Transcriptome analysis of BMDM from wild type and Rasgef1b_fl/fl_vav-icre mice treated or not for four hours with R848

Project description:Despite much progress, the specific role of some the putative intracellular signaling players upstream to inflammatory gene expression activated by Toll-like receptor (TLRs) in the immune response remains unknown. For instance, the TLR-inducible Ras guanine exchange factor member 1b (RasGEF1b) was previously shown to be expressed in activated macrophages, but its specific function is still obscure. To investigate the role of RasGEF1b in TLR-mediated innate immune response, we generated Rasgef1bfl/fl mice for tissue conditional gene deletion . We found that the expression and production of the neutrophil chemoattractant chemokine KC/Cxcl1 was impaired in response to TLRs stimulation of bone marrow derived macrophages (BMDMs) devoid of RasGEF1b. The production of KC/Cxcl1 was also impaired in in vivo models of LPS-induced peritonitis, and R848-induced acute lung injury inflammation. RasGEF1b was not required or sufficient to neither induce nor increase the transcriptional activation of Cxcl1 promoter. In addition, TLR-triggered inflammatory activation of NF-ΚB signaling pathway was unimpaired in macrophages lacking RasGEF1b suggesting a mechanism independent of the NF-B transcription factor. Our data presented herein revealed an important role of RasGEF1b in innate immunity by regulating gene expression, following TLR stimulation.

Project description:Apilimod is a reported TLR pathway antagonist in clinical trial. To understand the mode of action of apilimod, we applied global microarray analysis to explore the effect of apilimod on gene expression of RAW cell in the absence of presence of R848 (TLR7 ligand). RAW cells were stimulated with R848 in the absence or presence of apilimod (1μM). mRNAs were extracted at 0h, 1h,3h, 7h, and 22h for affymetrix analysis. The inactive analog of apilimod (API09) was included as a control.

Project description:Murine splenocytes were isolated from the spleens of C57BL/6J mice and were pretreated in vitro for three days in the presence of Pam3CSK4 (1 μg/ml), high pure LPS from E.coli O111:B4 (100 ng/ml) and R848 (5 μg/ml). PBS-treated splenocytes served as negative control. We used Qiagen Toll-like Receptor RT2 Profiler PCR Array kit to quantitate gene expression profiling of the TLR signaling pathway.

Project description:Analysis of whole mouse muscle and inguinal lymph node gene expression signature induced after 6h by in-vivo intramuscularly administration of MF59, alum, CpG, resiquimod (R848), Pam3CSK4 and DMSO and PBS controls. Analysis of splenocyte gene expression signature induced by the same treatments after 6h of incubation. MF59 and alum are licensed human vaccine adjuvants; CpG is a TLR9-agonist adjuvant; resiquimod (R848) is a TLR7/8-agonist adjuvant and Pam3CSK4 is a TLR2-agonist adjuvant.
