Project description:Affymetrix soybean genome arrays were used to identify genes differentially expressed in the immune resistance response at 6, 12, 24, and 48 hours after inoculation with Phakopsora pachyrhizi isolates TW72-1 or HW94-1; soybean + HW94-1 = resistant; soybean + TW72-1 = susceptible Experiment Overall Design: For each of 4 time points (6, 12, 24 and 48 hours post inoculation) a soybean PI200492 plant was inoculated with 2 treatments: HW94-1, or TW72-1. A mock inoculation was performed and sampled at time 0. There were three biological replicates of the entire experiment, for a total of 27 samples. During data analysis two samples failed the quality control metrics and were removed from the analysis, leaving 25 samples in the final data set.

Transcription profiling of soybean with respect to Pahakopsora pachyrhizii at V2 growth stage

Project description:A small number of genes (25) expressed differentially with respect to Phakopsora pachyrhizii at V2 growth stage.. We used Affymetrix soybean microarrays to detail the global gene expression underlying host reaction to Phakopsora pachyrhizii and identified differentially expressed genes during this process. Experiment Overall Design: Soybean cv. 5601T plants were grown in greenhouse in six 4-liter pots. Three weeks old plants at V2 growth stage in three out of six pots were inoculated with Phakopsora pachyrhizi and remaining three pots were used as control. Leaf samples from all pots were collected 72 hours after inoculation and RNA was extracted using Tri-Regent. Microarray analysis was performed using Affymetrix platform.

Response of Rpp4-silenced plants to soybean rust

Project description:Experimental design: 1 genotypes: PI- (resistant USDA Plant Introduction (PI459025B) line containing SBR Rpp4 resistance gene) 2 treatments: Rub inoculation with BPMV-VIGS construct targeting the LRR region of Rpp4 candidate gene. This BPMV construct and the plant tissue used for microarray was described in Meyer et al. (2009) Plant Physiol. 150:295-307. Control plants were inoculated with empty BPMV vector. At three weeks after BPMV inoculation, all plants were inoculated with Soybean rust (Phakopsora pachyrhizi) isolate LA04-1. Transcriptional response to soybean aphid infestation in susceptible and resistant soybean plants

Project description:Soybean aphids are phloem-feeding pests that can cause significant yield losses in soybean plants. Soybean aphids thrive on susceptible soybean lines but not on resistant lines. We used microarrays to characterize the soybean plant's transcriptional defense against aphids in two related cultivars, a susceptible line and a resistant line with the Rag1 aphid-resistance gene. We measured trancript levels in leaves after one and seven days of aphid infestation.

Project description:Chinese soybean (Glycine max (L.) Merr.) cultivars Rsmv1 and Ssmv1 were used for soybean mosaic virus (SMV) resistance genes screening. The Rsmv1 cultivar was highly-resistant to SMV but the Ssmv1 cultivar was highly-susceptible. We used microarrays to detail the global programme of gene expression underlying SMV inoculation and identified distinct expression genes between Rsmv1 and Ssmv1.
