Mesenteric adipose tissue of rats treated with oleoyl-estrone

ABSTRACT: The aim of this work was to determine if OE treatment affects the expression of genes that regulate lipid metabolism in white adipose tissue. Gene expression in adipose tissue from female treated rats (48 hours) was analysed by hybridization to cDNA arrays and levels of specific mRNAs were determined by Real-Time PCR. Treatment with OE decreased the expression of 232 genes and up-regulated 75 other genes in mesenteric white adipose tissue. Short-term treatment with OE affects adipose tissue capacity to extract fatty acids from lipoproteins and to deal with fatty acid transport and metabolism. The rats were given a daily oral gavage of 10 Î¼mol/kg/day of oleoyl-estrone (OED, Barcelona, Spain) in 0.2 ml of sunflower oil during two days, by means of stomach cannulae. Controls received only the oil gavage. After 48 hours of the beginning of treatment, the mesenteric white adipose tissue was dissected and frozen in liquid nitrogen and stored at -80ÂºC. Total RNA was extracted from adipose mesenteric tissue using Tripure isolation kit. Gene expression was analysed by hybridization to cDNA arrays (AtlasTM Rat Array 1.2 from BD-Clontech, Mountain View CA, USA). Two samples from control animals and two samples from treated animales were analyzed.

Tissue-level dissection of obesity-exercise axis in adipose-muscle tissues

Project description:We studied the opposing effects of exercise training and high-fat diet at tissue-level resolution to reveal gene expression and pathway changes in subcutaneous white adipose tissue, visceral white adipose tissue, and skeletal muscle. We revealed gene modules consistently up-regulated by exercise training, which include fatty acid biosynthesis/beta-oxidation/metabolism, oxidative phosphorylation, TCA cycle, and ROS response. Modules down-regulated by exercise training in all three tissues are antigen presentation, neutrophil degranulation, immune cell migration, phagosome-related genes, ECM-related genes, proliferation, and ribonucleotide biosynthesis. Taken together, exercise training improves fatty acid metabolism and cellular respiration similarly across the three metabolic tissues, and represses immune, ECM, proliferation and tissue-specific pathways.

Gene expression analysis of omental and mesenteric adipose tissue in Crohn's disease and ulcerative colitis

Project description:Our hypothesis is that in IBD patients intestinal bacteria translocation into the intra-abdominal fat depots affects adipocyte morphology and gene expression. The study aimed to study adipocyte gene expression of omental (OM) and mesenteric (MES) adipose tissue of ulcerative colitis (UC) and crohn's disease (CD). Total RNA was extracted from isolated adipocytes from omental and mesenteric adipose tissue of CD and UC patients. Microarray experiments were performed in duplicates of 4 different pools of RNAs extracted from adipocytes isolated from OM and MES of UC patients (n=5) and CD patients (n=5) respectively.

Project description:Two types of adipose tissues, white and brown, are found in mammals. Increasingly novel strategies are being proposed for the treatment of obesity and its associated complications by altering amount and/or activity of BAT using mouse models. We used microarrays to detail the global programme of gene expression in subcutaneous white adipose tissue and brown adipose tissue. White adipose tissue (Subcutaneous region) and brown adipose tissue (intrascapular) were isolated from LACA mice (male, 25 ± 3g ) for RNA extraction and hybridization on Affymetrix microarrays.

Expression data from Adipose Stem Cells (ASC) from morbidly obese and non-obese individuals

Project description:The adipose tissue is an endocrine regulator and a risk factor for atherosclerosis and cardiovascular disease when by excessive accumulation induces obesity. Although the adipose tissue is also a reservoir for stem cells (ASC) their function and “stemcellness” has been questioned. Our aim was to investigate the mechanisms by which obesity affects subcutaneous white adipose tissue (WAT) stem cells. We used microarrays to analyze differences in transcriptomic profiles between the adipose stem cells from morbidly obese and non-obese individuals. Subcutaneous white adipose tissues that were obtained during bariatric surgery (Obese) or liposuction (Lean) were donated by patients after obtaining informed consent. Three cases with BMI>40 kg/m2 (ASCmo) and three controls with BMI<25 kg/m2 (ASCn) were selected and processed extracting total RNA, processing and hybridizating on microarrays.

GDF15 links beta-adrenergic induced lipolysis with anxiety

Project description:Psychological stress activates the autonomic nervous system to protect organisms through an integrated response involving changes to both behavior and metabolism. Epinephrine is an important driver of this response, activating adipose tissue lipolysis3and inducing anxiety-like behavior. Interestingly, anxiety correlates with circulating free fatty acid levels and can be alleviated by a b-blocker that does not cross the blood brain barrier suggesting a peripheral signal linking metabolism with behavior, however, to date this has not been defined. Herein, we identify that the most upregulated secreted gene product induced by epinephrine within white adipose tissue of mice is growth differentiating factor 15 (GDF15). This increase in GDF15 occurs within one hour and is not observed in liver, brown adipose, kidney, or skeletal muscle, and is also observed with the more specific b2/3-agonist, CL316,243. Genetic inhibition of adipose triglyceride lipase (ATGL), the rate limiting enzyme for the release of free fatty acids in white adipose tissue, or genetic deletion of b-adrenergic receptors, blocks b-adrenergic induced increases in GDF15. Increases in circulating GDF15 are not derived from adipocytes but instead require free fatty acid-induced stimulation of macrophages within white adipose tissue. Remarkably, anxiety-like behavior elicited by epinephrine or restraint stress are eliminated in mice lacking the GDF15 receptor, GFRAL. Importantly in patients with severe burns, increases in catecholamines are associated with GDF15 and this relationship is eliminated with the b-blocker propranolol. These data provide important insight into the mechanisms linking metabolism and behavior and suggest that inhibition of GDF15-GFRAL signaling may be important for reducing acute anxiety.
