Tracing immunological interaction in trimethylamine N-oxide hydrogel-derived zwitterionic microenvironment during promoted diabetic wound regeneration

Project description:The diabetic wound healing is challenging due to the sabotaged delicate balance of immune regulation via an undetermined pathophysiological mechanism, so it is crucial to decipher multicellular signatures underlying diabetic wound healing and seek therapeutic strategies. Here, we develop a strategy using novel trimethylamine N-oxide (TMAO)-derived zwitterionic hydrogel to promote diabetic wound healing, and explore the multi-cellular ecosystem around zwitterionic hydrogel, mapping out an overview of different cells in the zwitterionic microenvironment using single-cell RNA sequencing. The diverse cellular heterogeneity has been revealed, highlighting the critical role of macrophage and neutrophils in managing diabetic wound healing. It is found that zwitterionic hydrogel can upregulate chemokine-secreted macrophages and downregulate extracellular traps (NETs)-neutrophils and facilitate their interactions compared with polyanionic and polycationic hydrogels, validating the underlying effect of zwitterionic microenvironment on the activation of adaptive immune system. These findings expand our horizons of the sophisticated orchestration of immune systems in zwitterion-directed diabetic wound repair and uncover new strategies of novel immunoregulatory biomaterials.

Cholangiocyte organoid culture in hydrogels from decellularized liver tissue

Project description:Culture of human cholangiocyte organoids in hydrogels derived from healthy liver extracellular matrix (LECM) extracts prepared from decellularized human livers are evaluated in an effort to establish a platform for production of cholangiocyte organoids for clinical regenerative applications. Human intrahepatic cholangiocyte organoids (ICO) grown in hydrogels made from LECM are compared those grown in mouse tumor derived basement membrane extracts (BME). Culture was performed with amino acids labeled with stable heavy isotopes to enable separation of ECM from the hydrogels from that of the produced by the cells with mass spectrometry (MS). MS data were used to evaluate the protein production of ICO comparing the different hydrogel substrates. The study also contains evaluation of the properties of the hydrogel substrates and focuses on expansion and differentiation of the ICO.

RNA-Seq of human spinal cord organoids generated in Matrigel and alginate

Project description:This experiment compares the transcriptome profile of human spinal cord organoids generated from iPSCs in Matrigel, 1% alginate hydrogel, or 2% alginate hydrogel. Spinal cord organoids were generated from human iPSCs and then encapsulated in the respective hydrogels before further maturation. Organoids were harvested on days 30, 60, and 90 for each group. In addition to the differences between hydrogel groups, neuronal and glial markers at each time point were also assessed for the development of the organoids.

Project description:To determine the performance of the decellularized GI tissue ECM hydrogel, we analyzed differences in mRNA expression levels in native gastric tissues, gastric organoids cultured in Matrigel and those cultured in decellularized gastric ECM hydrogel. We also compared mRNA expression levels of native small intestinal tissues, small intestinal organoids grown in Matrigel and those grown in decellularized intestinal ECM hydrogel.

Drugless Extracellular Hydrogels Enable the Reprogramming of Malignant Glioblastoma

Project description:Glioblastoma multiforme (GBM) poses a significant challenge in clinical practice due to its poor prognosis and high recurrence rate. Despite several advancements in GBM therapy, it often causes neural cell death near the tumor. Furthermore, the aggressive and invasive nature of GBM frequently leads to recurrence near the resection site. Herein, this study introduces a novel drug-free therapeutic approach by injecting a tumor-reprogrammable and extracellularly activatable hydrogel, termed ‘TREAT’ gel. The soft, injectable hydrogel, which is injected into the tumor resection cavity, adapts to the cavity’s shape, interfaces with residual tumor cells and induces genetic reprogramming of malignant GBM cells, while preserving brain cells and even restoring brain function.

Single cell analysis of iPSC-derived cortical organoids in biofunctionalized gelatin hydrogels

Project description:Human neural organoid models have become an important tool for studying neurobiology. In this work, we compared Matrigel to an N-cadherin peptide-functionalized gelatin methacryloyl hydrogel (termed GelMA-Cad) for culturing cortical neural organoids. Specifically, we compare five materials: (1) Matrigel, (2) GelMA-Cad with high crosslinker (HC), (3) GelMA-Cad with low crosslinker (LC), (4) GelMA HC and (5) GelMA LC. We profiled these organoids at the earliest stages to understand potential differences in radial glia formation.

Mechanically and Chemically Defined PEG Hydrogels Improve Reproducibility in Human Cardioid Development

Project description:Cardioids are three-dimensional self-organized heart organoids directly derived from induced pluripotent stem cells (hiPSCs) aggregates. The growth and culture of cardioids is either conducted in suspension culture or heavily relies on Matrigel encapsulation. Despite the significant advancements in cardioid technology, reproducibility remains a major challenge, limiting their widespread use in both basic research and translational applications. Here, for the first time, we employed synthetic, matrix metalloproteinase (MMP)-degradable polyethylene glycol (PEG)-based hydrogels to define the effect of mechanical and biochemical cues on cardioid development. We demonstrated successful cardiac differentiation in all the hydrogel conditions, while cardioid cultured in optimized PEG hydrogel (3wt% PEG-2mM RGD) underwent similar morphological development and comparable tissue functions to those cultured in Matrigel. Matrix stiffness and cell adhesion motif play a critical role in cardioid development, nascent chamber formation, contractile physiology, and endothelial cell gene enrichment. More importantly, synthetic hydrogel improved the reproducibility in cardioid properties compared to traditional suspension culture and Matrigel encapsulation. Therefore, PEG-based hydrogel has the potential to be used as an alternative to Matrigel for human cardioid culture in a variety of clinical applications including cell therapy and tissue engineering.
