively toxic to human cancer cells. Our in-vitro studies provide mechanistic explanations for the observed oncolytic effects. The availability of new NS1 variants in combination with a better understanding of their modes of action offers new possibilities for the design of innovative cancer treatment strategies.

Project description:Human parvovirus B19 (B19V) infection can cause transient aplastic crisis, persistent viremia, and pure red-cell aplasia. In fetuses, B19V infection can result in non-immune hydrops fetalis and fetal death. To systematically investigate the interaction between B19V and erythoid progenetor cells (EPC), microarray was applied to systematically analyze the dynamic transcriptome of CD36+ EPCs during B19V infection.
