Project description:Tissue fibrosis is a common pathway to organ injury and failure. It is characterized by an excessive deposition of extracellular matrix (ECM) in organs. Deciphering the fibrogenic processes is of utmost importance, as there are few effective therapies in fibrotic diseases 1. Systemic sclerosis (SSc) is a prototypical disease where fibroblasts (Fb) are key effector cells as they differentiate into myofibroblasts in response to chronic inflammation under the influence of transforming growth factor beta 1 (TGF-β1) pathway 2–4. In this study, we compared the proteome of primary Fb in different culture and stimulation conditions. Primary dermal normal human Fb were cultured at passage P3, P5 and P7 with and without Fetal Bovine Serum (FBS). At fifth passage, Fb were stimulated or not with different concentrations of recombinant human active TGF-β1 (0.04, 1 and 5 ng/mL) during 24, 48 and 72 hours.

Project description:Postnatal tissue quiescence is generally thought to be a default state in the absence of a proliferative stimulus such as injury. We now demonstrate that in the lung, quiescence in the adult is an actively maintained state and is regulated by paracrine hedgehog signaling. Epithelial-specific deletion of Sonic Hedgehog during normal homeostasis results in a proliferative expansion of the adjacent lung mesenchyme. Injury to the lung epithelium results in decreased hedgehog activation, accompanied by proliferative expansion of the adjacent mesenchyme. Moreover, reconstitution of Hedgehog signaling during epithelial injury attenuated the proliferative expansion of the adjacent mesenchyme. Hedgehog signaling maintains lung quiescence by attenuating PDGF signaling through blocking post-translational processing of PDGF receptor α/β into the mature isoforms. These results indicate that in postnatal tissues, epithelial cells can actively maintain mesenchymal quiescence via paracrine hedgehog activation, and that imbalances in this pathway could lead to aberrant mesenchymal expansion and postnatal disease. Fibroblasts were isolated from mouse lungs and grown in culture in triplicate wells. Samples were treated with vehicle or purmorphamine 5um for 24 hours and RNA was isolated for microarray.

Project description:<p><strong>INTRODUCTION:</strong> A decline in mitochondrial function represents a key factor of a large number of inborn errors of metabolism, which lead to an extremely heterogeneous group of disorders.</p><p><strong>OBJECTIVES:</strong> To gain insight into the biochemical consequences of mitochondrial dysfunction, we performed a metabolic profiling study in human skin fibroblasts using galactose stress medium, which forces cells to rely on mitochondrial metabolism.</p><p><strong>METHODS:</strong> Fibroblasts from controls, complex I and pyruvate dehydrogenase (PDH) deficient patients were grown under glucose or galactose culture condition. We investigated extracellular flux using Seahorse XF24 cell analyzer and assessed metabolome fingerprints using NMR spectroscopy.</p><p><strong>RESULTS:</strong> Incubation of fibroblasts in galactose leads to an increase in oxygen consumption and decrease in extracellular acidification rate, confirming adaptation to a more aerobic metabolism. NMR allowed rapid profiling of 41 intracellular metabolites and revealed clear separation of mitochondrial defects from controls under galactose using partial least squares discriminant analysis. We found changes in classical markers of mitochondrial metabolic dysfunction, as well as unexpected markers of amino acid and choline metabolism. PDH deficient cell lines showed distinct upregulation of glutaminolytic metabolism and accumulation of branched-chain amino acids, while complex I deficient cell lines were characterized by increased levels in choline metabolites under galactose.</p><p><strong>CONCLUSION:</strong> Our results show the relevance of selective culture methods in discriminating normal from metabolic deficient cells. The study indicates that untargeted fingerprinting NMR profiles provide physiological insight on metabolic adaptations and can be used to distinguish cellular metabolic adaptations in PDH and complex I deficient fibroblasts.</p>

Project description:To clarify the molecular alterations triggered by miR-9 to induce the conversion of breast normal fibroblasts to a CAF-phenotype, gene expression profile of normal fibroblasts transiently transfected with miR-9 or control was performed. We performed a gene expression profiling of immortalized normal fibroblasts taken from a noncancerous region of the breast at least 2 cm from the outer tumor margin. Fibroblasts were transiently transfected with miR-9 or control for 24 hours. 3 independent biological replicates for each treatment.

Project description:Arabidopsis plants (ecotype Ler) were grown under short day conditions (8h cool white fluorescent light at 100 uE, 16h dark) at 20 °C and ambient humidity in PGW36 phytochambers (Controlled Environment Ltd., Winnipeg, MB, Canada). Plants were grown in Redi soil (Grace & Co, Ajax, ON, Canada) in 12 cm pots at a density of five plants per pot without fertilization for 8 weeks. Light conditions were then changed to long day (16h light, 8h dark), and plants were fertilized once with standard 10-15-10 plant fertilizer (150 ul in 300 ml water per pot) (Schultz, St. Louis, MO, USA). Twelve and 14 days after changing to long day conditions we harvested sections from stems totalling 4-6cm (“5cm stem”) and 9-11cm (“10cm stem”) in length, respectively. All flowers, siliques, cauline leaves, and secondary branches were discarded and the remaining bolting stems were cut with a razor blade to generate the following sections: 0-2cm and 2-4cm from 5cm stems; and 0-3cm, 3-5cm, 5-7cm, and 7-9cm from 10cm stems. For array hybridizations, labelled cDNA derived from each section was co-hybridized with cDNA from 0-2cm sections as a reference.
