Project description:au08-04_dfo - analysis of deferoxamine treated leaves and roots - What are the effects of the siderophore deferoxamine on Arabidopsis leaves and roots? - Plants were allowed to grow for 5-6 weeks. The nutrient solution contains 0.25 mM Ca(NO3)2.4H2O, 1mM KH2PO4, 0.5 mM KNO3, 1mM MgSO4.7H2O, 50 µM H3BO3, 19 µM MnCl2.4H2O, 10 µM ZnCl2, 1 µM CuSO4.5H2O, 0.02 µM Na2MoO4.2H2O and 50 µM FeNa-EDTA. Plants were subjected to an 8 h light/16 h dark cycle, at 19°C, with 70% relative humidity. Leaves of six week old hydroponically grown A. thaliana Col0 plants were infiltrated with 1mM deferoxamine or sterile distilled water. Leaves were harvested 7 and 24 h.p.i. Transcriptome comparison of the irt1-2 mutant (pam42) and the corresponding wild type (Col-0)

Project description:IRT1 is the root high-affinity Fe uptake system. Despite severe Fe deficiency symptoms and reduced Fe levels in the shoots of irt1 mutants, we find that root Fe concentrations are higher in the irt1-2 mutant than in the wild type, unexpectedly. The goal of this experiment was to identify candidate transcripts contributing to the observed alteration in root-to-shoot Fe partitioning of irt1. We analysed gene expression in shoots and roots of the wild type grown under control conditions, the wild type exposed to severe Fe deficiency for 5 d, as well as of the irt1-2 (pam42) mutant grown under control conditions.

Transcriptomic characterization of spl7 and Col-0 ecotypes of Arabidopsis reveals new aspects of Fe-Cu crosstalk

Project description:Iron (Fe) and copper (Cu) are essential metal micronutrients that are necessary for many redox reactions. The uptake of these metals is tightly regulated in plants. Some redox processes can alternatively use Fe-containing proteins or Cu-containing proteins, depending on nutritional status. Copper deficiency can rescue a Cucumis melo Fe uptake deficient mutant, and Fe deficiency can result in increased accumulation of Cu. However, the system responsible for Fe-deficiency-regulated Cu-uptake is unknown. To understand the genes and gene networks associated with Fe-deficiency regulated Cu uptake and Fe-Cu cross-talk, we conducted transcriptomic profiling of roots and rosettes of spl7 (a Cu uptake deficient mutant in arabidopsis) and Col-0 (WT) grown under Fe, Cu and simultaneous Fe and Cu deficiency conditions.

Project description:Iron (Fe) plays a pivotal role in several metabolic and biosynthetic pathways essential for plant growth. Fe deficiency in plants severely affects the overall crop yield. Despite several studies on iron deficiency responses in different plant species, these mechanisms remain unclear in the allohexaploid wheat, which is the most widely cultivated commercial crop. In order to gain a comprehensive insight into molecular responses of bread wheat when exposed to iron deficiency, we studied transcriptomic changes in the roots and flag leaves of wheat plants subjected to iron-deficient and iron-sufficient conditions during early grain filling.

Iron-deficiency-induced changes in wild type and IRP1overexpressed transgenic Arabidopsis plants

Project description:a novel orphan peptide, IRON-REGULATED PROTEIN1 (IRP1) that is rapidly induced by Fe deficiency and improves growth on Fe-deplete media. In Arabidopsis, ectopic expression of IRP1 affected the activity of several genes involved in Fe acquisition and homeostasis, causing a dramatic increase of Fe in leaves and enhanced seed Fe loading. Heterologous expression of IRP1 in tomato plants resulted in increased Fe levels in fruits. Integration of AtIRP1 into the genome of crop plants may represent a novel strategy for Fe biofortification. wild type Col-0 and transgenic overexpressor IRP1-OE plants were grown under normal conditions or subjected to and iron-starvation for 3 days. Roots and shoots were collected with 3 biological replicates.

Project description:We reported the flg22-triggered immune responses in roots affect the iron deficiency responses and may link to the function of FLS2 and IMA1 in the root. To identify the underlying mechanism of how the root transcriptome profiles respond to +Fe, +Fe+flg22, -Fe, -Fe+flg22 respectively, and if the flg22 responses is dependent on the function of FLS2 and IMA1 in the root, we performed an mRNA-seq experiments in Col-0, fls2 and UBQ10::mCitrine-IMA1 with different treatments. The differentially expressed genes in response to +Fe, +Fe+flg22, -Fe, -Fe+flg22 were analyzed. It has 36 samples in total, with 3 biogical replicates for each condition and each genotype.

Transcript profiling of the roots and leaves of opt3-2 mutant and wild type Arabidopsis

Project description:Illumina 100bp single end sequencing was used to profile the transcriptomes of wild type (Columbia) and opt3-2 mutant Arabdiopsis leaves and roots under hydroponic conditions ( 4 weeks old, bolting stage). Sequencing resulted in 381 million uniquely mapping reads accross 36 libraries. Each sample was barcoded and divided into three sequencing lanes, while each genotype/tissue combination is represented by 3 biological replicates consisting of 3-4 individual plants (2 genotypes x 2 tissues x 3 biological replicates x 3 technical replicates = 36 libraries). Our results show the leaves and roots of the opt3-2 mutant have different iron sensing transcriptional programs in place which is independent of the iron status of the plant.
