Profiling of RNA expression in soleus muscles from muscle-specific Gna13 knockout mice

ABSTRACT: Gna13 transmit extracellular signals from cell surface G protein-coupled receptors to intracellular effector molecules. We have characterized the Gna13-dependent regulatory network in muscle through transcriptional profiling of the mouse soleus in Gna13 muscle-specific knockout (Gna13 MKO) mice or wild-type littermates with floxed genotype (WT).

Keyword: Circadian Transcriptome, time course 72 samples were analyzed, comprised of 4 experimental groups (Ctrl SOL, mKO SOL, Ctrl TA, mKO TA), with 3 biological replicates for each time point sampled every 4 hours for 24 hours. Project description:To understand the role of LSD1 in transcriptional regulation in muscle under voluntary wheel running (VWR) training, RNA-seq analyses of soleus muscles of skeletal muscle-specific LSD1 KO mice (LSD1-mKO mice) and WT mice after VWR were carried out. We found that loss of LSD1 led to increased expression of oxidative metabolism genes in soleus muscle.

Project description:To understand the role of LSD1 in transcriptional regulation in muscle under glucocorticoid stress, RNA-seq analyses of gastrocnemius and soleus muscles of skeletal muscle-specific LSD1 KO mice (LSD1-mKO mice) and WT mice after dexamethasone were carried out. We found that LSD1 inhibition led to increased expression of muscle atrophy associated genes and slow fiber genes in gastrocnemius muscle but not in soleus muscle.

Effect of USP21 deficiency on transcriptomes in skeletal muscle of mice

Project description:Skeletal muscle is the pivotal organ for energy homeostasis, but the regulatory mechanisms are largely unknown. Here, we report that ubiquitin specific protease 21 (USP21) is a regulator of skeletal muscle physiology. To elucidate features important for USP21, we performed RNA-sequencing analysis using the muscle tissue of muscle-specific USP21 knockout (USP21-MKO) mice or wild-type littermates with floxed genotype, screened and analyzed the differentially expressed genes. Transcriptomics assay enabled us to discover major signaling nodes persistently activated by USP21 ablation in close association with the genes linked to fuel oxidation. Consequently, USP21 ablation altered gross metabolic phenotypes.

RNA-sequencing of soleus muscle from Mustn1 heterozygous KO-first mice and wildtype littermates

Project description:To uncover novel genes and molecular pathways important for skeletal muscle remodeling and regeneration, we used a mouse hindlimb unloading and reloading protocol. We then performed analysis of global gene expression by RNA-sequencing (RNA-seq), looking at the transition from muscle unloading to reloading (GSE237099). This study focuses on the microprotein Mustn1 (Musculoskeletal embryonic nuclear protein 1, also known as Mustang), which consists of 82 amino acids. We performed RNA-sequencing of soleus muscle from 8-week-old mice carrying one allele (heterozygous) of Mustn1 tm1a (KO-first) and wildtype littermates.
