Gene expression analysis of papillary and reticular dermis microdissected from the same skin sample

ABSTRACT: The dermis is divided into two distinct layers. The upper, papillary dermis is characterized by thin and sparse fibers. The lower, reticular dermis is composed of solid tissue made up of thicker highly dense fibers. It has been reported that cultured fibroblasts isolated from the papillary and the reticular dermis exhibit different properties. In this study, we analyzed gene expression profiles of human papillary dermis and reticular dermis obtained using laser capture microdissection.

Expression data from human dermal fibroblast subpopulations

Project description:Fibroblasts are the main dermis-resident cells, yet they remain poorly characterized. Fundamentally, fibroblasts originate from the same population of mesenchymal cells but fibroblast subpopulations have been identified. Papillary and reticular fibroblasts were characterized based on their respective location in the papillary and reticular dermis. Here, we identified a new subset of fibroblasts, located in human papillary dermis and displaying specific features such as their organization into cell clusters. We used microarrays to detail the gene expression profiles of the novel fibroblast subpopulation we have identified compared to papillary and reticualr fibroblasts and notably the relative contribution of the different fibroblast subpopulations to the extracellular matrix of the dermis.

Human dermal fibroblast subpopulations display distinct gene signatures related to cell behaviours and matrisome.

Project description:The upper papillary and deeper reticular dermis differ structurally and functionally. Although the papillary and reticular fibroblasts produced distinct extracellular matrix (ECM), the matrisome of these two fibroblast subpopulations has not been defined. Therefore we performed a transcriptomic analysis of papillary and reticular fibroblasts freshly isolated from skin of young donors around 20, at a time they produced high level of ECM. Bioinformatics analysis delivered 230 upregulated and 139 downregulated transcripts in papillary vs reticular fibroblasts. Expression of various selected genes was validated by q-PCR. The papillary fibroblasts were characterized by a higher expression of genes involved in the defense function, in the regulation of cell motility and proliferation and in the MAPK cascade whereas reticular fibroblasts showed higher expression of genes related to the development of connective tissues. Papillary fibroblasts were characterized by the expression of a high number of matrisome-associated genes whereas reticular fibroblasts gene signature mainly related to the core matrisome and ECM regulators. The regulation of selected genes was validated at protein level attesting to the robustness of the transcriptome analysis. Altogether, our data brought new insights into an ECM signature that is coherent with the organization and function of the papillary and reticular dermis.

RNA-seq analysis of match paired papillary and reticular skin fibroblasts cultivated as pro-angiogenic cell sheets

Project description:The aim of our study was to determin if papillary and reticular fibroblasts cell sheets expressed distinct genes involved in angiogenesis regulation and extracellular matrix. Thanks to our RNAseq analyssis, we demonstrated that papillary and reticular fibroblasts express specific signature of genes related to secreted angiogenic regulators and matrisome genes, suggesting that each subtype of fibroblasts differently regulate the formation of capillary via secreted factors and the microenvironment they generate. We confirmed this hypothesis with functional angiogenesis model in vitro. Overall, we show that papillary fibroblasts have a greater angiogenic potential and support the formation of dense branched vascular netword. On the other hand, reticular fibroblasts allow the formation of fewer vessel of larger diameter. This resultats are coherent with native skin vasculature and suggest that each fibroblasts subtypes play a role in regulating skin vascularisation.

Beta-catenin stabilization of skin fibroblasts causes fibrotic lesions by preventing adipocyte differentiation of the reticular dermis

Project description:The Wnt/alpha-catenin pathway plays a central role in epidermal homeostasis and regeneration but how it affects fibroblast fate decisions is unknown. Here, we investigated the effect of targeted alpha-catenin stabilization in dermal fibroblasts. Comparative gene expression profiling of Sca1- and Sca1+ neonatal fibroblasts, from upper and lower dermis respectively, confirmed that Sca1+ cells had a pre-adipocyte signature and revealed differential expression of Wnt/alphaâ??catenin-associated genes. By targeting all fibroblasts or selectively targeting Dlk1+ lower dermal fibroblasts, we found that ï?¢-catenin stabilization between E16.5 and P2 resulted in a reduction in the dermal adipocyte layer with a corresponding increase in dermal fibrosis and an altered hair cycle. The fibrotic phenotype correlated with a reduction in the potential of Sca1+ fibroblasts to undergo adipogenic differentiation ex vivo. Our findings indicate that Wnt/alpha-catenin signaling controls adipogenic cell fate within the lower dermis, which potentially contributes to the pathogenesis of fibrotic skin diseases. The dermis was separated from back skin of PDGFRAeGFP postnatal pups (P2) by incubation with thermolysin (0.25 mg/ml) (Sigma T7902) overnight at 4Â° and further processed as previously described (Collins et al., 2011). Cells were labeled in PBS + 10% FBS TruStain fcX anti-mouse blocking buffer with the following antibodies: anti-mouse Ly-6A/E (Sca-1)-Alexa Fluor-700 17. Two populations of cells were collected in triplicate. The PDGFRaH2BeGFP/Sca1- and the PDGFRaH2BeGFP/Sca1+. Single-cell gene expression of lymph node B cell-interacting reticular cells

Project description:Efficient germinal center formation requires the coordinated movement of B cells between distinct regions of the B cell follicle where their fate and function is governed by the integration of cues from their interacting partners. While CXCL13 is known to be important for B cell chemoattraction and follicle formation, the molecular identity of B cell-interacting reticular cells remains ill-defined. Moreover, how CXCL13-expressing, B-cell interacting reticular cells are reprogramed to cater to the developing germinal center remains unclear. Here we use the Cxcl13-Cre/TdTomato mouse model to genetically target and decipher the cellular composition of lymph node CXCL13-expressing reticular cells under steady-state and inflammatory conditions. Moreover, we examine the consequence of cell-specific, genetic perturbation of CXCL12 on the molecular identity of the reticular cell network. Transcriptomic analyses revealed that B cell follicle reticular cell subset specification is predetermined in the steady-state, although additional transcriptional changes accompany germinal center formation. Although genetic perturbation of CXCL12 alters GC topology, the molecular identity of the underlying reticular cells remains largely unperturbed.

RNA sequencing of splenic reticular cells in healthy and LCMV-infected mice

Project description:We report the transcriptome of subsets of mouse splenic mesenchymal reticular cells and lymph node reticular cells obtained from healthy Ccl19-cre x R26R-EYFP mice on a C57BL/6 background. We also examine splenic reticular cells from these mice infected with lymphocytic choriomeningitis virus (LCMV) strain Armstrong (acute infection) or strain Clone 13 (chronic infection). Subsets of CD45- CD31- Ter119- reticular cells were analyzed including PDPN+ T zone reticular cells (TRC) and MAdCAM-1+ marginal reticular cells (MRC) from spleen and lymph nodes. Additionally from the spleen we analyzed PDPN+ CD140a- cells, PDPN+ EYFP- cells, and EYFP+ red pulp reticular cells (RPRC) in healthy mice. We analyzed TRC, MRC, PDPN+ EYFP- cells, and EYFP+ RPRC from mice infected with LCMV Armstrong or LCMV Clone-13 on day 8 or on day 30 after infection. Our study represents the first detailed analysis of splenic reticular cell transcriptomes in healthy and LCMV-infected mice, with biologic replicates, generated by RNA-sequencing. We find substantial differential gene expression across subsets of splenic and lymph node reticular cells. We reveal significant changes in gene expression induced after virus infection that differ markedly with disease.
