Transgenerational effect of Chlordecone [RNA-seq]

ABSTRACT: RNA seq project designed to understand the role of pesticide on transription of mRNA in the testis. We treated adult female mice with chlordecone with doses 0 (control), 100 Xg/kg/day and 1 mg/kg/day during gestation period E6- E15. Male born from Chlordecone- treated mice were crossed for a three generations. The testes from F3 generations were taken for mRNA extraction. For mRNA extraction we used columb. Three biological replicates used for each condition . We hypothesize that chlordecone cause changes in mRNA due to transgenerational effect of pesticides (Skinner et al, Science 2005). The changes in expression of certain genes can help us understand the effect of pesticides on many tissues. Number of sample :9 samples. We require Paired End sequencing and number of reads equal or more than 25M tags, strand specific sequencing

The testes of the third generations males were used for anti-H3K4me3 ChiP-seq analysis. We used two biological replicates for each condition.

To test our hypothesis, we exposed mice to chlordecone during embryonic development and we analyzed F1 female progeny epigenetic mark H3K4me3 in ovaries. We performed ChIP using anti-H3K4me3 commercial antibody (Merck Millipore, 07-473). DNA-protein complexes were immunoprecipitated, eluted and then DNA were purified.

Embryonic day 15 chick utricle single cell analysis

Project description:Single-cell proteomics can reveal the changing protein composition of differentiating cells. We used shotgun mass spectrometry to determine the abundant proteins present in single or small pools of subpicoliter-sized cells from the embryonic day 15 (E15) utricle of the chicken inner ear, when many hair cells are differentiating from progenitor (supporting) cells. The actin monomer binding protein thymosin β4 (TMSB4X) was present in E15 progenitor cells at nearly equimolar levels relative to actin, but dropped to one-tenth that value in hair cells, with little change in total actin. Single-cell RNA-seq analysis of E15 utricle cells showed that TMSB4X transcripts fell in abundance once hair-cell differentiation initiated. These results suggest that most actin is sequestered in progenitor cells, but upon differentiation to hair cells, actin is released, permitting assembly of the sensory hair bundle.

In this project we study whether the exposure to neonicotinoid pesticide, thiaclopride, during embryonic development, could cause gene expression deregulation in outbred Swiss mice. Mice were treated during embryonic development at dose 6mg/kg body weight/day. New-borns were grown until 35 days-old. RNA was extracted from testes of three different mice for each condition (control vs exposed) and stranded libraries were prepared.
