The sunflower transcription factor HaHB11 improves yield, biomass and flood tolerance via the quiescent response in transgenic plants

Project description:HaHB11 is a divergent member of the sunflower homeodomain-leucine zipper I subfamily of transcription factors. A sunflower microarray was hybridized with RNA from HaHB11-transformed leaf-disks and its analysis indicated the regulation of many genes encoding enzymes from fermentative pathways. A 1300 bp sequence exhibiting flooding-responsive cis-acting elements, upstream the transcription initiation site, was isolated and Arabidopsis plants transformed with a construct in which this promoter directs GUS expression. GUS expression was induced after flooding treatments and, in the same sense, HaHB11 expression was induced by flooding stress in sunflower. Furthermore, Arabidopsis plants were transformed with different constructs, varying in the promoter used, able to express HaHB11 cDNA. These transgenic plants exhibited differential phenotypes which the more remarkable traits were a significant increase in the rosette biomass and seeds yield. Moreover these plants exhibited an enhanced tolerance to flooding stress, both to submergence and waterlogging. Starch, sucrose and glucose contents were higher in the transgenics compared to WT and decreased less after submergence treatments. Finally, transcript levels of selected genes, involved in fermentative pathways, and the corresponding enzymatic activities were assessed both in sunflower and transgenic Arabidopsis plants after submergence indicating a general repression of the fermentative pathway which suggested that tolerance occurs via the quiescent strategy. Altogether the results indicated a role for HaHB11 in the flooding response and allowed us to propose this gene as a biotechnological tool to improve crops for increased yield and biomass and flooding tolerance.

Project description:affy_sunyfuel_drought_sunflower - Drought is the main cause or crop loss around the globe and it is expected to be more so in years to come. Helianthus genus comprises species that are able to grow in harsh environments suffering from severe water shortage. Cultivated H.annuus, i.e. sunflower, is a highly valuable oil crop due to, among other features, its ability to grow under low input conditions. The aim of our project is to unravel the transcriptional regulation underpinning drought tolerance in sunflower, with the ultimate goal of improving crop yield. Ninety six individual pots were arranged in six blocks of 16 plants each, which were sown in individual pots. Each block contained 2 plants of 8 different genotypes, one of which was used as control and the other one as treated plant. Plants within each block were randomly arranged. At the right developmental stage, treated plants were water-deprived and their water status was monitored as compared to the untreated control. Three out of the six blocks were harvested on a given date, so water stress among genotypes varied. ThERF1 regulates its target genes via binding a novel cis-acting element in response to salt stress

Project description:To reveal the regulatory network of ThERF1 in response to abiotic stress, we have employed an Agilent Arabidopsis gene expression microarray to study the expression profile changes between Wild-type and transgenic plants overexpressing ThERF1 under salt stress. A total of 154 and 307 differentially expressed genes with p-value < 0.05 and M-bM-^IM-% 2-fold change were significantly up- and down- regulated in ThERF1-transformed plants under salt stress treatment. Four-week-old seedlings of Wild-type and ThERF1-transformed Arabidopsis plants treated with 120 mM NaCl for 6 h were immediately harvested for RNA isolation. The Agilent Arabidopsis (V4) gene expression microarray (4M-CM-^W 44K, Design ID: 021169) was employed to compare the gene expression profiles of Wild-type and transgenic plants with three independent biological replicates.

Global expression profiling of wild type and transgenic Arabidopsis plants in response to water stress

Project description:Transgenic Arabidopsis plants with constitutively low inositol (1,4,5) triphosphate exhibit an increased tolerance to water stress by an ABA-independent pathway The phosphoinositide pathway and inositol (1,4,5) trisphopsphate (InsP3) are implicated in plant responses to stress. In order to manipulate the pathway and determine the downstream consequences of altered InsP3-mediated signaling, we generated transgenic Arabidopsis plants expressing the mammalian type I inositol polyphosphate 5-phosphatase, an enzyme that specifically hydrolyzes the soluble inositol phosphates and terminates the signal. Transgenic plants have no morphological differences compared to wild type; however, rapid transient Ca2+ responses to a cold or salt stimulus are reduced by ~ 30%. To further understand the role of InsP3-mediated signaling in plant stress responses we focused on drought stress. Surprisingly, the InsP 5-ptase plants lose less water and exhibited an increased tolerance to drought. Stomatal bioassays showed that transgenic guard cells are less responsive to the inhibition of opening by ABA but show an increased sensitivity to ABA-induced closure. The onset of the drought stress is delayed in the transgenic plants and ABA levels did not increase as much as in the wild type. Transcript profiling has revealed that DREB2A and a subset of DREB2A regulated genes are basally up regulated in the InsP 5-ptase plants. These results indicate that the drought tolerance of the InsP 5-ptase plants is mediated in part via an ABA-independent pathway. The constitutive dampening of the InsP3 signal in this system has uncovered novel regulation and cross talk between signaling pathways. Keywords: drought stress, expression study

Whole plant aged of one month were harvested and frozen in liquid nitrogen. The plants were ground with a mortar an pestls and RNA extraction was performed with the RNeasy extraction kit (QIAGEN) with cristal of PVP. The RNA concentration was measured on a NANODrop spectrophotometer.
