Gene expression in rice roots after Pythium arrhenomsnes infection versus mock infection

Project description:Next to their essential roles in plant growth and development, phytohormones play a central role in plant immunity against pathogens. In this study we examined the role of hormones in the antagonism of the plant-pathogenic oomycete Pythium arrhenomanes against the root-knot nematode Meloidogyne graminicola in rice roots. Hormone measurements and gene expression analyses showed that the jasmonate (JA) pathway is induced early upon P. arrhenomanes infection. Exogenous application of methyl-jasmonate (MeJA) on the plant confirmed that JA is needed for basal defence against both P. arrhenomanes and M. graminicola in rice. Whereas M. graminicola suppresses root JA levels to increase host susceptibility, Pythium inoculation boosts JA accumulation up to levels that can no longer be repressed by the nematode in double-inoculated plants. Exogenous MeJA supply phenocopied the defence-inducing capacity of P. arrhenomanes against the root-knot nematode, whereas the antagonism was weakened in JA-insensitive mutants. Transcriptome analysis confirmed upregulation of JA biosynthesis and signalling genes upon P. arrhenomanes infection, and additionally revealed induction of genes involved in biosynthesis of diterpenoid phytoalexins, consistent with strong activation of the gene encoding the JA-inducible transcriptional regulator DITERPENOID PHYTOALEXIN FACTOR. Next to that, our results provide evidence for induced expression of genes encoding ERF83, and related PR proteins, as well as auxin depletion in P. arrhenomanes infected rice roots, which potentially further contributes to the reduced nematode susceptibility seen in double-infected plants.

Transcription profiling of heptatocytes from mice given a high-copper diet

Project description:In order to study the physiological consequences of a high-copper diet on hepatic gene expression, 6 mM CuCl2 was added to the drinking water for a period of 1 month. After this period, livers of seven control mice and eight copper-treated mice were isolated and were subjected to microarray analysis and copper measurements. The hepatic gene expression profile of copper-treated mice was compared to non-treated mice using a pooled reference.

Gene Expression in Response to excess Copper Stress in Rice Leaves

Project description:Gene expression in response to Cu stress in rice leaves was quantified using DNA microarray (Agilent 22K Rice Oligo Microarray) and real-time PCR technology. Rice plants were grown in hydroponic solutions containing 0.3 (control), 10, 45 or 130 µM of CuCl2, and Cu accumulation and photosynthesis inhibition were observed in leaves within 1 day of the start of treatment. Microarray analysis flagged 305 Cu-responsive genes, and their expression profile showed that a large proportion of general and defense stress response genes are up-regulated under excess Cu conditions, whereas photosynthesis and transport-related genes are down-regulated. The Cu sensitivity of each Cu-responsive gene was estimated by the median effective concentration value (EC50) and the range of fold-changes (F) under the highest (130 µM) Cu conditions (|log2F|130). Defense-related genes involved in phytoalexin and lignin biosynthesis were the most sensitive to Cu. Our results indicate that plant management of abiotic and pathogen stresses has overlapping components, possibly including signal transduction. Keywords: dose response, stress response

Jasmonate-responsive genes in rice

Project description:The plant hormone jasmonate (JA) plays important roles in the regulation of defense responses in many plants. To clarify the response to JA in rice at gene expression level, we performed a microarray analysis using the Agilent Rice Oligo Microarray (44k, custom-made; Agilent Technologies, Redwood City, CA, USA). As a result, treatment of JA caused high upregulation of many defense-related genes including pathogenesis-related (PR) genes in rice. However, many of these defense-related genes were not upregulated in JA-insensitive transgenic rice plant overexpressing JAZ8deltaC.

Project description:Jasmonates is inductively produced as a major plant hormone responsible for defense reactions in plants against both biotic and abiotic stresses, such as pathogen infection and mechanical wounding. Jasmonoyl isoleucine is known to be a bioactive compound of jasmonate and plays a pivotal role for plant defenses. We identified OsJAR1M-bM-^HM-^Rrelated JA-inducible genes in osjar1 tos17 mutant (osjar1-2) rice leaves 0 - 2 h after JA treatment using 44k microarray. Expression profiling in the wild-type rice leaves treated with jasmonic acid for 0, 0.5, 1, and 2 h was compared with that in the osjar1 mutant leaves treated with jasmonic acid for 0, 0.5, 1, and 2 h using two-color method with three biological replicates.
