Project description:Laminin-mediated signaling via membrane receptors actively participate in most normal biological process and pathologies. Laminins involve in all step of metastatic cascade including tumor cell proliferation, migration, anoikis resistance, extravasation, epithelial to mesenchymal transition etc. The 5-containing laminins (LAMA5) are one of the three types of laminins predominantly expressed in the epithelial basement membrane in the adult intestine as well as during crypt-villus development. Likewise laminins 511 and 521 (laminin-10 and laminin-11) have been shown as important matrix component to facilitate stem cell survival and to maintain their stem cell properties. Thus, it is assuming that laminins play a role in the formation of premetastatic niches. To investigate the role of 5-containing laminins in colorectal cancer we performed the knockdown of LAMA5 gene expression in HT-29 cell line. Here we present the results of comprehensive gene expression analysis using Affymetrix microarrays in cells transduced by shRNA to LAMA5, control nonsense shRNA, and parental cells as well.

Transcription profiling of mouse laminin alpha 2 chain deficient animals vs wild type

Project description:Mutations in the gene encoding laminin a2 chain cause congenital muscular dystrophy, MDC1A. In skeletal muscle, laminin a2 chain binds at least two receptor complexes; the dystrophin-glycoprotein complex and integrin a7b1. To gain insight into the molecular mechanisms underlying this disorder, we performed gene expression profiling of laminin a2 chain deficient mouse limb muscle. One of the down-regulated genes encodes a protein called calcium and integrin binding protein 2 (Cib2) whose expression and function is unknown. However, the closely related Cib1 has been reported to bind integrin aIIb and may be involved in outside-in-signaling in platelets. Since Cib2 might be a novel integrin a7b1 binding protein in muscle, we have studied Cib2 expression in the developing and adult mouse. Cib2 mRNA is mainly expressed in the developing central nervous system and in developing and adult skeletal muscle. In skeletal muscle Cib2 colocalizes with integrin a7B subunit at the sarcolemma and at the neuromuscular- and myotendinous junctions. Finally, we demonstrate that Cib2 is a calcium binding protein that interacts with integrin a7Bb1D. Thus, our data suggest a role for Cib2 as a cytoplasmic effector of integrin a7Bb1D signaling in skeletal muscle Experiment Overall Design: Skeletal muscle (all hind limb skeletal muscle) from 4-weeks old laminin alpha 2 chain deficient mice and 4-weeks old wild type mice were isolated individually and RNA were extracted and hybridized on Affiymetrix microarrys. Three biological replicates from each group were analyzed.

Next generation seuquencing of 3D breast cancer cell culture in synthetic hydrogels presenting either a collagen mimic or laminin mimic

Project description:MDA-MB-231 (ER-) and T47D (ER+) breast cancer cells were encapsulated in synthetic hydrogels, formed with a multi-arm bioinert polymer poly(ethylene glycol), cell-degradable peptide crosslinks, and receptor-binding pendant peptides, and cultured for 3 days to assess initial cell responses to different matrix compositions. The synthetic matrices had a Young's modulus (E) ~ 0.6 kPa, in the range of bone marrow and healthy mammary tissue, and biochemcial cues to mimic binding sites found in collagen (GFOGER peptide) or laminin (IKVAV peptide). After 3 days in culture, mRNA was collected and RNA-seq performed to compare cell responses between the collagen and laminin mimetic cultures.
