Cell-type independent MYC target genes reveal a primordial signature involved in biomass accumulation

Project description:The functions of key oncogenic transcription factors independent of context have not been fully delineated despite our richer understanding of the genetic alterations in human cancers. The MYC oncogene, which produces the Myc transcription factor, is frequently altered in human cancer and is a major regulatory hub for many cancers. In this regard, we sought to unravel the primordial signature of Myc function by using high-throughput genomic approaches to identify the cell-type independent core Myc target gene signature. Using a model of human B lymphoma cells bearing inducible MYC, we identified a stringent set of direct Myc target genes via chromatin immunoprecipitation (ChIP), global nuclear run-on assay, and changes in mRNA levels. We also identified direct Myc targets in human embryonic stem cells (ESCs). We further document that a Myc core signature (MCS) set of target genes is shared in mouse and human ESCs as well as in four other human cancer cell types. Remarkably, the expression of the MCS correlates with MYC expression in a cell-type independent manner across 8,129 microarray samples, which include 312 cell and tissue types. Furthermore, the expression of the MCS is elevated in vivo in Em-Myc transgenic murine lymphoma cells as compared with premalignant or normal B lymphocytes. Expression of the MCS in human B cell lymphomas, acute leukemia, lung cancers or Ewing sarcomas has the highest correlation with MYC expression. Annotation of this gene signature reveals Myc's primordial function in RNA processing, ribosome biogenesis and biomass accumulation as its key roles in cancer and stem cells. Expression of genes in Runx2/Myc transgenic mouse thymus compared to control

Project description:Retroviral insertional mutagenesis (RIM) is a powerful tool in cancer genomics. Transgenic mice expressing two potent collaborating oncogenes in the germline, Myc and Runx2, develop rapid onset tumours that can be accelerated and rendered polyclonal by neonatal infection with MoMLV. Deep sequencing of lymphomas from infected Runx2/Myc mice revealed a network of progression genes. Transcriptional analysis of basal expression levels in transgenic cells showed that the progression network showed strong evidence of clonal selection beyond gene expression level. We examined the gene expression profile of triplicate 10 day old thymus from CD2-Myc/CD2-Runx2 and C57/CBA mice to examine early gene changes in comparison to RIM targets in end stage tumours. We found no significant relationship between basal expression in the transgenic system and genes that were targets for retroviral integration.

A Myc-mediated dormant state of naive ESCs

Project description:Mouse embryonic stem cells can be maintained in a naive state of pluripotency by culture conditions containing inhibitors of Mek and Gsk3. Myc activity is essential for efficient cellular reprogramming. We genetically addressed the role of c-myc and N-myc in naive ESCs cultured in 2i by inducing the deletion of both genes using CRE-mediated recombination. Our findings show that myc activity controls the biosynthetic and proliferative machines of ESCs without significantly affecting the pluripotency network.

Functional cooperativity between Myc and PI3K signaling for indefinite self-renewal property of embryonic stem cells

Project description:We explored the relationship between Myc activity and PI3K signaling in ESCs. Our data demonstrate that Myc and PI3K signaling function cooperatively for supporting pluripotent property of ESCs. Moreover, our data demonstrate that exposure of ESCs to 2i condition render both Myc and PI3K dispensable for preserving ESC status. Effect of PI3K inhibitor, LY4294002 on EBRTcH3 ESCs or their derivatives overexpressing c-Myc (wild-type or T58A mutant) was examined. Effect of LY4294002 on EBRTcH3 ESCs under 2i conditions was also examined. Furthermore, effect of Max expression ablation was compared between ESCs and those overexpressing p110 alpha.
