Developmental effects of maternal dietary vitamin A deficiency and P450 cytochrome 1b1 deletion in mice

ABSTRACT: Wild type C57Bl/6J, Cyp1b1-null, and a substrain of Cyp1b1-null that are resistant to diet-induced obesity (Resistant Cyp1b1-null) timed mated pregnant dams were administered either a defined vitamin A sufficient diet or matched vitamin A deficient diet from embryonic day 4.5. Offspring liver gene expression was examined at birth (post-natal day 0) and at weaning (post-natal day 21).

Project description:Signaling via the arylhydrocarbon receptor (AhR) is thought to contribute to exacerbation of allergic asthma by anthropogenic pollution. The physiological role of AhR and its downstream targets CYP1 family members upon exposure to aeroallergens remain unknown. We seek to characterize the role of the AhR pathway for the regulation of allergic airway inflammation (AAI). We employed knockout animals of the AhR and its downstream target cytochrome P450 enzymes CYP1A1, CYP1A2 and CYP1B1 and subjected them to preclinical allergic asthma models (ragweed and house dust mite) to assess the role of AhR and CYP1 family members in allergic airway inflammation. Histological, transcriptional and functional assays were used to uncover relevant cell types and mechanistic insights. In the ragweed model, AhRKO/KO and CYP1B1KO/KO but not CYP1A2 KO/KO or CYP1A2KO/KO animals showed exacerbation of AAI including elevated infiltration of cells in the bronchoalveolar lavage fluid and increased serum levels of IgE and allergen-specific IgG1. AhR KO/KO and CYP1B1KO/KO animals showed also more severe reactions in HDM-induced allergic AAI. Bone marrow chimeras as well as qPCR analysis and immune fluorescence histology indicate that expression of CYP1B1 in non-hematopoietic cells – presumably lung epithelial cells – is necessary to prevent exacerbation of HDM-induced AAI. Mechanistically, we show that CYP1B1 deficiency leads to enhanced expression and activity of CYP1A1 in lung epithelial cells and thus potentially to deprivation from endogenous AhR ligands. Transcriptional analysis of primary lung epithelial cells indicates a functional link of the AhR to regulation of circadian clock genes at baseline and massive alteration of gene expression upon allergen exposure.

Determine what gene changes in the Cyp1b1 null livers may be contributing to prevention of increased adiposity when givin a high fat diet. Three Comparison experiment. Comparing wild-type low fat vs Cyp1b1 null low fat, wild-type high fat vs Cyp1b1 null high fat and low fat vs high in wild-type mice. Sample size is 3 per group. Impact of dietary fatty acids on gene expression of liver-specific knockout of cytochrome P450 reductase

Project description:In the present study, we explored the hypothesis that the fatty liver phenotype and associated gene expression changes associated with the specific deletion of the POR gene in adult mouse liver could be abrogated by supplementation of the mouse diet with the very long chain highly unsaturated fatty acids, arachidonic acid (C20:4ω6), eicosapentaenoic acid (C20:5ω3) and docosahexaenoic acid (C22:6ω3). We expected the fatty liver phenotype would not be reduced by the polyunsaturated fatty acids linoleic (C18:2ω6) or linolenic acid (C18:3ω3), since these accumulated in the fatty livers of LivPORKO animals. This proved to be the case. However, we also made two surprising observations. First, control animals fed a diet enriched in PUFA had fatty livers and gene expression profiles similar to animals fed a lard diet, which was deficient in both PUFA and HUFA. Second, while a diet enriched in HUFA did result in reduced steatosis in livers of the LivPOKO animals, fat accumulation was still elevated relative to controls. Array analyses indicated most differences in gene expression were related to fatty acid metabolism and could explain differences in fat accumulation in LivPORKO livers with dietary treatment.

Maternal dietary vitamin A deficiency and diet dependent liver gene expression in mice

Project description:Initiation of a vitamin A deficient diet in mid-gestation, maintained in the post-weaning diet is sufficient to cause liver and serum retinoid depletion. Wild type C57Bl/6J timed mated pregnant dams were administered either a defined vitamin A sufficient low fat (12 percent kcal from fat) diet or matched vitamin A deficient diet from embryonic day 10.5. Vitamin A sufficient offspring were weaned onto either a high fat diet (60 percent kcal from fat) or maintained on the low fat 12 percent kcal from fat diet for 11 weeks (14 weeks of age). Gestational vitamin A deficient offspring were maintained on the same vitamin A deficient diet until 14 weeks of age. The impact of the maternal diet on a post-weaning high fat diet was compared to a standard maternal breeder diet followed by the post-weaning high fat diet.

Mutation of the cytochrome P450 CYP360A8 gene increased sensitivity to herbicide paraquat in Daphnia magna

Project description:Purpose: To create a CYP gene mutant from D. magna as a model for xenobiotic metabolism study Methods: We performed RNA-seq of wildtype 12 days old D. magna to find out the basal expression level of CYP genes. We picked up 5 highest expressed CYP genes and examined the temporal expression level from day 0 to day 12 by qPCR. We chose the highest expressed gene and performed mutagenesis to this gene by CRISPR/Cas9 to creat a mutant line. Results: From RNA-seq result, we found that CYP360A8 is the highest expressed CYP in D. magna. qPCR result showed that CYP360A8 has developmental pattern where its expression is low in juvenile, increase 6 fold by maturation time and stay stable till day 12. Mutagenesis experiment of this gene resulted in the obtaning of a monoallelic mutant. Paraquat exposure to this mutant line showed more sensitivity of the mutant compared to wildtype (showed by 1.3 fold lower of EC50). Conclusions: Our study showed the first mutant model of CYP in D. magna. This may be the start of creating a panel that list D. magna CYP gene with its inducing xenobiotics.
