Project description:To examine the response of Candida glabrata cells to iron-depleted and iron-repleted environmental conditions, transcriptional profiling analysis was carried out on wild-type and Cghog1∆ cells grown either in presence of BPS or ferric chloride. Mouse spleen expression profile after systemic Candida glabrata infection

Project description:Pleiotropic type I interferons (IFNs-I) fulfil multiple protective functions during infectious diseases, but can also exert detrimental effects for the host leading to immunopathology. Here, we report that IFNs-I promote the dysregulation of Fe homeostasis in macrophage populations and in the spleen during systemic infections with the intracellular pathogen Candida glabrata, leading to fungal survival and persistence. By using microarray expression profiling with RNA isolated from mouse spleens upon systemic Candida glabrata infection and subsequent in vitro interaction experiments we show that IFN-I signaling induces global transcriptional downregulation of key players in Fe homeostasis and profoundly alters Fe transport mechanisms within macrophages.

A unique iron homeostasis regulatory circuit with reciprocal roles in Candida albicans commensalism and pathogenesis [ChIP_chip]

Project description:The mammalian gastrointestinal tract and the bloodstream are highly disparate biological niches, and yet certain commensal-pathogenic microorganisms are able to thrive in both environments. Here, we report the evolution of a unique transcription circuit in the yeast, Candida albicans, which determines its fitness in both host niches. Our comprehensive analysis of the DNA-binding proteins that regulate iron uptake by this organism suggests the evolutionary intercalation of a transcriptional activator called Sef1 between two broadly conserved transcriptional repressors, Sfu1 and Hap43. The Sef1 activator of iron uptake genes promotes virulence in a mouse model of bloodstream infection, whereas the Sfu1 repressor is dispensable for virulence but promotes gastrointestinal commensalism. We propose that the ability to alternate between genetic programs conferring resistance to iron depletion in the bloodstream versus iron toxicity in the gut may be a fundamental attribute of gastrointestinal commensal-pathogens. ChIP analyses to profile genome-wide of distribution of Sef1, Sfu1 and Hap43 in response to various iron availability. 12 independent ChIP experiments were performed on 6 biological replicates of the untagged control and 2 biological replicates each of Sef1-Myc, Sfu1-Myc, and Hap43-Myc.

Transcriptomic profiling of Candida glabrata under eltrombopag (ETP) treatment

Project description:This study aimed to investigate the transcriptional responses of Candida glabrata to eltrombopag (ETP) treatment. RNA sequencing analysis identified 122 differentially expressed genes under ETP treatment, including 44 upregulated and 78 downregulated genes. The upregulated genes were associated with stress responses and iron homeostasis, whereas the downregulated genes were mainly related to mitochondrial respiration. GO and KEGG enrichment analyses showed that ETP treatment was associated with changes in pathways related to mitochondrial function, iron-dependent processes, and cellular stress responses. These data provide transcriptomic evidence that ETP affects iron homeostasis and mitochondrial respiration-related pathways in C. glabrata.

Functional Genomics Analysis of the Yeast Iron Responsive Transcription Factor Aft1 Reveals Iron-Independent Functions

Project description:The Saccharomyces cerevisiae transcription factor Aft1 is activated in iron-deficient cells to induce the expression of iron regulon genes, which coordinate the increase of iron uptake and remodel cellular metabolism to survive low iron conditions. In addition, Aft1 has been implicated in numerous cellular processes including cell cycle progression and chromosome stability; however it is unclear if all cellular effects of Aft1 are mediated through iron homeostasis. To further investigate the cellular processes impacted by Aft1 using genome-wide synthetic lethal and synthetic dosage lethal screens, we identified greater than 70 deletion mutants that are sensitive to perturbations in AFT1 levels. Our genetic network reveals that Aft1 impacts a diverse range of cellular processes, including the RIM101 pH pathway, cell wall stability, DNA damage, protein transport, chromosome stability and mitochondrial function. Surprisingly, only a subset of mutants identified are sensitive to iron fluctuations or display genetic interactions with mutants of iron regulon genes AFT2 or FET3. We demonstrate that Aft1 works in parallel with the RIM101 pH pathway and the role of Aft1 in cell wall structure and DNA damage repair is mediated by iron. In contrast, we show that the role of Aft1 in chromosome maintenance is independent of its iron regulatory role and that alterations in iron levels do not impact chromosome loss rates. Our study clearly demonstrates a novel iron-independent role for Aft1.
